本来我今天溜号出去打算跑步的，但阳光明媚春风拂面，路边的花儿又实在是招惹人意
乱心迷，不停的用手机咔嚓咔嚓。春天，终于来到了。。。。跑了半圈就回来了。。。
晚饭后先去park散步，姐姐忽然问，妈妈为什么你的泳衣都是two pieces，为什么你从
来不穿one piece呢？怔了一下，不知道该怎么回答姐姐。于是，解释说，因为one
piece会用掉更多的布料，妈妈为了节约布料保护环境所以就穿two pieces啦。噢！姐
弟俩立马觉得妈妈好伟大！
今天修改了loads/reps，而且因为弄清了蛋白粉的问题，于是2 cups slim milk+1
scoop whey powder装水瓶里带到GYM，在WT结束后立即喝，然后才去游泳，感觉非常好。

现在对打腿的技术要领，也就是膝盖不弯，大腿发力带动小腿和脚，理解的比较清楚，
执行起来也算马马虎虎。
直到昨天，发现另外两个技术要点的冲突：脚内旋和屁股夹紧。以前打腿时脚腕内旋不
成问题，我是比较严重的外八字脚，所以脚腕内旋幅度不够，为了增加内旋，导致髋部
adduct。昨天看到大家着重讨论控制腰的重要性，其中一个要点就是通过夹紧臀肌保持
腰部紧张不松懈，但夹紧臀肌需要外旋。所以我特意示范了一个动作，小酸说做不出来
，我现在才意识到原来自己体型半残。 |

极端即可。美国的年龄组成绩很有道理，B到BB大概对应50th percentile，只要高于这
条线，不偏科，由他去。
“发力太早”不是问题。有点小伤有点小失误是难免的，是小孩成长必需的。
个子稍矮又爱争胜的小孩一般容易采用相对高频的方式。这是克服四肢短手脚小的有效
手段，但不是唯一手段，在青春期发育前更是如此。对小个子，全身所有关节和肌肉柔
韧性都不要少于队友，同时应该重点加强shoulder & hips。

我一年前猛练abduct/adduct，早就能轻松夹个125 lbs了，但是最近做lunge和深蹲，
发现差距还是很大，也有不少进步空间。感觉使劲的不完全是一个地方。以前说实话我
抗拒weight training的一个主要原因，就是怕局部力量练大了整体并没什么爆发力的
效果，现在算比以前知道得多了点
还是要赞一下210 lbs leg press，我记得很久以前你说 180 lbs的时候我跳上去量了
量，能很勉强做个10下。不过我倒没重点练过那个 |

-14 Da有可能是-17 or -18Da (-NH3, H2O).
-74 和 -113可能是site chain或者terminal 的氨基酸掉了。
如果可能， 你可以把terminal的序列写出来， 大家可以看看。
不过你的分子其实很小， 7k应该误差相对较小。 |

peptide，只是经过fragmentation之后，fragments的pattern和从sequence预测的
pattern很像，所以我们觉得这可能是个peptide。
对于一个已知序列纯化过的蛋白，一般来说可以cover 80-9x％的digest以后的peptide
，但对于比较复杂的体系，比如cell lysate，那么因为peptide太多仪器通量不够，或
者因为非典型salt adduct，数据库序列不全等等原因，很可能会miss掉很多。具体
miss了多少，这个用现有的商业化软件很难evaluate。 |

you run it in the positive mode and this makes you think it is a fatty acid?
It can be ionized in positive mode very well without coupling to any adduct
ions. That means you might have some N containing functional group, which
also points to the molecular formula of C4H8N6O2. But I can not imagine any
very hydrophobic stuff as you claimed could have 6 N in it.

发帖数: 54 | 40 多谢大家的回复。MMP是N-methylmorpholine。我试过了3小时和16小时脱保护，结果没
有太大区别。经过重新分析质谱结果，我相信多出的分子量49（之前以为是50）是由于
有一个piperidine adduct（＋67）和一个aspartimide（－18）的结果。只是仍不清楚
到底是哪两个Asp的side chain发生了副反应。我查到两篇报导化学合成flag peptide
的文献，都是用fmoc化学固相合成的。一篇用的试剂和我用的大同小异，不过是手工合
成的。难道手工合成可以避免aspartimide的生成？我已经用很短的时间脱fmoc了。（
2X2min）
最后一招就是用Boc化学合成了，如果实在没有别的办法的话。
有人用fmoc化学作成过flag peptide吗？ |
