最近几年一直在做流行病方面的工作,知道点皮毛.斗胆解释下.
流行病学研究中有Randomized controlled trials和Observational studies,前者使用
随机样本来设计实验,Randomized controlled trials are experiments designed to
determine causal treatment effects in which treatment assignment is
determined through randomization.
后者是根据已有的数据来分析,An observational study is an empirical
investigation in which the objective is to elucidate cause-and-effect
relationships. 不可能象RCT那样随机分配实验组和对照组.
虽然宗教和科学就是不同领域的东西,但是没有科学基础的宗教是脆弱的. |

comparative genomics can tell us. In this paper, the authors discovered ten
new mycobacteriaphages, sequenced their genome and compared their genomes with
each other. The conclusion is revealing.
1) It shows the mycobacteriaphage genomes are highly mosaic, which means in
some part of the genome the sequence is homologous, while in other area, it is

我不很看好时下的Targeted Therapy，迄今的clinical trial结果都不太好(Gleevec是
例外)，针对PI3K，MAPK，EGFR, Ras到B-Raf的各种药，或是毒性太大，或是无疗效。
我自己比较中意angiogenesis和cancer cell metabolism，因此看好VEGF，mTOR，AMPK
，HIF和glycolysis。
我倾向于把癌症看作是metabolic疾病，而不是genetic疾病。我猜测未来的突破也许会
来自elucidation of the metabolic basis of cell transformation and theparies
based on metabolic perturbation。今天来研究代谢，手段方法比三四十年前多了很

另外，我总觉得他们用binreg(svd+probit reg)来预测pathway activity有个问题，就
是他们选的gene signature based on the correlation coeff between phenotype
and the gene exp。如果你仔细看看Bild 2006 的Nature paper，可以看出每个
pathway选出的gene list数目都在变，有时候75，有时候200，300等等，然后再作svd(
效果等同PCA)。这样的feature selection 到底对不对？
就像我们做clustering，只选top highly diff expressed genes做clustering可能有
问题，也可能没事？ |
