发帖数: 932 | 6 1. 首先，sub-lethal irradiation意味着低剂量的照射，并且没有完全的去除
Hematopoietic stem cells，照射完了以后，老鼠还能够存活,所以才称为sub－lethal。
2. 对于低剂量的照射，还会剩下那些细胞和你的老鼠strain还有照射的dosage关系很
大。Generally，最容易去掉的是B细胞和Naive T cells，还有neutrophil。Memory T
cells, dendritic cells,Macrophage， especially tissue resident DCs and
macrophages are relatively resistant to irradiation.
3. 对于这种低剂量的照射，in addition to hematopoeitic system,它对于
intestinal integrity损伤也很严重。所以，经常的，大家做BM chimera要小剂量多次

我上周四按照板上的建议，把CFSE浓度降低到0.5uM,10^7cell/ml室温label 8分钟。
昨天取popliteal LN FACS了一下，所有老鼠的都有很多的OTI细胞，也有很好的
proliferation，在wt/ht/mt里边proliferation的区别也看到了。
估计是我之前用的CFSE浓度太高了，T细胞CFSE label以后都死光了。
真心感谢版上大家的帮忙！
需要比较WT和MT的dendritic cell antigen presentation的差异，因为MT的老鼠本书T
/B细胞和wt有差异,所以把老鼠breed 到RAG deficient background里。
实验是这样设计的，把OTI/OTII （2-5×10^6 cell/mouse） 细胞通过静脉注射打进老
鼠体内，一天以后footpad注射OVA+LPS. 再过72小时harvest popliteal lymph node，
看OTI/OTII增殖情况。因为是RAG deficient 老鼠，所以做tail vein injection前没

或者anti-inflamamtory的分子，喂给老鼠有很强的抑制乳腺癌肿瘤生长的作用。然后
我们发现其实这个分子在肿瘤生长早期，在肿瘤部位非常强的促进了dendritic cells
和M1 macrophages,抑制了M2 macrophages,别的细胞比如T cells没有变化。我们现在
的想法就是这个分子刺激了很强的anti-cancer immune response,所以造成了后面的抗
癌效果。
想问一下，如果后面继续，想研究一下这个分子是如何刺激这个免疫反应的，该如何动

的cytokine expression区别。之前的数据表明，mutant GM-CSF DC有增强的migration
，和antigen presentation。我还想比较一下cytokine secretion。
目前有BD的mouse inflammation kit，还有life technologies的luminex kit (10 and

neuron有axon and dendrite.
完了完了,第一年修的histology, neuroanatomy都忘掉了.....俺好几年不做动物实验
了,现在开始做人身上的实验了,要是半年后再出不了什么好结果,我就不干research了.
哦,那个"1"肯定是neuron,毕竟俺看了几年的猴脑切片,我以前的几位导师中有一位是做
电极插在活猴脑里的,还要事先训练猴子,猴子可聪明了,还会从我手心里拿东西吃.
你为什么要从化学转到生物上来啊? 化学不是不太难找工作的吗? |

个人觉得，
发育最基本的问题，从形态建成，器官大小，器官再生都不理解。甚至哺乳动物的干细
胞数目如何确定维持都所知甚少。
衰老，aging，除了insulin我认为是真的。其他太多都是骗子。我知道某女在线虫，果
蝇里的aging工作别人完全不能重复，现在她又去做老鼠了。衰老有什么机理吗？
疾病，帕金森这么多年了，明白机理吗？
进化我们除了自然选择这个理论外，真的懂机理吗？我最近看Howard Chang lab的文章
。想到RNA，蛋白质，DNA的复杂互作，难以想象这些是如何进化出来的。如果说到神经
系统的复杂性，不同人的某个特定神经细胞的dendrites长出来的形态竟然完全一样，
这种复杂度进化是如何产生的？
难道说这些就是你说的“中间尺度”？那么宏观尺度，个体群体物种水平，发育的机理
是什么？衰老的机理是什么？ |
