我打算用Circular dichroism来研究DNA G-quadruplexes
我看paper很多人都是对比如（ATATAT)n 这样的probe进行研究
我现在是想对一段900bp的DNA进行研究，用CD Spectra可行吗？
另外不知道这个CD难不难做？有没有啥公司或者core帮着做？

the secondary structure of a protein. I suggest in your case, try gel
filtration chromatography first. Or native gel. These are probably the
easiest to do. |

(1).Fluorescence Emission,最简单的看看tyrosine, tryptophan的化学环境有没有因
为Mg结合而变化,从而对荧光有影响.
(2).同时可以尝试Circular Dichroism,最简单的蛋白二级结构,如果不追究变化的源头
,而只是用变化作为信号去测量金属离子和蛋白作用的,很简单方便.
以上两个方法典型的应用有很多,比如calmodulin和钙离子作用.
注意:因为你需要测的是chemical equilibrium,如果反应很慢的话,需要保证滴定过程
每个滴定点(浓度)都要达到化学反应平衡.
SPR
这个不适合,首先是因为信号来自bulk solution(流动的溶液)中的分子被吸附到固定在
表面的蛋白上形成复合物,本质上因为质量吸附造成sensor surface附近媒介的折射率
改变.所以被吸附的质量大小直接影响了信号强弱.你的实验是Mg在bulk solution,这么
小的离子,几乎不可能产生什么明显的信号.这也是ligand-protein binding里很少用
