Montana State University的一个搞中国历史的女教授Maggie Greene写了篇博客散文
，细数她在上海文庙淘到的80年代中国连环画。其中包括一本1980年科学XX（惭愧我认
不出这俩字，同志们帮忙看看下图）出版社广州分社出版的《星球大战》——要知道星
战第一集1977年在美国公映，1979年在香港公映。刚结束文革的中国大陆还处在相对闭
塞的状态，却能在香港上映一年后出版连环画，让搞历史的Greene非常惊喜。广东当时
跟香港跟得紧吧。
Greene兴致盎然地观察文化传播过程中发生的细微变化，比如当时中国人心目中的美国
家庭是这样吃晚饭的：
Greene笑说不仅桌上有一只电饭锅、一道热菜，而且主妇似在把一只烤鸭从toaster

旁白的男孩子。
记忆里第一次见到他，是十年前的这个季节，系楼？楼大厅边的楼梯口。那段日子，我

昨天和老婆聊天，老婆说在水木上看了一个男性选手写给他领导的四周年纪念文章，说

错。说起我可爱的老婆，基本上就是听见风，就要跟出去看看，简称跟风。于是任务交
待下来，让文采奕奕的我，在我们四周年的时候，也要发一篇大作，不求上什么nature
，science，但至少要上清华某水木的首页导读。
是呀，相识两年了，结婚也一年了，我还没有任何paper献给我可爱的老婆，实在是委
屈她了。别人四周年纪念，我们完全可以搞个两周年纪念嘛。每两年一纪念，等到80岁
，就可以出一个文集了。

Tried a different region again in my cell type that I had edited
successfully with CRISPR-Cas9 in HEK293 (note that these are not the cells I
'm trying now). The guides are essentially the same except for 2 extra bp on
either side of the sgRNA sequence. Tried a number of different combinations
for nucleofection but no deletions. The first lane is PCR from edited
HEK293 gDNA that I had from before, the rest of the lanes are various

化那个oligo，而不是从厂家订磷酸化的oligo。因为这个5'-end 磷酸化很关键，没有
磷酸化的Oligo不能被NgAgo识别。从理论上来说，如果oligo 的磷酸化不完全的话，就
有可能出现那些没有磷酸化的oligo和磷酸化的oligo 互相竞争结合基因上的位点而降
低酶的效率。所以弄不好这个磷酸化就是其中的技术要点。
貌似这个是目前第一个跳出来说重复出来了的人。静待更多的跟进报道。

现的优先权，会人为的“操作”某些实验过程及其结果，以使自己的文章最快发出来。
因为这些人知道，这个发现是很有可能正确的，只是囿于某些条件限制，领域内一直进
展不大，但是通过人为的“操作”数据，自己就可以把这个成果最先发表出来；过不了
多久，自己或其他实验室做了进一步的完善和重复，就没有人会去追究第一篇文章是否
有“夸大其词”甚至造假的问题了。
我现在担心的是韩可能也采取了这种策略：基于2014年的文章，可以明确存在合适的
DNA-guide 的Ago, 但一时找不到确实有效的蛋白，于是采取“夸大其词”的手段，过
度解读自己的结果（这里我不愿猜测其造假），目的就是为了尽早把这个概念性的文章
发表出来，取得时间差上的优势，然后再慢慢优化系统，寻找真有有效的蛋白。这样一
来，首发权就拿到了。
前段时间闹得沸沸扬扬的张生家事件，就是这种行为的典型注解。 |

然后14年高温的ttAgo是不能切割线性质粒的，文章中称ttago的切割需要负超螺旋结构
，切割的产生需要dna结构的帮助，而这个切割基因组的切割活性并不能确认。
退一万步讲，ttAgo在高温下能产生给切割，并不是说NgAgo在37度下就一定能切割，反
过来，如果NgAgo没有切割活性，也不能证明ttAgo就也在高温下没有活性。。这是两个
物种的ago系统，这两个之前没有什么必然的逻辑联系。
最近的例子，张锋的cpf1系统，张锋鉴定了好多物种的cpf1，很多在体外都有切割活性
的，都是cpf1系统，但是在哺乳动物细胞内，只有两个cpf1系统能够有效切割基因组。 |
