织中，在大脑皮层质及神经突触内含量很高。在外周组织，血清素是一种强血管收缩剂
和平滑肌收缩刺激剂。血清素还能增强记忆力，并能保护神经元免受“兴奋神经毒素”
的损害。如谷氨酸即对受损的神经细胞有很大的毒性，因此充足的血清素能在老化过程
中防止脑损害发生。

这篇paper进一步指出了STING,cGAMP这条pathway的重要性!
这篇paper用的是HIV/Retrovirus病毒感染的model，另外一个就是前面paper里面展示
的某个自身免疫疾病。
在STING KO杂交的disease model上，auto-immune disease几乎100%被block.
如果可以基于这条pathway开发出anti-autoimmune的drug，基本上是Nobel Prize级别
的。

这个study牛b之处在于，这是第一个metazoa的cyclic dinucleotides，有开创性意义
所以任何cGAS相关的科研都很容易出cns，自从今年jan的backtoback science papers
出来之后，今年cns上面好多片了
单纯从STING的重要性来说，目前还不知道啊~ cGAS是个新gene， 希望以后genome
study把cGAS划进去，如果cGAS真的跟尼玛autoimmune disease很有关联，那就牛b了，
当然，this seems very unlikely. |

他已开始猜了个病就和我们宝宝症状非常一致，可惜后来发现不是；而且他不是代谢病
专家。
问题是坚持宝宝不吃长链脂肪酸的奶粉后，他奇迹般地改善了；完全出乎医生们的预料
。我感觉医生列的是常见的代谢病，而我们感觉宝宝吃长链脂肪酸似乎是慢性中毒，大
概需要一个月左右才能体现出来。因为过程很慢，我们怀疑很多方法查不出来。
我们最大的问题是如果是这种病怎么查？不能确认的话他一辈子吃饭都受影响，基本也
就很难出门时间长一点了。尤其是他的肝已经在要换的边缘。医生们现在改口说他的病
程都是偶然，可我们不能同意。为什么每次都是长链吃一段时间就得病，尤其是这次停
了鸡蛋后马上改善。。

了，我自己也是得过acne的人，当时得了我自己觉得每天都很阴郁，心情不好啊！而且
我保证如果真的有药可以specifically target那个致痤疮的bacteria的话，其实比任
何药都赚钱，至少我愿意付大价钱买那个药，即使医疗保险不cover，相比下得什么
autoimmune diseases， cancer，inflammatory diseases的比例，得痤疮的人相对多

给楼主一点儿我的看法
1. 1X 2X protocol都试一下，看看能不能做出来什么expected results （KO more
severe），这样最好了
2. 考虑到study的novelty，建议楼主先把那个cytokine减低一倍的数据放在一边！ 先
把EAE实验完善，因为你应该相信in vivo的数据，这样更容易build story！ 广泛的读
EAE的paper，比较microarray数据，研究一下有没有可能是别的gene被up or down-
regulated！！ To be honest, 一个cytokine production变化一倍，其实不算一个非
常drastic的difference！
3. 如果有可能，把EAE in vivo的细胞sort出来做microarray。 把in vitro的数据留
着，看以后能不能再写一个story
4. 广泛读paper，看看哪些in vivo实验里面，相关gene或者cytokine变化 correlate

摘要
1970年代早期，多数中国科学家，在文化大革命中努力生存而无机会开展研究。两位年
轻的研究者屠呦呦和张亭栋，分别在发现抗疟新药青蒿素和揭示砒霜化学成分三氧化二
砷对白血病的治疗作用的过程中起了关键作用。回顾四十年前开始的历程，不乏曲折和
反讽。青蒿素工作源于中国帮助越南抵抗美国，三氧化二砷源于观察、验证和改进乡村
中医的实践。虽然他们的药物挽救了世界上很多生命，两位研究者迄今未获国内外充分
肯定，屠呦呦有争议、张亭栋基本默默无闻。相关的文献埋没于冷僻的杂志和一般不易
看到的内部会议资料。基于原始中文论文、文件和采访，我们在此呈现这些发现的历史
概貌。没有逃脱我们的注意，在古代和近现代中文文献及医疗实践中，可能还有尚待重
新发现的珍宝。
