值得一看. 我在SFO-ICN的航班上看了两遍, 强力推荐.
此片熔合了诸多元素, 一般看过几部片子的都能轻易识别出骨干情节就是基于爱情动作
片的套路,咳咳,表想歪了, 是结合了爱情片Groundhog Day和动作片MI1/2/3的套路. 但
相比靓佬汤致力于动作片事业几十年来层出不穷毁三观不倦的好片中片滥片, 又多了点
内涵; 与此同时, 相较于Groundhog Day, 此片又平添了一些戏谑. 实在是一个不伦不
类的杂交品种; 但盖棺定论+平心而论, 杂交得还挺不错.
与Groundhog Day这部彻头彻尾的男性片所探讨的主题不同(这个可以单开一片论文了),
EoT更多考虑的是: 如何以最简单最高效的方式进行周而复始日复一日的游戏通关. 这
对靓佬汤几十年的影艺生涯无疑带有一点黑色幽默的色彩---要知道,当年别人可是瞄准
了小金人全力进军的,什么Born on the 4th of July, Rain Man, & Jerry Maguire.

1 purify your PCR product with qiagen kit
2 use T4 PNK phosphorylate your PCR product's blunt end
3 ligate your blunt PCR fragment into any vector cutted with blunt end enzyme
(like EcoRV)
4 transformation and mini prep plasmid
5 then use hindIII and EcoRI cut the fragment out of vector
6 purify your hindIII-EcoRI fragment with kit and insert it into the vector
you want to use
this will add one more subcloning step but i am sur |

条件反复重复。
1 当初选用pQE8也是没有办法，因为pQE9我们实验室没有（pQE8和9的区别就是8在酶切
位点之间只有一个碱基，而9要长得多）。最开始的时候没有意识如此近的酶切位点很
难切下来，最起码同时酶切不可能切下来。但是只尝试过一次顺序酶切不行就没有再尝
试用pQE8了，我不记得是先切BamH1还是Hind3，后面尝试插一段片段在2个酶切位点之
间再切。
Nice alternative, shall solve the problem in this case...but I would just do
blunt ligation, nowadays, the fast ligation kits from different companies
such as Roche, takara, don't really care about whether it is sticky or blunt

网络写手刘某某，笔名“天一”，写了本名叫《攻占》的耽美小说，并拿来印刷贩卖了
7000册，从中盈利了十几万。刘某某撰写的小说《攻占》
所谓的“耽美小说”，相信很多年轻的盆友都知道，是以刻画描写男性之间的爱情为主
的。
但“男性爱情”本身不是这次的重点。重点在于，天一的小说里含有大量淫秽色情的描
写……
2017年，一本名叫《攻占》的书籍突然在网络上火爆起来，短短几个月时间，该书籍就
通过网络销售数千本。而这样一本“畅销书”也引起了国家扫黄打非办的注意。经国家
新闻出版广电总局出版产品质量监督检测中心鉴定，鉴定样本《攻占》为淫秽出版物。
办理此案的芜湖县公安局治安大队民警朱佩告诉记者，《攻占》是一本淫亵性地具体描
写男男同性恋性行为的书籍，其中还充斥着大量与性变态有关的暴力、虐待、侮辱等行
