下面的论文发表的时间，是中国两种转基因水稻一种转基因玉米“终于”获得安全证书
、并引起全国转基因大辩论的时候：2010年2月。
让我们看一下摘要的简单译文（全文未见）：
“用近红外反射法，气相色谱-质谱法、高效液相色谱法、电感耦合等离子体原子发射
光谱法，并以化学计量法研究三种（分别）可抗霉菌病和抗虫的转基因水稻中重要的营
养物质成分组成（按欧共体标准），发现其营养成分组成和物理性状有非预期改变，其
中蛋白质、三种氨基酸、两种脂肪酸、两种维生素及其他数种（微量）元素出现了非预
期的构成变化。差异幅度：氨基酸20-74%，脂肪酸19-38%，维生素25-57%，（微量）元
素20-50%，蛋白质35%；植酸没有显著变化。相比于非转基因水稻，转基因水稻的营养
成分构成和物理性状的非预期改变，可能与基因的转入有关，阐明变化后的作用需要经
过更多研究。”
需要说明的是：这项研究所用的上述几种方法，全都属于常规分析方法，不是专门检测
转基因成分的高新技术，因此这项研究似乎不是针对转基因成分的（网上尚未见全文）

问有没有和aromatic amino acid无关的测定方法？
已经知道不行的有BCA assay, Lowry assay, Bradford assay, Biuret assay,A280换算
。amino acid analysis也试过了，不知道是不是操作的问题，也没有结果。

chemistry. It is probably not the most cost-efficient way, but we did get
huge amount of Halo-Atto647N. Hope this would help.
Reagents and Reaction Set Up:
Dissolve 5mg of Atto NHS ester in 400ul of Anhydrous N,N DMF. (Final

phenomenon. Phage display is better than mine in terms of sequence space if
we are talking about the 20 amino acid residual due to the NGS thing you
mentioned. But my method is simple but limited to MS detection. I am
thinking to use it to none natural amino acid residuals, e.g. D-amino acid.
As such, no genetic methods is possible (or nearly impossible).

我的体系其实不会遇到这么极端的情况，其实就是很单一的L构型的芳香氨基酸，是纯
的物质，而且我想在水溶液中也不会有什么激发波长或者吸收光谱区分和明显的isomer
,tautomer存在吧。
我提出这个问题是因为看到文献里说L-3-AMINO-TYROSINE他们用288 nm激发，但是我自
己去测了一下这个非天然氨基酸的吸收光谱，吸收的峰值在280 nm和274 nm,288 nm已
经是很弱的吸收了，所以不太明白文献里为什么说激发3-L-AMINO-TYROSINE选择的是
288 nm,这样就可以避开体系里phenylalanine,tyrosine的激发了么，虽然tyrosine,
phenylalanine的量子产率都低很多。

group太贵，只好自己从DNA template trancribe出RNA，同时加amino-GMP在
5’加amino group，再从amino group变成其他functional group using NHS
ester.
我不是化学出身所有上述都是第一回做，提心吊胆，每步的yield也不知道怎么
确定。不知道有没有公司会

Adding H+ can be a simple solution. If you know the pH value, and also the
pKa of the amino acids, then you can selectively add H to some amino acids.
But if the pKa value of an amino acid is about equal to the pH, then I am
not sure whether you should add H or not.

1st time 2.7 mm, they gave me a possibility of down syndrome of 1/270 ( I
was 27 at that time), I decided to not do the amino test.But when I did the
ultra at 20 wks, they said nuchal fold is big and is an independent test of
nuchal transluency, then re-calculate the possibility to 1/100. I finally
did at the amino at 22 wks.
2nd time 9.9 mm (a big hygroma)
this time 1.8 mm
It was said that NT cutoff is around 2.5-3 mm |

意观察孩子的小手，如果他的手一直是攥着的，不能张开并且手指自由的活动，就比较
危险了，唐氏儿做大B超的时候，手基本上都是张不开的。
但是如果你也担心孩子有其它基因上的问题，就直接做amino吧，毕竟amino查的不仅是
唐氏，还可以查出不少其它问题。 |
