发帖数: 214 | 1 最近做了几次Elispot,目的是检测antibody-secreting cell的数目。用的是Immunlon
plate(通常做ELISA的那种）。结果预期应该有很多spot的孔里连一个spot都没有。但
显色的时候明显变蓝（我用的是TrueBlue substrate），而且孔底洗过后还是蓝的。因
此估计这种plate不适用于Elispot。
请问大家推荐用什么plate做Elispot? |

北京大学医学部免疫学系陈慰峰院士领导的T细胞研究室在肿瘤抗原克隆鉴定及
免疫应答分析、肿瘤疫苗研制等方面取得显著成果。近期，该实验室顺利完成了癌-睾
丸抗原（CT抗原）NY-ESO-1抗原肽（NY-ESO-1b）的临床前研究工作，他们研制的肿瘤
抗原NY-ESO-1b多肽疫苗现已通过中国药品食品监督管理局（CSFDA）药审中心评定，获
得新药临床研究批文。
在国家“863计划”等科学基金支持下，陈慰峰院士实验室建立了肽-MHC分子四
聚体（tetramer）、IFN-γ及GranzymeB/perforin释放的酶联免疫斑点法（ELISPOT）
、胞内细胞因子释放法等国际公认的检测特异性细胞毒性T淋巴细胞（CTL）的实验体系
。借助这些先进方法，该实验室证明了在肝癌患者中，约有1/3病人对NY-ESO-1b抗原肽
有特异CTL应答；通过ELISA方法检测，约有30.8%的患者体内自发产生了针对NY-ESO-1
抗原的体液免疫应答；同时发现在HLA-A2+ NY-ESO-1 mRNA+ 患者存在功能性 |
