如果谷歌一下“Custom Antibody”， 广告铺天盖地，Organic结果更是浩如烟海。选
哪家确实让人头痛。我以前做为客户从其他公司定制过抗体(Genscript, Sigma等)，给
公司打工时也给客户做过不少抗体，现在我所在的公司也在做定制抗体，所以我对现状
还是了解一些的。下面给大家一些我的个人观点。
像马牛羊驴甚至骆驼这类大动物的多抗多用于二抗或给病人紧急用（蛇素狂犬之类的）
，这东西贵得去了，大家用不着。最常用的还是四类：兔多抗，小鼠单抗，大鼠单抗和
兔单抗。兔杂交瘤的专利好象还没过期，所以只能由Epitomics或与之相关的公司做，
其他大家都可以做。技术上，无论单抗还是多抗，实在是没有什么难度，任何一个生物
WSN租间房子养几只动物（不需要无菌，敞开养就行）就可以搞起来。成功的关键主要
有两点：1. 抗原； 2. 多做几只。
抗原好，做抗体很容易，抗原不好，神仙也没用。做免疫的可能都用过Ova，就是鸡蛋

的ab，质量倒是不错，比其他公司的相同ab信号强，特异性好，有一只ab 我reuse了20
次信号依然强的一塌糊涂。不过要命的是shRNA处理后用这个抗体完全看不到knockdown

NIBS-王晓东实验室发现Necrosis的分子机制
在April 10发表的Molecular Cell杂志封面文章中，王晓东实验室描述了MLKL的磷酸化
是如何导致了细胞程序化坏死的分子机制。MLKL的磷酸化使其从单体状态向多聚体状态
的转化。多聚化的MLKL可以结合多种磷脂，从而由细胞质（弥散分布）转移到细胞膜和
细胞器膜上；并在这些膜结构中形成通透性孔道，从而破坏膜的完整性，引起细胞坏死
。这一点有点类似于细胞凋亡中的重要的蛋白Bax和Bak, 在细胞凋亡领域，Bax/Bak在
细胞凋亡过程在线粒体表面聚集，多聚之后形成孔道，从而释放细胞色素C(Cyto C),
诱导细胞凋亡小体，从而激活Caspase3,最后导致细胞凋亡。

剛剛信主的阿翔，每天早上起來總不忘記來到神面前禱告。他每天禱告的內容不外
乎求神保守他，使他平安無事，凡事順利。
過了一段時間，他開始從每天禱告變成三兩天才禱告一次，又過了一段時間，變成
每周主日上教會前才禱告一次，最後變成乾脆就到了教會之後，大家公禱的時候才跟著
