gel filtration出来的fraction还是需要别的方法来鉴定。原本是想native page的结
果比较可靠后用来检测gel filtration的fractions的。光从分光度来判断
gel filtration的fraction比较困难。1，蛋白质量少，很难有peak。2，单体蛋白就
550Kda,直接从bed volume就跑出来了。很怀疑跟4倍体能有retention time 上的显著

本就不重要，你只是要确定自己的蛋白是在哪些fraction里面）。gel filtration的每
个fraction根据和standard蛋白位置的比较，是可以知道大概的size的，那不就知道你
的protein的size了吗。你已知monomer是550Kda，到时候不就知道是monomer还是
tetramer了吗
我们的tetramer就是用gel filtration 分离纯化出来的，western 的结果也没问题。
现在怀疑monomer的存在。SDS-PAGE的话，假如原本是tetramer的蛋白也会变性成
monomer跑出来。这样就不知道原来fraction里的究竟是tetramer还是mononer.这就是
我为什么想用native-page的原因。monomer就有550Kda，一般gel filtration用的
standard 蛋白没有那么大的。最理想的结果是tetrmer在void volume,monomer不是。

家用的反渗透净水系统，应该是先用MICRO FILTRATION或者NANO FILTRATION过滤，然
后可能会过一下活性炭，然后过反渗透膜，应该是不会用离子交换的。水烧开以后去除
的是暂时硬度，去不掉永久硬度。 |

martingale可以看成是，在越来越多的知识下，看同一个随机变量，得到的看法。也就
是说这个随机变量的物理没有变，只是看法在变。而看法的变化也要满足物理没有变这
个限制。
举个例子，比如测量恒星表面温度这个函数吧，它是恒星表面的每个点，的一个函数。
这个物理量假定它在那，不变。但是我们的测量手段有限。比如最开始，恒星在我们看
来就是一个点，我们虽然知道它不是一个点，但是没有手段分辨恒星表面是个面，所以
恒星表面温度这个函数，我们只能给它一个数，也就是表面所有点都是一个数。这个数
实际上也就是恒星表面温度的平均数。反过来看，这个数值对应的定义域，是恒星表面
所有的点，这也是这个集合上的唯一一个可测集，literally。
可以想象我们的测量手段越来越发展，也就是能够分辨或测量越来越细致的恒星表面温
度。比如在下一个阶段，我们有能力把恒星表面分辨出四个区域，并测量标注每个区域
的温度，那么恒星表面温度就是四个数，每个区域一个数，或者说每个区域里的所有点

一般要买带HEPA filtration的吸尘器，但是很多厂家HEPA filtration做的很差，选好
点的seal就不会有灰尘吹出来 |

sand filtration --> Granular activated carbon filtration --> ultrafiltration
/ nanofiltration --> UV disinfection. Maybe add something to adjust the
taste. Spring water tastes a little better than purified water because lots
of dissolved ions are reserved in the water without being removed like
purified water, which give you a feeling of "natural taste". |

我最喜欢的动物是狗，所以养了最接近狗的宠物。
金鱼。
是的，金鱼温柔敦厚，见人摇尾巴，从人手里吃食，有的任人抚摸，能活十几年。
形态各异，颜色丰富，憨态可掬。
而且维护简单，就是每周换水，每月换滤片，有时间陪它们就行。
养鱼其实不久，家有三条分别叫 Bumblebee the Common, Ladybug the Fantail, Nasa
the Oranda，为儿子女儿我在另一个空间的投影。斗胆分享下拥有身心健康的金鱼的
十条撇步。
1. fish tank
一寸鱼两加仑缸。除去尾巴，如果一条鱼2寸，三条鱼的话就12加仑，不能再小了。因
为鱼会根据缸的大小控制自己的大小，但内脏会持续成熟，缸太小鱼就撑死了。如果不
想鱼被束缚在狭小的空间里，就象鸡场里的可怜的鸡，鱼缸得是3倍鱼长的宽，2倍鱼长
的厚，1.5倍鱼长的高，那么就是9倍的鱼长的立方。成熟的鱼大概会8寸到12寸，所以
40加仑左右的缸大概是必须的。水缸越大，水质越稳定，越容易保持鱼健康。

第一种贵一些，5 stage filtration,第二种便宜一些，3 stage filtration。
如果主要用来过滤pet allergen (cat dander)哪种更合适？或者两者功效差不多？
还有这个filter大概多久要换一次？ |

你说的确实是主流看法，但楼主是古法养鱼（缺少植物部分），虽然我不赞同，但结果
貌似还行。
最大的疑问是nitrate和排泄物的处理，浓度都非常高，即使水看起来清澈，但其实不
是特别健康。就象帝都雾霾成这样，还是几千万人生活着。
关于鱼缸和filtration的大小，你说的是最小值。一条10寸的鱼，如果只是10加仑的缸
，大概就10*20*10，基本就游不起来，跟养殖场的鸡差不多。所以，最好1寸鱼2加仑缸
。而且filtration要10倍，就是10加仑的缸，要每小时过滤100加仑的水。
关于换水，你说不能频繁，不能超过30%，其实只说对了一半。就是在水质一般的情况
下，换水过多会导致水质变化，从而shock鱼。但如果水质非常好，水里除了少量
nitrate没有别的，那其实只要注意温度变化要小外，换90%都是可以的。一些大拿这么
做，但是一般爱好者还是每周20%比较稳妥。
还有纯鱼缸，就是裸缸，个人觉得也不是最好。金鱼喜欢sift，建议铺一薄层重沙（比
如Tahitian Moon Sand），或者有些algae供其消遣。真的植物我觉得是必须的，鹅卵
石小假山通通不要。 |

running buffer. I usually use 100mM NaCl to run gel filtration, it depends on
the protein.
I don't know what you gonna do to store the protein you purified. I usually
concentrate the protein down to 10mg/ml, and save it for crystallization.
I save the protein in 5% Glycerol, 10mM DTT, 100mM NaCl and 20mM whatever
buffer. So I usually make the buffer, concentrate the protein down to a
reasonablly small volume, add the buffe |

If you have a doubt about this, you should load the eluant directly without
any pulldown to see how much full length protein you recovered.
I don't know why you are bothered by the free amino acid. They will be
washed out in the pulldown process. I suggest you NOT to do the gel
filtration at all.
What you need to do is this:
1.centrifuge to pellet the ribosome. Are you sure you are using 100,000 rpm?

They are inverted, that means the total solution is put in the bottom of the
unit, and the filtrate will flow up to the reception unit that is inserted

dimer。如果gel filtration能同时得到dimer和monomer，分别测定它们的活性啊。

If I understand the question correctly, we need to prove that w(t) is B.M.

Since the process is a Gaussian process, we only need to check the
independent incremental property and the std of w(t_2)-w(t_1), using the 2
part integration you mentioned. |

Yes, we can not use the filtration generated by b(t), but I think
in this case there is no problem to enlarge the filtration.
Basically, if Let the first part be X(t) and the second part be Y(t),
then from the definition, we could see that we can actually add
their QVs together which gives out t. |
