糖尿病field,有一种说法是adipose tissue在快速增殖的时候,angiogenesis跟不上
obesity的expansion,造成local hypoxia。而hypoxia引起inflammation,
然后recruit macrophage,inflammation继续加重。
这种恶性循环最后导致global insulin resistance 。

代谢和免疫?都不算是worm/fly的太好的用武之地吧 除非你有很specific的问题要问
并且别的系统不好回答。比如说你要搞pathogen/host interaction 这个worm/fly好歹
还是活得动物;之类的。
worm/fly都没有adaptive immune system(虽然要说起来Toll还是果蝇里clone的)。
worm/fly也都没有正儿八经的mammalian代谢的遗传/组织元素。比如说两者都没有正儿
八经的insulin(只有ILP);都没有liver, adipose tissue,就算是有类似的组织功
能也不是那么一样;果蝇幼虫和线虫都没有控制食欲的神经系统,基本就是一辈子吃到
死的意思。。

嗯 挺多neuro+metabolism,不过我是觉得做得不够好了,尤其是老鼠里。如果你比比我
们对peripheral metabolism (in adipose, liver, muscle, pancreas, etc)的理解和
对brain的理解 能差个数量级吧感觉。
呵呵 还真有人interview问我这个问题。。其实我从来就没想过继续做的说。。所以也
就压根没和他讨论过。。。觉得原来的话题太小众了 不好玩。。

apoptosis了以后就release free fatty acid,然后macrophage也来了,
然后又是inflammation什么的,情况就越来越糟糕。
adipose tissue在身体里面的作用就跟加油站,炼油厂差不多。
平时管理好了,相安无事。
但是加油站要是炸了,energy都给瞬间释放了,肯定没什么好事情。

有2个postdoc offer,
1) 方向是GWAS,主要是验证某个SNP和某疾病的相关性。经费未来5年都比较充足。学
校是某一大牛校。PI是此疾病领域的临床医生,应该在该疾病领域比较牛。
2) 方向是脂肪细胞分化,主要研究brown adipose tissue分化。经费未来5年充足。
学校是中牛校,PI是AP。
1)的优点是大牛校,PI也牛,缺点是GWAS这个领域水分比较大,不知道对我未来的
faculty前景是否会有不利影响,另外本人也不大喜欢functional genetics,感觉这个
领域很容易做到头。不知道各位对该领域有何看法。
2)的优点是领域比较热,研究前景似乎不错,缺点是PI是新AP,目前还没有好的文章
发表,虽然PI的博后老板是大牛,博后发的文章也不错。也就是说有一定的风险。本人
倾向2),但是因为博士期间做的是癌症相关的研究,对上述领域都不是非常熟悉。希
望懂行的人指点。

muscle specific insulin receptor KO mouse
这个老鼠的肌肉是insulin resistance,但是整个老鼠并不insulin resistance。
是个比较奇怪的mouse model。
相对靠谱的解释是说,adipose tissue变得更加insulin sensitive,
然后就起到了compensation的作用。

最近要提取从白色脂肪里面提取SVF,根据protocol我确实看到了pellet,用TRI-
reagent也提取到了mRNA,nanodrop测出的浓度有800ng/ul,但是我试了cyclophilin,
gapdh,18s几个内参,都测不到CT值。已经重复几次了,郁闷的不行。请教各位,有什
么建议么?
SVF protocol
1)isolate WAT,
2) Incubate tissue @ 37C for 45min with shaking @ 80 rpm (Collagenase I,
1mg/ml)
3)300g for 10min
4) Dissolve pellet in Tri-reagent.

换一下酶。我个人感觉collagenase不够强。组织解离不够。 我用collagenase 2mg/ml
+dispase. 或者直接上trypsin LE 都可以。

现在不是组织解离不够,我能拿到细胞,也能测到RNA 浓度,可是housekeeping gene
18s,GAPDH和Cyclophilin都测不出,这个是什么原因?
楼上的你有没有测过SVF的gene表达呢?用什么housekeeping gene呢?谢谢

RNA 跑跑胶看看.如果housekeeping 都测不出,你拿到的那就不是RNA,或者是降解了.如
果RNA 确定没问题,是不是PCR 的reagent有问题了?拿一个阳性RNA 来测.一步一步的排
除,还能找不到原因?

Insulin sensitization would result in increase in RER, which shows the
elevation of carbohydrate oxidation and the concomitant reduction of fat
oxidation. Therefore, the overall energy expenditure would not be altered.

现在好多所谓的大paper 完全违反生理学原理,没什么意义。 Please keep in mind,
energy demand determines energy expenditure, and NOT the other way around!
Without the increase in energy demand, a mere expansion of oxidative
capacity, such as induction of browning of WAT, will not do anything!
Browning of WAT is operative under cold because the body energy demand under
cold stress is increased (to maintain temperature homeostasis). I don't
