ClC-ec1最终能成为“大牌“, 固然归功于Roderick的那篇paper, 却主要也是沾了它的
mammalian亲戚的光. ClC指的是一个氯离子通道家族(Chloride Channels). 在
Mammalian中一共发现了9个ClC蛋白, 几乎个个都有很重要的作用 (见上次的ClC
review.pdf, 我就不一一累述了). 顺便提一下一个有趣的知识, 电鳐 (Torpedo fish)
也有一个ClC蛋白, 叫做ClC-0. 电鳐之所以能够放电, 就是靠它.
很多mammalian ClC和ClC-0的单通道电流都被记录到了, 可是这个电流和ClC的结构有
什么关系却一直不清楚. ClC究竟为什么可以选择性的通过氯离子, 而不会通过钾钠之
类的阳离子, ClC的开放与关闭是如何调控的这些问题一直困扰着很多ion channelists

1. Prepare 4.6ml 15-60% sucrose gradients in 5ml polyallomer tubes (Beckman

我现在读PhD有几个research area的选择:
1)一个是有关处理化石燃料尾气的化学合成(Ionic Liquids)；
2)一个是chemical biology，具体说来就是RNA structure and therapeutics，也涉及
到biophysics(optical tweezer)；
3)还有个是metabolic engineering,说变了就是genetic engineering，molecular
biology之类的（本科做多了，不想干）
我现在纠结于未来找工作前景和自己的兴趣熟练之间，希望化学版前辈们在这两点帮忙
分析分析
我的想法如下：
2和3应该是自己比较了解的领域，2是比较中意也合适的但是会担心不好找工作；
1好像应用性强，但是做的是不太熟悉，导师也会有这个顾虑。 |

buffer. The reason lies in the mechanism of DNA molecule electrophoresis is
so different from SDS-PAGE. DNA band shift is not a unusual result if DNA
sample denature or conformation is not consistent because DNA molecules
migrates on a electrical filed by the negative charge of its own phosphate

No, I did not talk about band distortion. Read my question carefully. Fig 4a
and Fig 4e. No shift when cleavage sites are 30bp and then clear shift when
cleavage sites are closer.
3) Visualization/staining and capture will add non-linear effects to DNA

用non-ionic surfactant, Triton X100. 当分散剂加到一定剂量， 如 2 wt% 以上时
（比如2 wt%, 2.5 wt% 和3.0 wt%），suspension 放很长时间不再sediment. 但不知
如何表征2 wt%, 2.5 wt% 和3.0 wt%之间的差别， 也可能溶液太黑，分辨不出具体差
别。另一问题是，怎样分析上述溶液的溶解度， 也就是分析碳黑在溶液中可分散的最

whiteside的文章没？pH 3-7羧基表面修饰的nanparticle是很容易团聚的，建议pH8以
上。MPA surface pka is ~5.8
第二，你的buffer ionic strength是多少？这个直接影响debye length,这个参数直接
关系到两个particle之间的“安全”距离。

试了一些表面活性剂(SDBS, Pluronic acid, PEG),但是效果不是很理想，过24hr还是

future. My advice:
1 let your supervisor know that you are looking for postdoc position, he/she
can directly settle down a postdoc for you.

engineering departments for postdoc |
