aardlbx 表达的刚性大概是指 3-10 helix，当VSD感受外界电压变化时，3-10 helix结
构是接近垂直（上下）的运动，alpha-helix则接近旋转（类似于螺旋式的上升下降）
。相对于alpha-helix螺旋，3-10 helix的上下运动显得“刚性”一些。
其实这2个结构在VSD的S4跨膜段中都有，而且可以互相转化。VSD的不同关闭开放状态
就是S4二级结构在 3-10 helix和alpha-helix的转换。3-10 helix仅形成于S4的相邻2
个正电荷氨基酸，空间上位置相对恒定。在通道激活，外界电压在促进S4的运动过程中
，每相邻的2个正电荷氨基酸完成一次3-10 helix和alpha-helix的转换，就是一次不同
的关闭状态，close的C1，C2，C3就这么来的。
kdfox11 引用的pnas是catterall的工作，他老人家是3-10 helix的死忠，这篇文章俺
碰巧看过，支持钠通道的VSD的 3-10 helix（所谓偏刚性结构）。

先说题外话啊，哈哈。说到电生理，其实就是研究生物体如何把电信号转化为生理反应
。而电压依赖性离子通道（VGIC）是这个领域的研究热点之一，这个热点的核心又是
VGIC通道如何通过电压敏感区域（VSD）感受电压，VSD感受电压后又如何调控通道开放
？一句话，当外面来了电压，VSD在细胞膜间怎么运动？
在钾通道VSD结构没解析出来，有几种模型被提出来。
代表性的有2种：一种就是细胞膜相对静止，VSD的S4直上直下，即helical screw。另
一种VSD的S4相对静止，细胞膜直上直下，叫moving orifice。
这时期代表人物很多，我只记得Isacoff和Horn。Isacoff要提一下，一方面他是Lily
Jan的学生，另一方面他同时也是最早研制出TIRF的前沿人物之一。犹太人，很聪明。
如今研究VSD这一块实验室虽然很多，但基本上就出自几个大家族，诸如Miller C,

第一个模块的实验结束以后，我们进入第二个模块的实验。这个实验的指导教授叫Al Courey。
这个模块的实验主要是要从Hela细胞里纯化AP-1。AP-1是一种很重要的sequence specific转录因
子，在很多生理过程里都有作用。AP-1其实不是一种均一的蛋白质，实际上一种二聚体结构，要么
是Jun-Jun 或者是Jun-Fos dimer。Jun和Fos都是alpha helix的结构。Helix的一边有很多Leu,和
另一个helix形成Leucine Zipper的结构，而helix另一边就是碱性的氨基酸居多，所以可以和DNA
结合。这个转录因子看起来就象一双筷子夹着DNA。

3 helixes from each
protomer，但是两个protomer 不是像手掌对手掌那样的完全对称interact, 而是两个
protomer之间交叉对称 ，i have

disrupt the dimer but failed. 因为
protomer不是整个helix在interact, 而是有一定角度的，所以不知道为什么两个
protomer之间的作用力会这么强。我们用
的是glycerol gradient sedimentation analysis 来确定 s value， 我试过用没有
salt 的 gradient, 结果蛋白变成了
tetramer; 在高浓度salt gradient 下，仍然是dimer.

helix还好呀。两指肯定没问题，三指我就用来forward和backwards，有错误率不过也
不算高，四指我基本就很少用了，不过用的时候都还可以。
yoga好像只有二指和三指，没有四指操作。二指没问题，三指好像比helix差点。 |

发帖数: 3668 | 20 我用了5，6年的T61，然后跳到yoga 2 pro，现在又跳回thinkpad helix。
T61->yoga 2 pro，除了键盘稍微适应了两天，其他方面都是巨大提升。到现在，yoga
2 pro买了也有9个月了（现在我老婆在用），没出过问题。还挺满意。
yoga 2 pro->helix，感觉整机重量大了好多。不过又用回了小红点，还是比较舒服。
笔和tablet mode是我跳helix的主要原因。 |

10实在有点丑。
helix还有个问题就是pro 键盘居然不是标配，升级pro键盘要320块，这价格简直是在

发帖数: 21830 | 22 下一代你弄我thinkpad跟现在的明显的区别是yoga系列和x系列合并。下面是我老大致
估计的型号演化：
x系列：
x1c4将是x1c3的电路改进型，外形配置都不会有什么变化，最大的区别是cpu更新，和
fingerprint reader改成2维scanner.
x250的后续型号是x260，这个型号改变比较大，x260将与yoga共用一个chasis,同时
yoga系列的后续型号改为yoga 260.
yoga260你们大家已经见到了，所以x260大致应该和yoga 260基本一致，唯一的区别是
屏幕不能后翻。
x260/yoga260的重量都是3lb，基本延续现有x250的特征，改进是把x250的双电池改为
单电池，yoga系列添加x250的企业级设备：smart card reader, wwan和指纹锁，同样
，这个指纹锁因为微软收购upek,改成2维扫描类似手机上的产品。
T系列：
T系列估计比较稳定，基本跟x1c4一样，是基于现有设备的电路更新。
Helix：
这个是最不确定的产品线，helix1还比较成功，helix2因为用了2种键盘这个傻逼设计

发帖数: 1677 | 23 flexible loop一般是连接两个二级结构的，比如alpha-helix----loop----alpha-
helix,如果直接删掉loop,这两个alpha-helix就都被破坏了，因为没法turn了。所以要
加至少一个aa来提供连接，ala是有侧链的最小的aa,加了一般不会引起任何机构上的变
化。如果一个不行，就要再多加。
基本上的思路就是把那个loop变得最短。 |

Not really. The first theory was triple-helix, probably because of some
misunderstanding about exact structure of the molecules and how they bonded
together.
Watson and Crick were the first to propose and demonstrate a model of DNA
double helix. Although it's almost inevitable that someone else would have
came to that solution within a couple of years, if not a few months. |
