很多药物都会引起autophagy（细胞状态不好、DNA damage, mitochondria stress, ER
stress）等等。
如果不是特别重要的化合物，不建议follow.
如果某个特别的基因KO or express引起autophagy还值得一做。
另外autophagy和autophagic cell death也需要区别对待。
很多autophagy并不会引起death. |

键性的实验支持。搜了一下通讯作者的背景，好像也没有抱过什么大腿，他自己也没什
么呼风唤雨的能力，这样的工作居然也能发NI？是审稿人的脑子秀逗了还是我才疏学浅
看不出这文章的高深之处？

我觉得这篇文章烂的原因：
1 题目不够准确。Autophagy proteins的缺失影响了Inflamamsome的功能，这是文章里
面用两种敲除老鼠证实的，但是不能说 Autophagy proteins regulate innate immune
responses。另外就是 the release of mitochondrial DNA mediated by the NALP3
inflammasome，这个结论的实验证据太单薄了，只有Figure 6d这一个实验证据，觉着
这么核心的结论还是应该多角度证明一下的。

autophagy也是一个比较常见的降解通路，虽然最终经过lysosome降解蛋白质，但它不
等同于经典的lysosome降解(NH4Cl抑制的那种)。
常见的能block autophagy降解通路的药物有：3-MA, Bafilomycin A1 等等 还有能促
进的如Rapamycin等 可查阅相关文献。实际上autophagy最近这些年一直做得比较火热
。嘿嘿~

is a well-studied organism, often used by biologists to examine fundamental
cellular processes.
Unfortunately, its cells are small and to start with Dr Ohsumi could not

十一公做结晶时，蛋白的功能是已知的。而这个日本人是找到autophagy的关键蛋白，
还不只一个。至于他提出的autophagy这个名词，并不重要。就像apoptosis，你叫
programmed cell death 也行。关键是他找到了components。有他找到的这些蛋白并证

我觉得nibs还是挺鼓励新人做冒险的工作的，另外丰富的人力资源也很重要。
张宏的工作我比较熟悉，他回国前3－4年基本是延续phd的时候系统，发了几篇
development，db,pnas的文章，后来他们在做一个筛选时无意中发现跟autophagy有关
的东西，然后就大半个实验室转向autphagy，期间的人力和资源很是可观，记得当时
autophagy领域一顶尖牛人来了nibs了解后表示。“我绝对没有这样的资源。。。”
你能想象美国一个实验室8－10个人一起做筛选么？ 最后一篇工作4－5个共同第一作者
。。。这就是那两篇cell 的真相。
nibs的研究生和技术员聪明能干，比美国学校大多的fresh phd不知道好用到哪里去了
，另外本科留下等出国的和客座的学生不要太多。。。这也就是nibs奇迹的秘诀，你把
他们放到美国，同样的仪器，同样的经费／人工比，他们大多实验室绝对出不了这么多
这么好的工作。
神经所。。这几年优势没有那么明显了，生物物理／北大和很多地方都可以招到非常出
色的年轻人。而且庶我直言，我觉得ion里面poo/dan yang体系的人有点多，另外我可

本来想笑而不语，还是科普一下吧：
2007年之前，清华生物系和医学院基本没有任何core facility，连动物房好像只有不
正规的一小间（我不做动物，记错勿怪）。到2010年，细胞平台、蛋白质测试平台、动
物房等core facility基本建完。以前在Rockefeller的邓海腾加盟清华执掌Mass Spec
，对我们帮助巨大！在那之前，总是去NIBS。没有这些基本的facility，不管你多牛，
举步维艰，哪有什么竞争力？我私下经常对后来的人忆苦思甜。我当年去清华的时候，
