一年前刚看到这方面的paper就很惊艳,不过被做device的labmate打击了,说是虽然
看上去很美但操作难度极高。之后同是这个哥们时不时跟我说IVC的进展——sorting
越来越方便,droplet寿命越来越长。。。说不定很快我dream up的一些应用就能有人
做出来。当然不会是我——对laymen来说还是太难了:(

兄弟我是个粗人,干发酵的。不知道这么多花花儿道道儿。瞎说说~
很多微生物或油料作物,会积累油脂形成lipid body,细胞膜表面上的lipid只是很少
一部分。
这些lipid body上面结合了不少蛋白。lipid body主要是甘油三酯(TAGs)啥的。不知
道对你有帮助没?
有兴趣可以翻翻下面的paper,更多的自己翻翻吧。

嘛蛋白能quantify lipid? lipid droplet上那些蛋白perilipin之类的恐怕不行吧,连
我都想的到的东西,不然人家早用了。而且我记得虫子的消化道那一圈可是有挺强的自
发荧光的,至少GFP channel如此。

毕竟是HT screening而不是selection。我小见识浅薄一惊一乍大家别见怪呵呵。

目前大家关注的最多的和TAG有关的基因是DGAT,另外还有PDAT也较相关。此外还有和
lipid droplet形成有关的基因也比较受关注。

Raindance和Bio-Rad的ddPCR的原理是不一样的。
Raindance出来的结果不直观,而且不是绝对定量。
ddPCR定量的原理利用的是泊松分布,在低浓度范围内定量挺准,缺点是定量范围比较
窄。

Two methods:
1) Streak the strain out on YPD plate (or selective plate), seal the plate,
and mail it in a pressure resistant envelope.
2) Grow the strain up in liquid culture (overnight is enough), put one
droplet (50-100ul) onto a small sterile waterman paper, wrap it with sterile
foil. Mail it in a regular envelope.

南京大学的张辰宇教授是国内生命科学领域少有的、有自己独创概念的学者,其主要
的工作都集中在miRNA方面。关于miRNA的介绍很多,这里不做科普。10年的时候,张教
授课题组发表了两篇看起来没有什么关联的论文:
1. Identification and characterization of microRNAs in raw milk during
different periods of lactation, commercial fluid, and powdered milk products.
2. Secreted monocytic miR-150 enhances targeted endothelial cell migration.
这两篇文章都挺有意思,第一篇发在Cell Research上,讲的是从牛奶,不管是哺乳

肝细胞细胞质有很强的自发荧光,主要来自lipid droplets或者vesicles。如果做
imaging的话,大家都有什么trick可以减少自发荧光。
我的感觉是在体外培养时间越长自发荧光也增强。

标 题: 调研了一下清华最近回去的PI

云老师发了个贴说清华目前的找人标准,很多人不相信。特地调研了一下清华生命学院
最近回去的人的情况,如云老师所说,绝大多数都有2cns,如果只有一篇cns,那基本
上还有>2的子刊。这个不仅仅局限于胖老师经常说的结构生物学,而是所有方向都基本
如此。
当然这些新回去的绝大部分都是千青(有些不是的是因为回去那一年千青还没开始)。
网页上的Title有的是教授,有的是研究员,估计当时还没有统一。但校内一律都是副
教授,也就是相当于这边的AP。我还特地打听了一下,国内的TT制度好像没有Tenured
Associate这一档,清华5年考核通过了就是正经的full professor。
有些老师看不上国内的科研,我就仅列出两位回国后的publication,咱就不说颜宁等
这种Early T的个例了。说两个没太有名气的,还没有评上T的,各位老师可以参考一下

我也是做结晶的。 你这种情况多半是溶液不对。所谓高盐是相对TEV酶切的。比如
500mM盐。 你得先保证你的蛋白稳定了才能谈其他的对不对。楼上说的加DTT估计也有
帮助。0.5mM TCEP还是弱了点。
再说了,谁说高一点的盐不能结晶的?最普通的做法就是,screening时,set up好
droplet后,密封前,在所有的well solution里补加等浓度的盐,比如500mM.

I have a series contact angle measurement images (jpeg file), which contact
angle and droplet position changes a little from image to image. To avoid
error when analyze image by hand, is there any software (Image J or ...)
that can analyze the image automatically after I define the contact angle

I never heard of negative diffusion coefficient...//expect more details.

本人本身不是做高分子聚合的,只是在一个particle stablized emulsion体系里面用
了styrene作聚合单体。发现增加里面的particle浓度导致聚合速率降低,该particle
是7% 的SWNT负载在SiO2上。增加particle浓度从表观上是减小了聚合物颗粒的大小(~
um)。我看见不少emulsion polymerization都是增加particle浓度增加了反应速率,因
为emulsion droplet变小的话会导致反应体系compartamentalization降低bimolecular
termination的可能。我现在怀疑是不是该颗粒会quench聚合的radical,本人用AIBN
作为引发剂。该颗粒具有SWNT, COOH, Co, Mo和SiO2在表面上。本人提出另外一个可能
是emulsion drop变小,减少了monomer bimolecular reaction的速率。 那种更具有可
能性。先谢谢了。

那这个就是pikering emulsion。我猜,假设所有initiator都在core里面,如果增加
particle浓度,droplet变小,但是数量会增多,总体来说core里面styrene对于
initiator的相对浓度降低,所以速度降低,这个反应用的多少度?AIBN的t1/2不一样
吧。

I am mostly interested in the dynamics of the droplet. So I will solve the
Navier-Stokes equation coupled with electrohydrodynamic force. Yes. I think
the electric field is static and is solved by Laplace equation. I did CFD
before. This research may require me couple the electric field effect into the NS momentum equation.

液滴燃烧显然是所有液体燃料燃烧的基础,这个领域在二三十年前很火,Law,
Sirignano等大牛做过系统的研究,超临界微重力环境下的也都做过。这个领域的研究
为液体燃烧提供了理论基础。
但液滴燃烧和喷雾理论结合的不是很好,显然喷雾是实际燃烧系统中肯定遇到的问题。
如果能把两者好好结合起来,用详细准确的液滴燃烧模型去优化喷雾燃烧模型,显然是
条出路。
NASA的项目,估计可能是微重力相关,或者推进系统。。。
这个领域出可以去各大内燃机,燃气轮机公司
留可以在学术界做做燃烧模拟
其实任何科研方向都是有出路的,只要自己不把路走死就好了。。。

呵呵,燃烧领域现在funding很少。而且作为international student没有绿卡很难找到
相关工作。即便找到,也不推荐。因为大部分项目都是和发动机,或者是NASA相关的。
到时候一不小心被FBI盯上就惨了。
还是看看有没有别的offer吧。

My pleasure =)
Mr. Lith used to be a SP trainer in kaplan years ago, then he started this
workshop. He knows all the details about CS, and he is very helpful too.
Kaplan 5 day should be better, as droplet mm suggests. However, if time and
costs are concerns, this 2-day workshop is a good substitute. Save your "
vacation" days for later interview leave, which will take MANY days.
