What kind of cells, which fatty acids are you using? Please give more
details.
Here is what I used for hepatocyte.
Preparation of the fatty acid stocks
The free fatty acids are prepared by dissolving each individual free fatty
acid in 100% ethanol. Most are readily dissolved in ethanol, though some
require heating to achieve fully solubility.
A final concentration of 100 mM for each of the fatty acids as follows:
31 ul 1 M Palmitic Acid Solid, 1 g / 3.3 ml EtOH

这几种脂肪酸中,Palmitic Acid 和 Stearic Acid常温下是固体,计算摩尔浓度比较
容易。其他几种常温下是液体,但是大体上他们的密度和水的密度很相近,1ml近似1g
。这些脂肪酸都极易溶于乙醇。溶解Palmitic Acid需要略微加热(50C)。
The free fatty acids are toxic unless they are presented bound to purified,
fatty acid free, endotoxin-free serum albumin, which is used at a typical
concentration of 0.1–0.2%.
Take 31ul 1M palmitic acid, 2.8ul 1M Palmitoleic acid etc and bring the
final volume to 1000ul with 100% EtOH, you will get 0.1M FFA. Add 76ul of

我现在做的两个fatty acid isomers的混合物,结构差异很小,一个是一个primary
alcohol,一个是secondary alcohol,有没有什么化学反应可以区分的?
因为需要大量的分离,所以虽然HPLC可以实现,但是还是想找到尽量简单的方法。

不是医生,只能提些建议。最好还是多咨询医生。
1. 保护孩子首先。
2. 从你描述的症状来看。应该是不能代谢长链脂肪酸, 没法beta-oxidation
LCHAD=Long-chain 3-hydroxyacyl-coenzyme A dehydrogenase deficiency
VLCAD=Very long-chain acyl-coenzyme A dehydrogenase deficiency
最后导致长链脂肪酸堆积,导致肝损伤,直至肝硬化。
3.避免长链脂肪酸。这类的疾病用Carnitine or MCT (medium chain triglycerides)
oil supplement.跳过长链脂肪酸代谢那一步(请尊医嘱)
4.我想应该是long chain FA氧化通路上的基因的mutation导致的。

fatty acid合成和分解是两个不同的过程。
身体当中自己合成fatty acid的情况其实不是太多。
一般来讲,fatty acid都是有的。减不减肥的关键是看fatty acid是被降解产生能量,还是被储存
成triglyceride。
越说越生化了。。。

诈一看这文章发的还不错,JNCI影响因子13,14点左右。不过这篇文章的风格是非常
clinical的,很大程度属于resource based的研究(就是最关键的难度不是研究思路,
而是你有那么多的病人资源),所以吗,呵呵……而且我一看就以为一作一定是个MD医
生,没想到居然是个单纯的PhD做的,当然这都是题外话,一篇文章的好坏和发在哪里
其实没有必然联系,和谁做得也不一定有关系,而且这作者也不是日本人…….我们还
是先看文章
这篇文章讲的事情和实验其实都很简单直观,科普的说就是弄了一堆病人和正常人的血

我们的实验是直接拿胰岛做的,体外的
normal glucose, no fatty acid, insulin secretion +
high glucose, no fatty acid, insulin secretion +++
high glucose+high fatty acid, insulin secretion ++++++++ (this is close to 大餐)
normal glucose, high fatty acid, insulin secretion + (this is close to low GI diet)就是说只要血糖不高,摄入fat不太会刺激胰岛素分泌
(加号不是定量的,就是为了说明大概的趋势)
这个还有一个implication就是,如果非要吃high GI的东西,不要跟fat一起吃。既高糖又高脂的东西会大量刺激胰岛素分泌,然后就是大量的fat storage。

1.简单的回答是,会。
复杂的回答是:食物中的TG在肠子里被分解成fatty acid和monoacylglycerol (MG,
就是连
着一个fatty acid的glycerol),这两个东西被肠壁吸收,在mucosal cell里面重新形
成TG,
然后和其他一些lipid和protein pack到一起,形成chylomicron,被release到淋巴系
统里
面,最终进入血液。
chylomicron then bind to lipoprotein lipase (这个酶可以把TG分解成fatty acid

I have been wondering why the natural selection has allow gene that is prone
to Acne to survive. This recent article might give a good understanding. I
did increase my omega 3 intake recently and did notice an improvement. This

linolenic fatty acid is a healthy contributor for your hearts because it is
unsaturated,but it is not stable in a high temperature such as frying.
linolenic fatty acid 在高温下(油炸)会容易降解,和转化为trans-fatty acid,so
linolenic is not good for a frying oil in restaurants and food industries.

下面的论文发表的时间,是中国两种转基因水稻一种转基因玉米“终于”获得安全证书
、并引起全国转基因大辩论的时候:2010年2月。
让我们看一下摘要的简单译文(全文未见):
“用近红外反射法,气相色谱-质谱法、高效液相色谱法、电感耦合等离子体原子发射
光谱法,并以化学计量法研究三种(分别)可抗霉菌病和抗虫的转基因水稻中重要的营
养物质成分组成(按欧共体标准),发现其营养成分组成和物理性状有非预期改变,其
中蛋白质、三种氨基酸、两种脂肪酸、两种维生素及其他数种(微量)元素出现了非预
期的构成变化。差异幅度:氨基酸20-74%,脂肪酸19-38%,维生素25-57%,(微量)元
素20-50%,蛋白质35%;植酸没有显著变化。相比于非转基因水稻,转基因水稻的营养
成分构成和物理性状的非预期改变,可能与基因的转入有关,阐明变化后的作用需要经
过更多研究。”
需要说明的是:这项研究所用的上述几种方法,全都属于常规分析方法,不是专门检测
转基因成分的高新技术,因此这项研究似乎不是针对转基因成分的(网上尚未见全文)
