版上有人用过么？求一下经验，怕被拒绝。
1.Benefit里面写的max coverage per item $10,000 这个能多次claim吗？
比如我这次摔坏镜头了，修了500,下次又摔坏了又要修300，或者后来又被偷了，还能
继续claim吗？(直到赔到购买的价格？) 还有来回的shipping cost能否包含到claim
里面？
2, 里面有一个规定The item was not reasonably cared for to prevent the
damage or theft。这个很模糊，如何界定？比如其实我是因为自己失手摔坏的，能否
说我摔坏镜头是因为三脚架质量问题，三脚架不稳自己倒了。这是合理的理由吗？
还有theft哪些情况有效，放在车里被偷了是否有效？镜头在邮寄的过程中，快递员放
在apartment门口被偷算吗？握在手中，被人当街抢劫，是否也算theft?

标 题: IKEA damaged 家电after delivery求助

上个月在IKEA买了一些厨房电器，当时IKEA送达时外包装都良好；因为contractor的时
间关系到昨天才拆箱子，发现洗碗机的正面角上有很大一块dent，可能是装箱之前就有
问题。当时Delivery时因为外包装都是好的就没有多拆开检查，请问有没有人有这种经
验该如何联系他们退换？查了IKEA的网站，说是delivery 48小时内要和他们联系，另外
电器的warranty也不包含delivery damage.是不是只能找信用卡公司dispute了？ 多谢！ |

保险。
最近小儿把它给摔了，屏幕有了一长道裂纹，倒是还能用。根本就没问苹果，知道人家
不会管的。给CapitalOne打了个电话，给转接到了Visa Signature的客服，结果被告
知，它家的Accident Damage，只管雷击啥的，放车里被人偷了都不管。
看来Visa Signature这卡的这项服务基本上没什么用！ |

基本做法是blood cell先用MMS （Methyl methanesulfonate）treat大概2hrs，用PBS
洗干净，重新用新鲜的medium培养被损害过的cell让其进行repair，2hrs，4hrs,
24hrs
然后qPCR来扩增我感兴趣的genome region
我的预期是，MMS引起的dna damage比如double-strand break会在后面的2hrs, 4hrs,
24hrs自行修复
但qPCR出来的实验结果是：Ct value随着2hr, 4hr, 24hrs越来越大，也就是说DNA 含
量越来越小，和我的预期正好相反
不知道有没有做DNA repair的高手解释下这是咋回事？

发帖数: 274 | 14 MMS之后用q-PCR直接检测基因组DNA在DNA repair领域可以说闻所未闻，没人知道PCR出来
的是什么,因为MMS诱导的damage类型很复杂，DSB只是可能性之一。取决与你想研究的
repair
pathway, MMS不一定是最好的agent.
对于MMS sensitivity, 比较经典的assay有survival curve, pulse field
electrophoresis，但这
些都不能说明特定区域的damage repair.

领域的人用的assay是完全不同的，简直不能互相理解。一般来说，MMS damage repair
更多是用来test BER/NER pathway/factors，研究这些pathway很难用specific
endogenous locus. 对于DSB repair来说,MMS更多只是yeast领域里面用来suggest
epistatic relationship.
从你的描述，感觉你倾向于认为你的locus of interest可能undergo DSB upon
genotoxin stress. 检测最简单的方法就是genomic DNA southern blot for
rearrangements，或者gammaH2AX ChIP，PCR-based assay比如sequencing也可行if
PCR is possible. 当然前提是break frequency要比较高,不然什么assay都白搭。一些

我是ChIP data发现这个repeats：
有H3K56ac的enrichment，同时也是BrdU enrichment的地方
BrdU意味着active DNA replication？而H3k56ac也是replication和damage response
相关
所以读了一堆文献，感觉经常看到的就是MMS可以induce DSB所以就先用MMS来下手（说
错了请指正）。敢问内行们，如果你们看到BrdU和H3K56ac这样的marker，会接着怎么
做呢

I decrease the modulus of the element to a very small value when the damage
occur at that element. For instance, when matrix fails, I take E2=0, G23=0.
If fiber breaks, E1=0, G12=0, G13=0....
As I know, it seems your interest is on the micro scale analysis of the
composite (not macro :-) ), Crack analysis of composite is very complicated.
Take cross-ply composite for example, the crack may happens in the

标 题: 前几天租车没买租车公司的damage waiver

月初去了一趟boston，在Thrifty租了一辆经济型小车，加满油，两天一共100刀，如果
买damage waiver的话要再交70刀。因为觉得自己用discover卡是带租车保险的，就没
买，前台一个劲威胁我要是不买的话出了事你自己负责哦，威胁了三次，我坚持不买。
然后开出去的时候还有点心惊胆战。boston的路实在不好开，当天晚上风雨交加的就在
一个昏暗的小路上蹭到了马路牙子，右前轮的塑料轮毂给刮花了。
交车的时候他们也没检查，打了收据就good to go了。。。。
过去两周了，discover 的网站也没显示有什么异常
应该没事了吧。
后来听师兄说他一般也不买租车公司的保险，因为一般信用卡都有带rental car
insurance。
