发帖数: 844 | 3 如果lz在讲附上的胶，我的看法是：
1.上面那个带是well里边滞留的，下面那个是free CTP还是free RNA不好说。但整个胶
曝光时间不够长。我做过无数EMSA，也是RNA aptamer，RNA 从MAXIscript得到后不用
纯化，直接取1 ul去跑denaturing gel，看看有没有transcripts，产量有可能因为变
化的，所以每批都要检验。我组里有个博后不信邪，在这上面浪费了几个星期，一直以
为是这个那个仪器不好用。
2.lz提到protine minus也有多条带，这在native gel里边太正常了，因为RNA有多种
folding的可能性，即使mfold只给出一种结构，我在胶里也能看到很多条带。蛋白特异
性的带可能跟RNA一下带superimpose，但是你会看到下边一条带变弱，上面一条带变强
，这样你要换种浓度的胶跑跑。
3.制胶和电泳液可以用1/4xTBE或者1/2xTGB，前者我用的比较多，这个每个蛋白性质有
关，一般都要试试。
4.ARGA胶有些时候可以替代page，想B52的RNA APTAMER只能在ARGA胶才能看到shift，

我的RNA都做过PAGE purification，产物方面应该没问题。
你说free CTP我没理解。看你下一个回复，是不是不用radio用的其他方法？
嗯我也听说过这种情况但自己没见过。不知道出现这种情况的时候怎么计算percentage
of bound RNA呢？把所有band加起来作为total？另外，怎么说服别人某个band是
complex band其他都是free RNA呢？
第二次被提醒buffer的问题了。一定要试试。多谢！

RNA in vitro transcription 用Applied biosystems的Maxiscript足矣
biotin没有试过，下来准备试试，可以避免用同位素。 |

有窗户，不知道是白天还是黑夜。在这又冷又黑的晚上，同学们都回家过圣诞去了，一
个蓬头散发的小女孩在办公室里坐着推公式。她不能回家，因为她还没有文章，没有文
章怎么申请博士后啊？她从家里出来的时候还穿着一件外套，但是有什么用呢？那是一
件很大的外套——那么大，不知是哪一年买的。为了推公式的时候更方便，她推导写草
稿纸的时候就把它脱掉了，同学们常常嘲笑说，那件外套还是超对称刚提出那个时代的
款式。
小女孩只好一个人做推导，一双小脚冻得红一块青一块。她面前的文章划得满满的，桌
上的草稿纸上一个式子写了满满三页还没写完，其中还包括很多的点点点[1]。这一整
天，她都没有啃一口汉堡王买的一块钱的汉堡包，只是天黑前到系里最有钱的做伪科学
的生物物理实验室的全自动特浓咖啡机那儿偷偷打了一杯热水喝。公式又长又难，根本
推不出来，谁也没帮过她，谁也帮不了她。
人们都在准备圣诞礼物，高高兴兴，欢欢喜喜，小女孩多可怜啊！她又冷又饿，哆哆嗦
嗦的推公式，桌子上摊满了草稿纸。发黄的日光灯管把光颤抖地砸在她的干枯的长头发

facial drooping, left hemiplegia and severe back pain betweeen the shoulder.
Upon arrival, no cranial nerve involovement,no language deficit the strenght
of LUE/LE improved to 3/5. CTA/CTP/CT no signs of defici. Out of scan room,
patient kept improving, strength now normal excpet left hand intrinsic
muscle 4/5. Patient was making jokes all the time and pain was also much
