厄运缠身。同样是人,差别咋就这么大呢。请新锐研究者藤井聪教授从心理学的角度来
给大家解说吧。
越是关照他人利益的人越走运
看看您的周围是否有这种现象?有些人不管是工作还是私生活中,不知为何总能左右逢
源一帆风顺。相反地,有些人像是命运女神的弃儿,无论做什么事情都不顺利,越活越
出溜。那么,为什么有人幸运、有福、运势强劲,有人倒霉、悲催、厄运连连?这两种
人究竟有何不同呢?
我们提出“认知焦点化理论”来解释这个问题。这是一个以人们心底里究竟最看重什么
为切入点进行的心理学研究。
简单而言,就是从人们在待人处事时能够从多大程度上关照他人情况这个观点出发,我
们尝试对人进行分类。
请看图1。横轴表示社会和心理的距离,以自我为原点,依次顺序为家人・恋人
→朋友→熟人→陌生人……越往右关系越远。而纵轴是时间轴,表示在待人处事时思虑

you can download the cosmid sequence to your computer, and then according
to the position of tag-24 on that cosmid, you can select out the sequence
you need.
however, I never did that way. I prefer wormbase.

很奇怪,cosmid的NGS测序很难找到公司做(genome就很平常)我之前好像也发过帖子
问的,已经找了很长时间了,才找到这家公司的。确实,按理说应该换公司了,但是一
来他们熟悉情况了,没准trouble-shooting可以简约点时间,二来我确实苦于找到别的
好的地方。联系过几家core facility,反馈也不是很positive。
仁兄,有推荐的吗?我还会有很多这样的测序。

I don't have the protocol w/ me, but I believe that you can look
it up in the journal Biotechniques. It is a quite reliable method
which is basically:
1. get a set of degenerate primers, usually designed specifically
for your organism.
2. design a specific primer which binds to one end of your DNA.
3. do 3 rounds of PCR, 3 minute elongation time, 10 minute extention time.
i) very low anealing temperature (35C?) for 25 cycles. take 1 ul,
do a 1:20 (or 1:100?) dilution and

Size doesn't matter, cos a regular cosmid/fosmid would be over 30 kb. Sorry
that I don't know anything about "Cre-ER construct", but I think it's the
protein that you want to express really matters. But not the size.

Invitrogen 的DH10B是适合做大Plasmid还有cosmid的
不好的是本身有Sp抗性

Question1: So large plasmid always behave this way or not? the biggest one I
worked was 20kb, it was ok with typical condition.
Question2: so cosmids or Yac/Bac don't follow the same way as they have
multiple rep origisns, right?

1. It depends on what genes were in your plasmids. for example, it is ok to
manipulate some DNA viruses with large genome using conventional cloning
method but when it comes to some RNA viruses, you can't even clone its
fragments more than 3K.
2. I have no experience on Cosmid/YAC/BAC.
