最后一张图里的bead应该是塑料的，和fly上的bead作用不一样。fly用bead，主要是模
拟昆虫的圆头（不少昆虫这样，例如蛐蛐），用bead chain模拟昆虫眼睛。另外一个作
用就是加重，bead一般是金属的，沉水速度快。
楼主钓组最后一张图上的bead，我一般是用来调整固定高度（长度）的，用力时可以滑
动，调整一些间距等，很好用。
rig2图上的红色bead，估计有些引诱鱼的因素，例如我们常用荧光bead穿在leader上钓
海鱼。或者可能是助浮的，底钓鳟鱼的时候，有时候leader上穿浮球，使得钩子和饵离
开水底。 |

有GFP, Flag等tag.Overexpression 做Co-IP.
先做A和B的相互作用，用protien A beads，IP后boil.先IP B, WB A.对照有（1）non-
transfected, （2）only transfected with A, 试验组（3）transfected with A and
B. IP后发现（1）没有A,（2）中有weak band of A,（3）中有strong band of A.
然后怀疑A蛋白太大，可能和beads或者antibody for IP有non-specific结合，所以（2
）中有weak band of A.之后change strategy: IP A, WB B.后来发现有同样情况。
请问下大家有没有发现蛋白和protein A beads non-specific结合的情况？

bail。
但是还有两个问题：那覆盖在珍珠上的一小片金属是什么呢？好像不是bead cap。bead
cap是不是一般用在穿一串bead的时候，用在bead的两端，而且比较大，全部盖住bead
头的？
最后一个问题是，珍珠怎么挂到bail上呢？

照；磁珠beads。发现在裂解液中加入beads-抗体后，beads会有不同程度的粘结.总体
趋势是H3〉目标蛋白〉IgG。 其中IgG-beads几乎没有粘结的现象。这个观察可以理解
为不同抗体富集蛋白的能力和总量不同，H3最强，IgG最弱。这种粘集在不同的wash
buffer处理时，没什么改变，但是当最后用TE洗的时候却一下子消失了。似乎TE破坏了
蛋白之间的相互作用使beads变得不再粘连了。
问题是TE中并没有什么特别的成分，除了不包括detergent和LiCl，Tris和EDTA的浓度
和最后的wash buffer相同。PH值TE:7.99；wash buffer:8.14也没大区别。
请同学们指教一下，为什么加了TE后，beads不再粘连了？这种现象是否暗示有蛋白产
量上的损失？以及最后一步如果省略在wash buffer 1,2,3后用TE洗beads会有什么结果？

我看protocol 通常有3种
1）先加Beads到样品中，然后把上清移到另一个管子里，然后再加beads和抗体过夜。
2）先将Beads和裂解液（不是样品）混合，rotate 1h，然后再离心，把preclear的
Beads+抗体+样品液 混在一起4度过夜。
3）将抗体和样品4度过夜，然后把Beads加进去，4度转4h左右。
3种方法，哪种背景更少啊，还是说差不多？谢谢大家了 |

---- 枪
用什么枪？每个刚入门打飞碟的射手都有这个问题。最简单省事的回答是，只要是霰弹
枪，都可以。没有规矩说不可以用平双，也没有规矩说不可以用16 gauge, 20 gauge,
28 gauge, 和410打trap；如果仅仅是一年去碟场打几次，每次打中一部分碟就满意了
，那么用的枪真的不是太大的因素。
但如果射手的目标是经常打，而且追求打尽可能多的碟，那选用的枪就很重要了。我自
己挑选碟枪以及帮助他人挑选的时候一般用一下的标准：
1. 长管
Trap的目标绝大多数是在30-40码的距离上被命中的，所以枪的指向性要很好；长枪管
的指向性远远好于短枪管，因为sight radius更长。参加trap比赛的枪，30英寸的枪管
是最短的，32英寸非常普遍，34英寸的也不少见。关于指向性的重要，在下面的技巧帖
中更深入的讨论。
有些人说用长枪管是因为初速更高，这是不确切的。霰弹枪和步枪不一样，弹丸在管内
并不形成有效的密封，而且火药一般在16英寸左右就燃烧完毕了，不存在有效加速，所

关于protein-beads ratio，我labmate用的方法是：先用少量beads，
bind protein at highest coverage，这个batch用来做beads可能带来background
的实验。然后recycle supernatant，加过量beads，把能bind的protein都bind上。
感觉比较make sense，就是有点麻烦呵呵。
btw，以前用M-270 carboxylic beads，现在改用这个，发现capacity低很多，我
随便就要用几百个microliter。。。

