这个问题困惑我整一年了,发出来向各位大牛求教。
当然了,套话先说一遍:一个 model 并不是 details 越多或者越少就越好,而是看多
复杂的 model 恰好可以适应这个 model 所 target 的应用。
现在切入正题:
有些组做 model 的趋势是越来越复杂和具体,横向看,越来越多的 components 被
integrate 进来,各 module 之间的 crosstalking 也越做越多;纵向看,各个
module 也越来越向 molecular level 迈进,同时越来越多地把 spatial info/
locations and diffussion/mass transport 模拟进来。
另一些组则是越做越简单,试图从大量纷繁复杂的模型中找出简单的共性来分析,或者
是从特别复杂的 |

人都能做的,既烧钱又周期长。很多时候还没有定论,全是crosstalk,牵一发动
全身,不太容易purturb one specific signaling。stem cell research也是这
样,一篇篇的好paper,说白了大都也是灌灌observations,鲜有碰触mechanisms
的。有人做stem cell differentiation期间的expressionprofiling,已经很算是
想干些mechanisms的事儿了,那又如何?profiling这种工作,其结果好比给你张
“错误概率未知,可能很大”的地图,除了他们自己人,谁敢拿它去指路?扯远了,
总之俺是尽量回避纯粹的mechanistic study,等天才们做出来的时候,俺在旁边鼓
鼓掌就行了。 |

不过不管什么方法,包括你说得专门仪器,是不是都会有一个荧光光谱overlap的问题
?overlap部分的信号就没办法分开吧,会有crosstalk,我查了一下常用label的光谱
,好像如果同时三个或更多,大多会有很强的重叠,如果滤掉重叠部分,光强就太弱了。 |

要扭转这个趋势,生命科学应该主要提倡智力竞争而非工作量竞争。要重点资助能影响
整个生命科学研究的新技术,新方法的发明(比如deep seq,单分子成像),以及能影
响全局的新发现,新观点。
添砖加瓦式的发现,比如多数蛋白的相互作用,pathway间的crosstalk,蛋白结构的解
析,基因组的全敲除,系统的筛选等等,应该列入工程性学科,其发现主要以resource
的方式发表。工程性的研究主要以公司化管理,可以招募很多本科级别的工作人员。如
前所述,能系统研究和解决这些问题的新方法,新技术,新思想应该得到更多资助。
最好的杂志在选择文章时,优先发表新意很高的核心数据,而非高大全的东西。例如两
个蛋白相互作用,只要数据足够可信,不一定非要要求面面俱到,体外,细胞,生物整
体,跟疾病的关系一应俱全。许多文章的核心数据就一个figure,但是由于杂志/
reviewer要求完整story,要求数据完备,许多推论性的数据也要做,导致近年的文章
supplemental data越来越多,工作量越来越大。

决定的经常就是politics的问题。Politics是绝对不听Science的。
还有,你这种说法,其实就是废话。就好比国内的学校,开会的时候总是教育学生要有
思考创新。但是创新这种东西并不是教育出来的,而是自己悟出来的。你自己做课题想
到一个东西,也不是别人教你要创新你才去想出来的,是你本来就有那个水平。

是非常noisy的。如何在非常noisy的micro尺度得到macro尺度上的精确调控本身就是一
个很难的问题,很多人都在研究这个。这个问题不清楚,用现有的工程方法是没办法
insulate很多crosstalks和side effects。所以我建议LZ最好先不要搞太复杂的系统。
重要的是找个大牛,先发几篇好文章,等职位安全了再挑战复杂的系统。 |

cysDML and cPILOT, 相关工作在ASMS会议报道过。他们所用的方法基本还是基于传统
方法改良如Thiopropyl Sepharose 6B resin和Cys TMT,这些基于抗体和resin方法的
富集效率比较低,从鉴定数目就可以看到。我们的富集效率可以达到97%,很轻松鉴定
到10000+ unique peptides。
目前没有同时含有iodoacetamide+phosphate的tag。phosphate可能会被phosphatase酶
切掉,phosphonate不能,因此更加稳定。
至于为什么要富集Cys peptide,因为其low abundance,essential for the biological
activity and structure, highly sensitive and reactive.

学校的校长助理是个50多岁的白人, 今天发了一封信, 说去年被诊断为ALS, slowly
losing the use of hands and legs, 所以以后就会坐电动轮椅出现在校园里工作了.
这个学校的孩子从2岁到12岁都有, 他在信里详细的列举了不同年龄的孩子看到他坐电
动轮椅会有什么样的问题, 家长应该怎么回答, 非常细心. 对大的孩子, 如果有兴趣的
话, 也可以和他讨论相关的神经系统的问题以及电动轮椅的机械问题. 还建议老师们可
以见缝插针的教孩子们平时碰到坐轮椅的人应该有怎样的礼仪, 他可以现身说法.
查了下这个病, 神经系统引起的肌肉无力, 目前只有药物可以延缓病情, 诊断以后的

to crosstalk between your high switching digital signals and or is it really
power supply related due to the high switching current sourced from the
supply as your suspicion indicates ?
If the noise indeed manifests itself on power and ground then decoupling
near the power supply would help to mitigate it. If your glitch is on the
signal only, then decoupling the supply won't help, and instead you should
