target，这些结果基本都是预料中的，做不做大家都信，对field一点帮助都没有，也
就中国embryo不值钱能烧，在美国收这近百个embryo做这实验难多了，费效比太差。
nature science据是正常的，如果他能放卫星说效率能做到90%啦，或者有办法控制off
target不产生mutation等等，看看nature还会不会装ethical的比，肯定是立马收了发

是很清楚。然后在transfer那天，我的针灸师拿了这篇东西给我，我的ivf诊所也没说

挺好的。
时并没发现什么，所以可以肯定是最近才有的。
个小手术，他们说不 remove 也不一定会影响 implantation，因为这个 polyp 很小，
但是如果 embryos 万一正好在有 polyp 那里就不会成功 implatation，即使是他们做
ET 时避免那个位置也不能保证 embryos 不移到那里，他们不想 "take the risk"。
更好。所以我就要好好养好身体为一个月后的 ET作准备。唉，自我安慰一下吧，呵呵。
上还要去 做ultrasound and bloodworkd，另外和 Dr Schoolcraft 谈作 polyp
removal 的事儿。
remove 掉，所以可能要把 embryos freeze，下个月再回来 ET。如果不把 polyp 掉就
移植，会影响implantation。
说有可能只是linning 变厚了，folded, 然后 Dr Schoolcraft review 之后觉得不太

embryo so my re decided to transfer right away. Back then I didn't trust
my re very much as I was very sad about the fact that only one egg got
fertilized. I searched online and could't find many examples of 2 day
transfer. Some articles said that on day2, the embryo is still in tubes.
In fact, the environment in uterus is better than the dishes. Be sure to
ask about the quality of the embryos before transfer. The |

empty sac pregnancies terminated with D&C because the embryo stopped
developing for two weeks while the Hcg was still high. Three months after my
last D&C procedure I got pregnant again. Now I am 4 months pregnant with a
healthy baby. My OB seemed not too worried about my previous history. She
just thought the embryo were not healthy. The advantage of D&C is that you
can get preganent again sooner than a nature miscarrig |

、人等动物在胚胎发育不同阶段的侧面图，其原版收藏在伦敦大学的Bodleian图书馆，
它的作者是十九世纪德国Jena大学的生物学教授海克尔（Ernst Haeckel）。从这张图
得出的结论是，虽然这些动物成年后形态各不相同，但是由于它们是由共同的祖先进化
而来，所以在胚胎发育过程中均有一个形态相似的阶段，这就是“胚胎重演论”（
Ontogeny recaptulates phylogeny）。

这张无敌手绘的作者是恩斯特·海克尔（Ernst Haeckel，1834 - 1919），著名的德国
生物学家、哲学家和艺术家。经他描述并命名的物种成千上万，我们熟悉的很多生物学
词汇，比如“门（phylum）”，“生态学（Ecology）”，“原生动物（protist）”

Female radiation workers who become pregnant and declare the pregnancy in
writing to the RSO shall be limited to a dose of 500 mrem for the
gestational period of the fetus.

一般几十到100micron吧。
用multi-photon直接in vivo看也只能深入到脑子里100－200micron吧。

半疑：如上面有人提出的，动物和人体内有很多组织器官是偏酸性的，为什么没有出现
癌变呢，如果这个STAP cell的理论是正确的话。
半信：STAP cell的totipotency。要知道，目前已有的ESC系，几乎都不具有
totipotency，即只能形成embryo，但不能形成胎盘等extra-embyonic tissue.但小保
方文章的最主要的“卖点”就是这个STAP cell具有totipotency.如果这个STAP cell不
存在的话，那么小保方他们是用什么细胞做了chimeras与四倍体补偿实验？只能是
blastocyst（inner cell mass）之前的细胞，比如2-cell，8-cell或morula embryo。
但是，这样一来，造假的过程就不仅仅是小保方一人可以完成的，必定还有做chimeras
与四倍体补偿实验的实验室。 |

GFP。所以，就这个实验本身，是可信的。可能性只有两个：1）totipotent STAP cell
真的存在过；2）这个chimeras实验从最初都造假了，inject的不是STAP cell,也不是
STAP-derived cells,而是小鼠blastocyst之前的细胞。如果是第二种可能，说明造假
的不是小保方一人，而是至少两个实验室联合造假。那么，问题并非披露的那么简单，
这可能也是为什么RIKEN要重组发育生物学中心的原因。 |

另一个有 off-target嫌疑的技术？没有证据表明，abp1-c1 是否影响到其它target，
尽管回交了一下；然后，他们也提到可能有紧密连锁的其它突变（off-target所致）无
法清掉，所以就筛了另一个（非Cas）突变abp1-TD1。乖乖，也没有表型，但是同样无
法证明abp1-TD1影响了其它基因。反正，两种可能都有：以前的结果被他们认为是off-
target造成，别人也可以说，你们现在的结果也没法排除这个可能性，那发出来干嘛？
等以后第三方再来“纠正”？就为了数文章数？本来，他们吧 abp1-c1 和abp1-TD1杂
交的F1 表型验证一下，对于排除可能的off-target也稍微更有说服力一些（如果这些
影响是隐性的）。
－－验证负结果跟正结果不一样。至少他们证明了基因不表达（我对此存疑），但植物
没有表型。off target导致负负得正的机会有多大呢？他们至少做了abp1-c1跟WT回交
clear background。我赞同你的观点，他们确实应该做几个complement，abp1-c1Xabp1
