i read in a fic in which some guys traded limericks with each other to kill
time, the author musta polled them right from a book. those she had
there were rather crass, but i enjoyed them anyway. :) |

发帖数: 321 | 19 想看看蛋白A是否和蛋白B 结合，蛋白A有个myc tag, 蛋白B没有任何tag. 所以IP的时候
就是用anti-myc (9E10)来 pull down蛋白complex，然后用anti-B抗体做WB。
我的negative control 是只表达B蛋白的细胞（没有A蛋白转染表达），这个negative
control 在WB上看不到B，也就是说B和anti-myc抗体或agarose beads 没有 non-speci
fic interaction. 我觉得这个阴性对照是合理的。
可是老板今天非要说这是个 bullshit negative control, you should use mutant pr
oteins (known for no interaction with each other) as negative control.
我听了之后，真TMD要发疯了，这两个蛋白连是否有interaction的确凿证据都没有，我
怎么知道要突变哪里才能导致这A和B没有interaction?

meaning the Heston-Nandi (HN) model or a general SV model? For the HN model,
i do not know. But for SV models, in FIC area, the SABR is heavily used and
in FX, double-heston is used sometime.
Also, which model should be used will heavily depend on your products. e.g.
i do not think you can easily avoid SV models when you price the vol
products like vix options, etc.

1) 有人卖06 Exige的原装座位，shoulder lap的洞本来就有，他在driver侧底部打了一
个，可以装5-point harness了。跟我原装的一样黑色皮座，风格应该比较接近。drive
r侧的$650 shipped，一对的话要$1300，也许还能再讲点下来。
2) 有人卖Tillet的CF座位 http://www.lotustalk.com/forums/f172/new-lotus-speci
fic-carbon-fiber-seats-65200/。driver侧加bracket加运费大概1150。
现在犹豫应该选哪个。Exige的好处：
颜色一致，风格接近，只装一侧应该反差比较小
便宜
安装省事，都不用换rail的
Tillet的好处：
比Exige的舒服
全新
底下本来就有洞，不需要再打，而且这样相信比后打的洞强度要好

看着《美丽心灵的永恒阳光》就怎么也挥之不去的想到《乌云背后的幸福线》，这两部
片子太像了。
1.镜头都不太讲究美感，像是随意拍成的，一部片子里面全是男女主角的镜头，特写，
全景，你，我，你和我。
2.几乎没有什么辅线和hidden plot，没有花边。
3.最重要的一点是人物。两部片子的人物都有点cuckoo，都在逃避什么，对爱情都没有
信心都害怕受伤，但是对对方又割舍不下，一起携手走过一段特殊的历程，在这个历程
中，他们试探对方，压抑自己，释放自己，再次试探对方，一轮又一轮。
这两部片子经常给你忽近忽远的感觉，有时候你和人物有相同的感觉，有时候你作为旁
观者很同情人物的经历，而有时候你又感觉这是生活中的又一出戏。

法。我儿子头段也是迷MTH，老师跟我说他在学校也要看这个书，她就跟他说，我们看点
别的有意思的书吧，MTH你在家看就可以了，我们在学校看点其他如何？学校的non-fic
tion book看得也相对多一些。 |
