NFAT被去磷酸化后，从cytosol入核。所以打算提取nuclear fraction检验NFAT 蛋白水
平。
我用了pierce 的kit， 不好用， 结果不make sense。
自己试了几个protocol， 背景太高，结果也不理想。
请教做过类似实验的xdjm 推荐个protocol，或者kit也成，能提取nuclear fraction

发帖数: 305 | 8 H3在cyto里检测到是有可能的，但如果cyto里多，就不太对了。H3应该和DNA结合很牢
，其实并不是一个很好的Nu. marker，因为可能会造成分离假象 --- cyto fraction被
Nu protein轻微污染，但无法检测到H3。
不知道LZ如何用镜检确定的细胞fractionation。cyto里面有H3，首要怀疑的还是裂解
过程出错，buffer或者操作。
不急，慢慢来。 |

abundance of protein bands with the stain of ponceau S. However, after i
fractionated the sample, it seems nothing can be detected, even in the

Lysis buffer 绝对是对的，我用了好几种，而且都在显微镜下观察了。老是在
cytoplasmic fraction 一大堆的H3，太让我伤心了~ |

俺还是觉得问题出在细胞裂解，不知道你镜下观测细胞核用的是什么染色。如果是直接
bind到DNA上的如DAPI之类，只要DNA不被降解，都可以染出来，无法提示你关于
fractionation的信息。
如果你还是相信你的操作，做个H３的IF看看。也许是重大发现？ |

mM KCl, 1.5 mM MgCl2）溶胀细胞后，dounce 10次或者加1%NP40，每次我都用普通的
相差显微镜检查过细胞，溶胀后细胞膜没破，但Dounce或者加入NP40后，90%以上细胞
膜破掉，细胞核似乎还是完整的。可是当做WB时候，很多只在核内的蛋白（比方说转录
因子）都可以明显的在cyto的fraction里边检测到，我估计还是有渗漏。你能检测到H3
可能也是这个原因？ |
