发帖数: 5365 | 1 接触凋亡不久，做Annexin V-PI staining 看到有Annexin V-PI+的细胞群。按理说
PI阳性的细胞胞膜一定不完整，Annexin V也会结合上去的啊，是什么导致这样的细胞
群？
我想来想去觉得可能是debris，但问题是我看的结果是凋亡而不是活细胞，这种情况下
gate( flow cytometry)基本上设了thresh hole后绝大多数events都要收集的，是不是
这样造就了这么一群“细胞”？哪位比较熟悉flow 和apoptosis的牛人请指点以下，谢

主要是被我们实验室一个号称一直做cell death的德国博士后唬住了，那家伙意正严辞
地宣称不可能PI 进去了annexin V还染不上 ，让我这个刚开始做apoptosis的菜鸟迷惑
了一下。
我做flow的时候用了fluorence intensity thresh-hole, 用的gate也是离了一点距离
到原点，按理说应该把最小的碎片给排除了，这种PI单阳性的细胞是不是死亡的时候胞
膜出现了PI可以穿透而Annexin V不能进入的小孔？或者是Annexin V在细胞内和PS结合
不完全造成染色阴性？ |

发帖数: 287 | 3 我用过 Invitrogen 的 Annexin V-Alexa488/7-AAD kit，蛮好用。为了回你这个帖我
去 Invitrogen 网站的 Fluorescence SpectraViewer 查了一下，结果贴在后面了。
RFP 的 excitation 和 7-AAD 一样，但是 emission 不一样。RFP 的 emission 是红
色而7-AAD 的 emission 是红外的，flow 机器应该能把两个分开。如果 RFP 的表达量
和细胞凋亡没关系的话更应该如此。真的想省心可以考虑 Annexin V-Alexa488 和
DAPI 一起用，这样三个颜色分得很开肯定不会有问题。 |

很好用，7-AAD或者PI搞multi-color staining有点难度 |

而TUNEL 是根据DNA damage来测Apoptosis. The method should only identify cells
in the last phase of apoptosis.对吗? |

PI+only (%#) --necrosis (dots on the Left side top)(died)
PI+/Annexin-V+ (%#)--later apoptosis (dots on the Right side top)(died)
PI-/Annexin-V+(%#) --early apoptosis (dots on the Right side bottom)(Not died)
PI-/Annexin-V- (%#)--alive & No apoptosis cells (health cells)(dots on the Left side bottom)
I use The sum of "PI+/Annexin-V+ (later apoptosis)" and "PI-/Annexin-V+(
early apoptosis)" to count Total Apoptosis (%)

PIsorting 最上边的是死细胞，下边是活细胞，然后中间就是凋亡细胞。但问题是中间
的细胞很大一部分都是 FITC Annexin-V negative。该怎么解释阿？ 细胞没有固定和
permeablize。下星期要给老板汇报。不知道怎么解释。特向大家请教。提前谢谢你的

发帖数: 287 | 11 有细胞重新做一次么？如果有的话加一个 permeablized cell 做 positive control。
膜破掉了之后 PI 和 Annexin 都可以自由进出细胞，所以这个 condition 应该是
Annexin 和 PI 都 positive。如果这个条件都做不出信号说明你的试剂坏掉了，要是
这个条件有信号但是你的样品没信号那就是诱导凋亡的条件问题。这个实验做完之后不
管结果如何你都有东西和老板讨论了，good luck! |

发帖数: 236 | 12 在贴壁细胞中siRNA了某基因，之后就有大量的碎片漂浮在medium里，镜下看就是小圆
泡，diameter < 10um，而细胞即使trypsin消化下来之后也并不是规则的圆形，这些碎
片并不能uptake trypan blue，计数活细胞(trypan blue-, diameter > 10um) 的数量
有明显减少, 但以trypan blue+ 的％算，并不比对照组减少很多，大概从95％减少到
90％
用FITC-Annexin V和PI double staining，做flow没有看到明显Annexin V+细胞群，
AnnexinV+ PI+细胞群会增加但也不会增加太多(本身％也小于10％)
用50uM Z-VAD-FMK caspase inhibitor 与siRNA共同处理细胞, 这些碎片就不会产生，
计数活细胞的数量也不会减少, 和对照细胞没有差别
请问这是否说明细胞有死亡？能否看出是何种死亡方式？谢谢大家! |

morphology in microscopy.
Since all control work well, you should trust the FACS results. |

也有可能是ANN V 抗体(连FIC)的不work了(时间长了,污染了...).(我就遇见过)
当然,如果apoptosis多,也有可能少量的PI+ ANN V-
建议用新KIT再做一次. |

另外看有没有caspase activation with western of cleaved Caspase or luc
substrate.
不是所有的death都值得研究的，除非，你看到的death有具体的pathway
我们做screen的知道，很多compound扔下去细胞都会死。 |
