是herbal supplements， 但是海关人说不知道里面的ingredient，不能放。其实这些

is about drug design that produced peptide with almost identical activity
as the cell surface's natural conjugates(an import hormone). Sorry, I don't
think it is appropriate to reveal more details about it.. The peptide is
much, much easier to produce than the real hormone.

gamma-secretase 虽然直接负责amyloid的成熟/生成,
但是并不是导致amyloid 变性的原因。抑制了它的功能很难说和Alzheimer's
有什么直接的逻辑关系。
而且这个酶在正常细胞里就存在，说明它肯定是有正常生理功能的。
这么广泛的到处去抑制它，不产生不良后果才怪了。 |

the amyloid plaques are either protective or doesn't really cause AD. First
, transgenic mice with tons of plaques don't have any disease phenotype.
Second, drugs destroy plaques in human don't do anything to improve the
outcome for patients. Third, scientists have been looking only at one side
of the amyloid, why happen to the cytosol part that's also cleaved off.

影响也非常清楚，所以这个酶和AD显然强烈相关，针对这个酶动手，就致病机理上是最
自然的事情，所以这不是个随便乱蒙的例子。现在没多少drug target比r-secretase更
清楚。也许你的总的论点是有道理的，但具体到这个r-secretae上，我想你举了错误的
例子。
唯一的问题是r-secretae是最重要的Notch pathway上必须酶之一。这个基因一旦彻
底敲除（把卵子mRNA也去掉），基本上什么都发育不出来，流产都免了。在果蝇胚胎发
育时大部分细胞都会变成神经细胞，是CPU级别的pathway。Notch pathway在成人中的
功能不完全清楚，但已经知道的部分也相当重要，比如免疫系统的维持。所以针对r-
secretae的毒性估计就从此而来。

在384 plate上,加上药物,3天后收细胞,提RNA,转cDNA,再做qPCR. 但是现在发现收细胞
的时候,仪器在remove medium的时候吸走了很多的细胞,使每个孔细胞数目很不一致,这
样做出来的qPCR variation太大. 目前我用V bottom的plate养细胞,不知道大家有这方
面的经验没有,怎样才能避免第一步remove medium时尽量不影响细胞的数目? 有没有可
能不remove medium,就直接在培养基里直接加lysis buffer,拿到RNA呢? 多谢了 :) |

还听说过EGFR epidermal growth factor receptor，这个领域的药是传统意义的
targeted medicine吧？不是immune therapy吧？
cell therapy好像也挺火，有的公司把病人免疫细胞取出来繁殖后再注射回去，按理说
这也是immune therapy一种吧？
cancer drug以后的发展方向是什么？个人拙见是human genome sequencing廉价后使
personalized medicine成为可能。
