半天都用不了。谁大老远去这个小破城市的downtown 。好比来美国旅游去看China
town 。去cancun 周边玩儿要参加当地的tour,各种tour 太多了 ,每个一去去一天可以
买包午餐的。酒店区的all inclusive 未必就是贵的。很多廉价酒店也all inclusive
。当地吃饭很便宜没必要选酒店all inclusive 的。 |

机票和hotel,还是蛮贵的，all-inclusive好一点的很贵，尤其是随季节价格变化很大
，但在expedia上面酒店随季节变化不大，所以想自己用点数订机票，然后在expedia上
订3天all-inclusive的hotel,然后在costco上订3天非all-inclusive的hotel，求建议

hotel zone那里订all inclusive的。前面的几天在PDC因为都在外面玩，所以看很多人
的意思没有必要inclusive，所以我就想订一个不是的。看看pdc 不是inclusive的大家
有推荐的吗？

Presidenti InterContinental 上午退了房，小朋友们都赶着要去Royal Solaris All
Inclusive。到了，房间自然没ready，存了行李，订接下来两晚的晚餐。然后在海边吃
中饭。天巨闷，人爆多，热闹得我想逃走，逃回InterContinental 去。这次的体会就
是这家洲际可以一直住下去，我一气儿可以在那个沙滩椅上躺几天都不腻。
住进房间去感觉好一些，高层，海景，虽然窗户小小。另外两张double beds之外还有
一个嵌在墙上的twin bed，免去了晚上被小朋友折磨的痛苦。搞不懂为什么在all
inclusive的酒店里两张Queen beds的那么难找，本打算实在不行就把小鬼踢下床去的。
7字竖上的酒店，浪大，沙子也粗一些，适合小朋友在岸边玩水，浪里来浪里去，呵呵
。也算满挑战的，浪头小时，可以跳上去让浪推着跑；浪头太大，大到一人多高，就要
从浪底钻进去，从浪的背后钻出来。某人逞能不肯摘的墨镜，最后送给了大海做礼物。
然后各种吃喝，swim-up bar。Marco Polo 的晚餐倒是非常出彩，牛排烤鱼还有头台和

sale拿个VS2的给我，我摘下眼镜看了一下，说这个inclusion好丑。
sale跟你一样，说我胡说八道，不可能看见。对我嗤之以鼻。
我跟他详细描述了inclusion的位置和形状特征，他拿显微镜看了一会，终于说我说的
描述完全对。
这可能跟我近视有关，摘下眼镜，就相当于正常眼镜加了个放大镜。
应为我能看见inclusion,所以对clarity的要求就高些。但是budget有限，学生的俩月
工资（$2000-$3000）,就买了个0.41 ct 的。但是 IF.
你们3，4千就搞 1 ct的，钻石是降价了吧。

先说说俺为嘛在playa del carmen住。因为在playa del carmen住。这里离各大景区单
程较cancun近30min到一个小时，每天就能节省一两个小时。象俺这种在外面疯玩的人
，能省时间可是很重要的。而且在网上做的功课是，playa del carmen更具备墨西哥风
情，cancun太人工化，象miami.而且cancun就只离金字塔近一点。所以去金字塔就准备
从cancun走。女人岛离cancun较近，但不好玩。就选择了离playa del carmen近的
Cozumel.
再说说俺为嘛在playa del carmen不住all-inclusive. 看了俺的行程，估计就知道了
。整天在外面晃悠，基本回酒店就累趴下了。而且回来还不一定能赶上餐厅的晚饭。在
外面饿了就吃。随意。呵呵！
除了Cozumel的潜水需要在岛上单独定，Dos Ojos的洞穴潜水怎么着也得到当地订。所
以就Dos Ojos和Xcaret & Xplore就一起在代理那儿订了。这样能便宜据说一人小一百,

当正儿八经的蜜月了。 媳妇儿做了所有的research， 定了机票旅馆， 我就来记个流
水账。
传说Mexico警察很垃圾，专门找自驾游客的喳。 我们一个很好的朋友去Cancun，租车
，还被警察弄进jail关了2天， 所以我们这次的定位就是选resort， 然后跟tour， 不
自己开车。 网上很多all inclusive的package， 但是最后我们定了一个5星Fiesta
Americana Coral Beach， 机票加3晚住宿每人700。 最后两天我们单独定了一个4星的

四月六号早上，我们开始了第一模块的实验，Burgess教授负责。这老兄研究了一辈子E.Coli的RNA polymerase。E.Coli RNA polymerase 核心酶是一个蛋白质复合体，只有合成RNA的活性，却没有
识别DNA的能力。Sigma系列因子能够识别启动子, 并且与核心酶结合，从而实现转录。我们这个实验
的核心主角就是其中一个因子，叫sigma32。Sigma32可以识别heat shock genes的启动子。Sigma 32
过表达在大肠杆菌里面的时候，绝大部分形成了不溶的包涵体(inclusion body),我们的任务就是要用变性
剂溶解变性不溶的蛋白，然后做重折叠。
所有的buffer几乎都已经配好了，所以直接做就行了。我们先用1% Triton 来溶解细菌本身的一些蛋白，
inclusion body 相当稳定，不溶于triton,所以这一步，绝大部分垃圾就被去掉了。

其实我这个题目不是很准确，我们去的严格讲不是cancun，而是playa del carmen,在
cancun南边一点的hotel zone.
先说说我们hotel的选择吧。我们之所以没有选择住在cancun一是因为不太喜欢city的
环境，另外离city远一点的地方离drug dealers也就远一点，更安全些。playa del
carmen 15年前还是个小渔村，最近几年才发展起来的，当地有一些保护环境的政策，
比如房子都不能建3层以上的。playa del carmen的hotel zone是个gated community，
安全措施做得很好，环境非常nice。出了hotel zone就跟中国的小乡镇差不多。我建议
如果住playa del carmen的话还是选择住hotel zone里面，平时出来走走什么的环境都

院水生生物研究所的谢平研究员在博客上披露，在邮件沟通中，该刊要求作者在包含台
湾的中国地图中用“大中华”，并要求删掉南海九段线。此事随后引起该杂志编辑部关
注，向原作者发邮件对事件做了澄清与说明。Nature Asia网站也发布了正式声明的中

“very difficult” to open in Boston because of its statements on marriage
equality.
Mayor Thomas M Menino suggested he would block the chain from opening in the
city because it “discriminates against a population”.
The chicken food chain says it is founded on Biblical values. Last week, the
firm’s chief operating officer, Dan Cathy, said Chick-fil-A was “guilty
as charged” on its opposition to equal marriage rights.