怎么个crosslink法？我是加了1st Ab overnight再加beads的。
还有elute我是用的SDS sample buffer，然后就boil，100 C，10min。boiling beads
和elute是两个独立的过程呀，为啥elute可以取代boiling？是我理解错你的意思了吗？
关于ECL plus，我觉得用ECL曝光久一点，貌似也跟plus一样的效果，而且plus还会令
blot颜色深浅不均一，不知道你会有这个问题吗？如果有，怎么解决比较好？

Crosslink Ab to beads with Imidoester
Crosslinkers (Pierce). Incubate Nuclear extract (or whole cell lysate) with

After washes, elute the co-purified protein complex with low PH glycine
solution instead of boiling beads. |

thrombin切后，离心，此上清为eluate 1, 然后再用cold 1x pbs洗4次，跑胶，发现没
有目的条带，几乎所有目的条带都
是在最后beads fraction里。怀疑是目的蛋白本身和beads结合，而不光光是通过GST－
GSH结合。
然后用reduced GSH换出GST，然后用1M NaCl洗beads，希望能够破坏蛋白和beads间的
作用，但是没用诶。。。

2 reasons
1. ChIP buffer contains SDS and SDS kills M2 beads (refer to M2 beads
handbook)
2. you never know how much antibody you used. 10 ul of M2 beads perhaps
contain hundreds ug of ab. and saturate your chromatin. you need titration,
but impossible.
you may want to try M2 antibody which is a monoclonal antibody and the same
as the one conjugated with M2 beads.

首先对小娃来说，减法是加法的 “reverse”，小娃能理解。。。但除法是乘法的 “

从artbeads.com 买了1oz 的silver wire, 几个crystal的beads, 然后开工。
用silver wire 弯了18个link连成项链，
再弯出6个方块，
和3个水滴形的frame.
用小圈连起来，水滴形的bead吊在frame中间，
再用粗些的wire 做成坠子，把前段敲扁，剪成椭圆形。照片上是修剪之前的，所以形状
比较怪异。
全部连起来，穿上beads，再加上wire的钩和环就完工了。
试戴效果。
总共用了将近1oz silver wire, 9个bead,大概30刀，两天时间。 |

crimp bead固定住，第二根线就偷懒直接拿22gauge的wire来扭了……
唉，看来我要么需要添置一个crimp plier（这个工具貌似可以把crimp bead对折），要
么需要再去买一些尺寸大一点的crimp cover了 |

发帖数: 1604 | 39 嗯，就是incubation前后各取样测浓度。我是用Pierce的BCA assay kit做的，因为我
的peptide没有280吸收峰不能直接nanodrop测浓度。但是因为我样品少，用的是
microscale做的，所以incubation之后的浓度实际上已经很接近noise水平了,真正的
binding efficiency我感觉应该会超过80%的。
Streptavidin beads的instruction上应该有说它的binding capacity，一般用的标准
是mg biotin单抗/mg beads，你得用你的蛋白的size和单抗的size大概齐毛估估一下，
估计一个适当的蛋白浓度来做incubation，beads多一点没关系，蛋白用多了就可惜了。
嗯，不过beads也不便宜就是了。:)

Cells carrying beads are bad for their trafficking in vivo. But magnetic beads enrichment is quick and relatively economic, as compared to sorting. For in vitro study like RNA purification or culture, beads purification is fine.
Sorting is expensive and labor-intensive (I suppose your lab do not have
your own sorter and do not sort cell frequently). But without beads binding
, cells are more healthy. |

如果都一致的就无需担心啊。
另外我一般用20ul beads，貌似远远小于大家讨论的量。可我觉得20ul handle 1mg以
下的总蛋白足够了啊。当然这取决于你拉下来的那个蛋白量。我拉的是一个早期信号蛋
白，一种adaptor protein。可能不多。我觉得beads多很annoying，因为会带来非常大
的IgG band。当这个band非常强的的时候会类似屏蔽那种，影响周围方圆几百里的弱信
号。如果你分子量离50不远的话，我觉得70都很费劲。所以我情愿用少一点beads。 当
然如果做那种crosslink beads的就不存在这个问题了可能。我说的是经典ip。不知道
我的想法对不对，请教大家。谢谢。 |

刚刚做完最麻烦的部分，拆旧的drywall joint bead
不知原来为什么用铁铝类的，不防潮啊，都绣得很薄了 (因为drywall joint 很靠近浴
缸) 用锯子锯下来，然后铲除周围的漆和compound
磨平了后，换上Vinyl joint bead (不需要用fastener, 卖joint bead 地方有卖3M的
喷胶，喷出来是红色的)
然后上了compound, 等24小时，明天就进入技术活阶段，打磨compound 表面
进程算是过半了 |
