他提到一点,当时他们那片paper一开始投了science悲剧,理由是“we hadn’t made
any scientific discoveries beyond finding that channelrhodopsin-2 functioned
well in neurons”,这话单独来看道理上无比正确。在这片Paper之前,大家已经知道
Chr2能感光也能响应光开放cation channel了,所以这篇文章确实只说明Chr2能在神经
元里表达并工作而已,什么新机制都没提出。后来发了Nat. Neurosci.
但是呢?把Chr2和neuron两个概念联系在一起,不需要任何别的机制,就已经是革命性
的突破了,因为可以从此用光很有效的(这个定语也重要)控制神经元的活动了。这个
东东弄个Nobel不成问题,如果Chr2自己发展的好,Ed和Karl分享这个nobel也不是完全
没有可能,尽管不大。可惜reviewers看不到,人家就觉得没有机制,没有新发现,完全

神经元甘氨酸受体通道的电生理研究,后来做过GABAA受体还有protocadherin蛋白的研
究,但我对老年痴呆之类的一点概念都没有,现在上网查也只能了解各大概,也没找到
老年痴呆和我熟悉的GABAA受体或是protocadherin之间有关系的相关文献,有没有高人
指点一下该怎末入手设计这个面试课题啊,万分感谢。
补充一下,先前通email他们好像对我的电生理研究背景比较感兴趣,另外我熟悉的实
验技术包括细胞培养,免疫荧光,western blot,基因克隆等。还有我查了一下,他们
研究所有人做β-Site APP Cleaving Protein 1 (BACE1)和CutA Divalent Cation
Tolerance Homolog (E. coli) (CUTA)的研究,还有人做AD小鼠模型。

sunnyday's desalting method is the best to try. I will be surprised if it
doesn't work. I guess you have to use a syringe because nobody likes to
inject 32P into their FPLC. In that case, you should be careful of the
fraction size. Don't mix two well separated peaks during fractionation.
kaka1000's vivapure spin column离子交换柱 is a cation/anion exchange column,
to which unlimited volume can be loaded. In the end, elute with B buffer.

the single negative charge. These negative charges tend to blow the double

要把固态物分散成以水为分散介质的Emulsion或Suspension。要求粒径在2-20 microns
之间,anionic或cationic particles,常温稳定,Shelf Life一天足够,当然越长越
好。 浓度没要求,别太稀太稠,能够泵输。
基质和水的密度接近,不溶于水,惰性。 我觉得做成稳定的Suspension应该比较困难
,正好它溶于丙二醇及芳香溶剂,把它先液化然后再做成Emulsion比较好。
做Painting和Coating的当然是专家了,但都是核心机密,不会告诉我的,虽然我根本
不涉及他们的领域。 网上也少有资料。
我搞出一个配方,但觉得应该有足够的空间可以提高,哪位高手能有建议没有?
包括:乳化剂及用量,乳化器及乳化条件,等等。先谢过。 |

cation,阳离子这个, 用的金山词霸念“开审”. Lingoes念"开太恩"
这个还好说,偶一般念后边那个。
但是crystallization呢,念"crystal-来贼审" 还是 "crystal-太嗯"?
基本上,遇到lization等偶都习惯性的拼成"来贼审"...
但是在词霸和lingoes上的发音大多是"太嗯".
身边没人指导,求助大家怎么念比较合理,地道

把一个电学元件(化学电源、蓄电池、电解电容、二极管等等)看作一个整体,则电流
(正电荷)流入的一端称为阳极anode,电子(负电荷)流入的一端称为阴极cathode,
这是普遍适用的原始定义。
衍生词汇有:阳极化anodize,阴极射线cathode ray
与此有关的概念是正极“+”和负极“-”。正极指电势较高的一端,负极指电势较低的
一端。在大多数情况下,阳极和正极、阴极和负极是重合的,因为电子会自发从电势较
高的一端流出。但是对于化学电源和原电池而言是个例外,因为化学势的存在使得电子
能够克服电势差反向流动,使化学能转化为电势能,所以此时阳极是负极,阴极是正极。
另外容易混淆的是阴离子anion和阳离子cation。这两个术语原来分别指在电解质溶液
中趋向anode和cathode的离子,曾被翻译为“阳向离子”和“阴向离子”,现在泛指带
负电荷和正电荷的离子。

有人用过这个柱子吗?为什么,我们的每次总是用了几次后,用同样的标准样(std)
去测,peak area 值就会变大呢(相对标准偏差还是很小,也就是系统的rsd还是好的
,就是同样的东西,测出的peak area 变大了)?看起来这是一个cation-exchange
column,有什么要注意的呢?有什么方法能恢复呢?
另外,通常HPLC的柱子使用寿命是多久呢?我们的这个新柱子刚用了一个月,就变成这
样了。我们对于UNKNOW的值得测定是建立在unknown和std的peak area 的比较上建立的
,而不是建立标准曲线。所以直接导致unknown里的含量结果偏低。所以,我也有点怀
疑这个方法的有效性。
当然,我们每天要测20个样品左右。。这个会用影响吗?我都不好意思跟老板讲这个柱
子又有问题了:(

CH3SO3H,浓度是10mM,流速0.5ml/min,柱子长35mm,用ICS3000时 三个离子的保留时间
都在1.3min左右,用DX600,Na 和K保留时间0.8min,Ca大概2min,用CS12A试时,相同
条件下,钠的保留时间大概8分钟左右,钙大概28分钟,ICS3000因为别人也在用,
injection loop是2ml,这可不可能是原因呢 |
