发帖数: 1125 | 1 挺拥挤的领域了，基本的新组分／转运／和药物／表型筛选已经做烂了，在疾病发育和
高等动物里的功能也有不少人做了，领域里面大牛灌水文章不少，真有意思的倒不是特
别多。
资金还好吧，至少能扯上不少疾病，很多原来做centrosome的现在都转做cilia了，比
纯做centrosome好骗钱..
如果能找到新的结合点挺好的，一般的研究的话很拥挤，除非进大牛实验室，否则不推

现在我比较感兴趣的是canonical wnt(Wnt/beta-catenin)和Non-canonical Wnt(主要
是Wnt/PCP)之间的相互作用和联系。个人感觉，一般前者和细胞增殖和分化相关，后者
和细胞极性和形态发生相关，二者往往是相辅相成但又有拮抗作用的。二者之间的调控
又和cilia相关，cilia的作用最近几年以来一直在升温。Wnt在胚胎发育、心脏肾脏等
器官的发育和修复、某些疾病和癌症里都有很重要的作用。像一个大谜团，很有挑战性
。。。
我继续准备周三的口试了，改天好好聊。^_^ |

发帖数: 1015 | 8 centrioles 相互垂直是不是由于结构需要，毕竟在没有成熟前他们需要互相支撑？
在之后的cell cycle， centrioles靠近核膜，会形成有丝分裂的纺锤体。他们靠近
cell membrane,会形成cilia。他们在成熟过程中肯定需要不同的蛋白复合物，但我觉
得不太可能二者之间有什么大的差异，毕竟功能相同。
至于疾病，由于centrosome or cilia-related 疾病很多是遗传性的，比如BBS，NPHP,
MKS，很多胎儿在怀孕期间就流产了，存活下来的也是症状太严重，药物应该没有什么
作用吧。

是，这个两个蛋白公转到hela细胞中，看不到任何的共定位。其中一个蛋白和cilia形
成相关，能到看到它定位在basal body，另一个蛋白功能未知，弥散分布在细胞中，细
胞核信号强一点。自己不是做cilia，对这块不熟悉。请教各位大牛，如何才能看到共

我们实验室做海胆和海葵的胚胎早期发育中体轴的建立和胚层分化，主要就是Wnt/beta
-catenin和Wnt/PCP这两个通路。现在好像PCP比较热门，和cilia、细胞极性、体轴发
育、果蝇的单眼和翅膀的毛，高等动物内耳毛的发育等等基础问题有关，又和cystic

1。为什么垂直？
－没人知道，有一些推测，但是没有数据支持。。。也算是这个领域的一个有趣的悬案
，我老板自己对这个问题很有兴趣。
2。Procentriole和matured centriole的区别
－组成差别很大，比如appendage只在mother的distal上面，
－另外有很多蛋白只在procentriole (sas-6)上表达，有的只在matured上表达(cp110)
，有的都有表达（centrin..)
-功能的区别研究不是很多。
3.有没有target centriole的药物。。。
－有关centrosome cluster 的小分子筛选做过了。
－有关centorosme reduplication的也做过了。
－一般的centriole duplication 的小分子。。这个就是我的一个子课题。。。
问题是centrosome和很多疾病有关系（小脑症），但是太rare，跟cilia相关疾病比起
来根本没有重要性，而centrosome跟肿瘤的关系虽然100多年前就有人发现，至今也没

问题是centrosome和很多疾病有关系（小脑症），但是疾病本身太rare，跟cilia相关
