multiple enzyme digest,光用trypsin是不够的，还要Glu-c, Asp-N啥的。然后基本能

coupled with orbitrap 或者qtof. FT-ICR应该不行，扫描速度太慢。

charge state(CS>=+3)。不知道你的peptide大都是多少CS。对于tryptic peptides，
基本都是+2-+3，所以大多CID比ETD效果好。而现在越来越多地用到其他酶解的peptide
比如Lys-C, Glu-C等产生long peptide，那ETD的优势就很明显了。
Fusion用的软件比前几代仪器在data-dependent MS2的设计上有很多优化。你可以针对
不同的CS设定适合各自的CID或ETD（包括ETD的reaction time)。比如+2只用CID，+3和
+4的用both CID and ETD。高于+4的只用ETD。ETD对+3CS可以用60ms，+4就是60 * (3/
4)2= 34ms。其他对于>+3 CS的ETD time以此类推。
最后，不知道你对于MS2是用IT检测（低resolution)还是Orbitrap FT检测（high

DMF可以充当由羧酸和硫酰氯或者磷(V)酰氯生成羧酸酰氯的催化剂。
萃取除大量DMF的关键是（a）反应停止后，把有机相倒入或者滴入水相（会放热），同
时剧烈搅拌。水相体积至少是有机相体积的5-8倍以上（推荐10-20倍）。这个水相应该
保持酸性或者弱酸性（pH 3-5），比如加入1/4-1/2体积的蒸馏水的饱和氯化铵水溶液
（加入额外的水防止氯化铵析出），或者酸性的Na2HPO4-NaH2PO4的高浓度缓冲溶液。
（b）然后用低极性的有机溶剂萃取，比如乙醚，二氯甲烷，TBME等。乙酸乙酯也可以
。（c）合并后的有机相用饱和氯化铵水溶液洗1-2次。（d）后继操作视产物性质而定。
即使这样萃取了，还是有少量DMF会混到粗产品中，但绝对是少量的DMF。走柱子或者重
结晶是我常用的后续手段。

I strongly suggest using one large, wide gap hook instead of two unless you
are using a jig that is more than 13” long, then and only then should you
use a top and bottom hook.
Two hooks does nothing but scar up your fish and cause injury to yourself
and foul each other causing missed strikes
It is very easy to make your own assist hooks
As a rule of thumb I keep all my assist hooks 1/3rd way down from top of jig.
