发帖数: 589 | 12 最近几天用Pierce的Seize X Protein A kit做IP，要沉淀的是一个膜受体。用的抗体
（兔IgG）应该没有问题，因为做WB和flow cytometry都成功。使用了DSS让抗体和
Protein A交联。用了300～400 ug total protein沉淀，4度翻转过夜。之后清洗用的
是自己照着kit说明配的PBS,用kit自带酸性elution buffer洗脱，收集了3个组分。可
是之后用SDS-PAGE+WB检测，受体的带完全没有。我知道这个受体肯定是表达的。请问
可能原因有哪些？
另外Seize X的kit刚好用完，有没有什么其他的kit便宜质量又不错的？曾经考虑过
dynabeads，但是貌似还得另买一台磁力沉淀器？

可能出的状况
1）不理解为什么要用DSS？交联后抗体是否还有活性需要检测。
2）elution buffer是否能洗脱目标蛋白？建议直接加上样buffer。
3）细胞裂解条件是否合适，IP时的buffer条件是否合适？做IP的细胞裂解条件是否和
之前WB检测时的一致。膜蛋白可能需要比较强烈的buffer条件。但同时，IP的buffer条
件可能不能太极端，需要摸条件。 |

如果你那个蛋白的size离heavy chain light chain不近，
你先试试不用cross-link做IP好了……
你elution volume是多少？都跑胶了吗？
如果volume大可以concentrate一下，全跑
这个KIT你可以自己order各个试剂，elution buffer也可以找到recipe啊
另外，顶楼上，膜蛋白不好做，搜一搜别人的paper，看用的是什么条件
IP的抗体也要多试几个……
300-400ug有点少啊，多弄点sample啊 |

用DSS是为了让抗体不被洗脱，以后还可以重复用agarose beads。交联这一步我是严格
按照厂家的protocol做的。Elution buffer根据厂家描述是酸性，pH2~3。洗脱应该是
没有问题的。洗脱后没有中和pH,但是厂家说这样没有问题。细胞裂解用的是Cell
Signaling的lysis buffer，用来做相同蛋白的WB从没有问题发生过。
你是建议我用Laemmli Sample Buffer洗脱吗？这样即使洗脱，agarose beads也无法重
复用了吧？ |

用DSS一是不想看到IgG的带，还有就是当初想重复用agarose beads，因为IP需要的抗
体量太大了，想省钱。
洗脱体积是190 ul，IP前的total protein加了350 ug，洗脱后用了36 ul跑胶。
用那么多蛋白做IP实属无奈，我用的是原代细胞，只有那么多量。
请推荐个浓缩洗脱组分的办法吧，可以的话我打算把剩下的样全跑胶了。 |

发帖数: 305 | 17 个人建议。不要光想着省试剂钱，你的时间也很值钱的。不如用最优化的条件做一遍，
得到正结果后再改进来省银子。几点你可能需要注意。
１）SC的抗体很多不靠谱。当然，也有好的。
２）最好不要用DSS。蛋白交联情况和处理时间有关，结果无法用其他实验验证。如果
是这一步的问题，你都无从查找。
３）低PH洗脱液可以从Agarose上洗脱抗体，但是否能保证打开蛋白－抗体的结合未知
，与多种因素相关。
４）你IP时的缓冲体系是什么，不会是１XCS的裂解液吧。
５）你蛋白上样太少了。直接加小体积上样Buffer煮，全上了做WB。
６）膜蛋白IP不好做。 |

定能IP了，再考虑CROSS LINK？
有关重复用agarose beads的问题
1 IP需要多少抗体？没觉得省很多啊……既然用的是原代细胞，你的DISSECTION和
CULTURE的人工费用应该比抗体贵很多吧
2 要确保经过 1和DSS反应 2被ELUTION buffer摧残 后的抗体仍然好用……
浓缩洗脱组分（随便换buffer或者desalt）可以用spin column，

+ CoIP的kit，用的是disuccinmidyl suberate (DSS)，貌似也不贵多少，还省事。有
人用过这个的推荐么？Kit好像也有magnetic和agarose的可以选，是不是一般magnetic
的方便点？ |

个药物载体的构建都不在正文里提了。 Supplementary Methods里面的图1（S-Figure

and the evaluation in vitro）显示SiRNA和带骨靶向多肽的脂质体混一下就能得到有

发帖数: 319 | 23 PA amazon不错的书(补充：就是REVIEW很不错的）本年度以来我已经看过了10本左右基本没看到书，都很烂，我关注的是Class E, AM, collector modulated, 100 watt左右的，基
本没什么书application notes看得也是tens of,讲得很泛
1970年前后的经典paper有些不错，不能直接用，有变化，保护电路讲得很少
有用的知识都在ham里面的实际design，和数字接口的更少，但是有些DSS的design
基本还是没有类似高速数字设计black magic那类书。
