今天有空去查了一下CRISPR专利的历史,UCB的专利已经有claim通过了审查(allow)
,只是还没有issue(时间问题)。我在后面列出来了其中一个通过了的claim,同时把
张锋issue了的专利里面的一个claim也列了出来。大家看能避开不?张锋后来的Cpf1看
着能避开UCB的Cas9。
有意思的是发现最早的一篇CRISPR相关专利申请其实是Luciano A. Marraffini(张锋
合作者)在Northwestern University时申请的,priority date Sep 23, 2008,早了
好几年。看看claim,只有interference,还没进化到切割,可惜一个claim也没批,居
然华丽丽的被一堆112给毙了,也就是审查员觉得那专利写的不清不楚,业内普通人士
看着不可能做出claim的结果,连101,102,103都没用上就挂了。西北也是早早的
abandon了,当年连PCT都没申请,可见并不重视。张锋专利里也没有Marraffini,虽然
science文章有他,我猜可能也是他只做真核interference方面的工作吧,张跟他合作

, 2007; Rogers et al., 2007). The abundant opportunities for prokaryotes to

functional (e.g. the presence of introns in eukaryotic genes, inefficient

比方说,有一个还不错的文章,引用了我的两篇文章十几次,但是大部分都是都是同时
引用了好几篇文章。比方说下面这个例子:

小黄人不吉利啊, 不过细看应该是赢了。
The dispute centred on CRISPR’s usability in two different kinds of
biological lifeforms: prokaryotes and eukaryotes. UC had devised a way to
edit the genetic makeup of prokaryotes using CRISPR in 2014. Broad followed
a year later with a method to use CRISPR in eukaryotes. UC had subsequently
contested the patentability of Broad’s method saying that it was a
derivative of UC’s method and couldn’t be patented separately.

大部分的变异都是近中性的。要移除近中性突变需要非常长的时间,也就是大部分被水
平转入的基因(在一个很大的时间尺度内)都处在“被移除”或者“被固定”过程中,
这个过程是极其缓慢的,这些基因一直没有被移除而处于实际存在状态。
比如金小蜂(Nasonia)体内被转入了细菌(Walbachia)的近完整基因组。

大分子照相相关的炸药奖加上这次的,已经发了14个了(见下表)。
之前主要是基于X衍射或者核磁共振的技术开发和应用,拍一些单分子的裸照。82,91
,02年颁给X衍射和核磁共振的技术开发,剩下11个的给了应用。
而冷冻电镜,这次算是第一个(可能是第二个),直接给了技术。用它可以看更加复杂
、分子量更大的大分子复合体。这些大个头的结构当然更有复杂也更有意思。
这个技术开启了一个新时代,细胞里重要的复合体实在太多。接下来可能拿奖的那就是
:真核细胞的细胞核通道复合体;一一公搞的剪接体(这个结构无论在生命进化和疾病
相关都非常的重要);G蛋白偶联受体(跟N多疾病有关)等等。
彦宁的工作好像确实没有什么希望,葡萄糖转运蛋白跟上面这些不是一个量级的。

Mobile genetic elements
MGEs are non-essential (accessory) genomic elements composed of genes and
structural features that facil- itate their spread both within and between
organisms (Doolittle and Sapienza, 1980; Orgel and Crick, 1980). They are
found in the genomes of all prokaryotes and eukaryotes. MGEs share many
features and often share a common ancestry with viruses. The main difference

We show here that, unlike most other prokaryotic Argonaute (Ago) proteins,
which are
DNA-guided endonucleases, the Natronobacterium gregoryi-derived Ago (NgAgo)
can
function as a DNA-guided endoribonuclease, cleaving RNA, rather than DNA,
in a
targeted manner. The NgAgo protein, in complex with 5’-hydroxylated or 5’-
