硕士在国内上的，要做蛋白纯化，06年的时候，可能全校也就两台akta蛋白纯化系统吧
，一台就在本科教学实验室。2010年，在德的第二年，系里面组织参加akta的培训，到
了现场，我一下子就愣住了：刚买的16台仪器......
随后，我想起来了我们本科专业实验，2003年的时候，我们的生物技术大实验，要做蛋
白纯化，2个人一组。其中包括离子交换层析和凝聚过滤层析，都是我们自己装的柱子
，然后链接蠕动泵和核酸蛋白检测仪。

，如何才能获得弃掉鸡肋的资格，或者说，得什么样的能力才能拿到这种offer而且不

自己背景一般： 2流211＋985大学， 数学佳，被忽悠学生物制药。高中根本不喜欢生
物和化学，大一时用了很久很久才明白DNA是咋复制，咋2倍，咋4倍 。。。。。
大学混了一下，想转专业，没有魄力。
毕业时，2004年，一个大国企的offer, 1200人刀，一个私企offer, 做销售。
仰天长叹，我他妈还不如回去筛沙呢！
于是考研想学蛋白药物，可惜被top2拒，调剂到一所谓top科研院所，被忽悠搞干细胞
。对胰岛发育神经发育稍有射猎，3年4篇文章，不全是一作。于是觉得自己还是个搞科
研的料，猖狂一时。毕业时一心想出国，老婆也厌倦了北京的堵车。
拒掉pfizer的销售，sanofi-aventis销售，另一外企的研发及国外培训机会，offer大
概5000人刀。
2007，顺利来到德国一老牌名校。教授说 alle anfang is schwer, 还说做科研重要的

发帖数: 49 | 41 我正在做2个蛋白的纯化（分别纯化），一个约270kda，另一个约90kda。以前一直
在用GE prepacked Superdex 200（bed volumn=2.4ml, loading volumn=50ul) 的一个
小柱子做微量的size exclusion纯化然后用来做生化试验。
最近对所需蛋白量要求变大了，所以正在找合适的大柱子能让我一次的loading
volume到~5ml. 碰巧前几天旧的UV detector的灯泡也坏了，问别人说换新的要700多刀
，还不能保证修好。现在我盘算着能不能干脆整套新的。
我问了下周围的同事，有人给我推荐GE的AKTA pure系统。我看了下觉得功能太多
太花哨（估计也很贵），对我这种只需要单一步骤蛋白纯化的人好像太浪费了。所以上
来问问有没有用过或者熟悉这方面工作的朋友，给我推荐下经济又简单实用的一套
chromotography系统。
顺便问下，如果我买单个组件自己DIY一套能省些钱么？比如pump, column,

发帖数: 1 | 42 gel filtration(GF)柱子最常用的就是GE的了. 一定要5ml的话必须是Hiload column.
16/60 的最大能上5ml.但是看你用的应该是Superdex 200 GL 5/150. 这个柱子分辨率
比hiload高.你的样品未必在Hiload上也能分开. GL的柱子有 10/300的,24ml bed
volume.能上样0.5ml. 如果溶解度不是问题,浓缩10倍上这个柱子好了. 另外,你真是就
只过一个GF柱子没有其他比如affinity column? 蛮奇怪的.
GF的柱子都是要HPLC级别的pump才能带得动的. 这样的系统不可能便宜的. 要省钱就放
弃那些自动功能,诸如多个inlet,outlet还有多柱切换什么的.AKTA的purifier可能便宜
一点.
DIY就算了吧.这些公司的系统设计都是不考虑兼容其他公司部件的.自己组合太折腾了. |

注意事项
1，PhD不能申请Bachelor 或master的工作。
2，没卡没戏，政府工很多需要公民。
3，工业界经验，没有没戏，学校经验归零。
4，自动化机器或系统，如手工纯化蛋白，不会用AKTA Avant没戏，养过细胞，不会上
10L以上的自动发酵罐没戏，等等。
其他的自己去对对吧，以上基本一票否权。如果看完后没有问题，祝福各位。
如果有问题，以后别觉得PhD去公司很容易。 |

点功夫买个灯泡自己装，900刀搞定。这年头经费紧张，越来越多的实验室受不了大公

发帖数: 288 | 50 前一阵看一个公司招人的广告，上面写的是 找phd 要求会蛋白纯化啊，会filtration，
尤其强调会 AKTA FPLC 之类的，觉得自己都特符合，结果被告知人家只要化工专业的。
回信说，我biology 的background很强，不合适他们。。。＝（
我是传统生化，成天的提蛋白，所有的系统，bacteria，yeast，insect，mammalian
都练过。还成天的develop assay，每天眼冒金星的计算那十几 二十个 大k，小k的，
，，，
结果发现，找生物的工作，不如养细胞的和做动物的，找做蛋白的，又不如学化学的。
班上的前辈们，大家觉得，现在industry 这方面的opening 还有吗？还是说，不需要
招phd
来做了？我该找哪个方向的呀？
或者，我不应该太注意他们要求什么技术，而应该注意我做的project 是不是match那？
可是，一般的广告上，只写会什么，会什么的，没有说具体是那个方向呀。。。
都想找块豆腐撞墙算了。。。 |
