1. enucleate the eyes
2. slit the cornea, put the eye in the fixative (usually 4% PFA), for 10min
3. remove cornea and lens and vitreous, and put the eyecup (retina+RPE+
sclera, etc) back to the fixative for about 30-40min. 1hour should also work
.
4. wash with PBS and dissect the retina from the eyecup.
5. put the retina to PBSTriton (1%) permeabilize
6. staining part
这个protocol适用于~95%的蛋白染色。
但是如果遇到一些比较tricky的东西，可能需要调整fixative，固定时间，
permeabilization等等。

我是做眼睛的， 我做过如下的比较，
1. Whole globe: O/N fix in 4% PFA
2. Whole globe: 1hour RT in 4% PFA
3. Slit the cornea (more access to PFA) : O/N in 4% PFA
4. Slit the cornea: 1hour RT in 4% PFA
I compared immunogenicity of CD31 and PDGFRb.
Results were:
1>=4>2=3
As for the brain, I usually perfuse and fix with 4% PFA and post-fix in 4%
PFA O/N.
Today I didn't perfuse, I guess I may just leave the brains in 4% PFA for
36hours.

treatment: a. protect eyes from sun, dust, use sunglasses or goggle
b. lubricate with artificial tears 4-8x/day to reduce your irritation
c. the rest of your conjunctiva looks not inflamed, which would be treated
with topical steroid or NSAIDS
if it is growing into the cornea so that interfere with your vision, or
irritation severe enough interfering with your contact lens, consider
surgery.

you need to also compare different types of laser, visx star s4 is the best
availible in the US. Canada has the most current laser, MEL 80, said to be
much improved over visx s4. but FDA has not approved MEL 80 for US.

如太阳地里,或者商朝里,或者看大面积白色的东西,就会很吃力好像站猛了时眩晕的感
觉.要是突然到了很黑的地方,也会很吃力.
大约10天前,看过一个眼科医生,结果他上来就跟我说他是主要管retina的,随便看了看
说是结膜炎,给了点眼药水.让我去找gerneral ophthalmology的医生,还给推荐了个.结
果我打去那个医生,他又说他是管cornea的,好像不太想给我看似的.都不知道该看谁.然
后查网上,没看到谁是gerneral ophthalmology的.眼睛的问题好像起了些变化,非常着
急,主要开车很担心.
这个到底该看什么医生啊? |

contact lens use artificial tears all the time. Chronic lack of tears can
lead to permanent cornea damage.
It is probably worth looking into the cause of dry eyes. Is that Sjogren's
syndrome? |

(角膜) has nothing to do with being short-sighted. What you will need to
ask for is some published clinical evidence that it really works, not some "
famous doctor" claiming that it has saved many kids. This is why evidence-

的OD就不做CRT，做CRT的OD就都说CRT有用。我也问过我的一个眼科的MD，她对CRT有没
有作用不愿评论，因为不是她的领域，但她觉得reshape cornea，属于非必要invasive
