布，如何计算出CCD在不同积分时间下，光谱峰值的count number。比如知道了相机镜
头表面的照度是10000lux,我怎么估算CCD在不同波长接收到了多少个counts?
我需要计算饱和度。如果有了这个count level,外加很容易测的饱和时的counts,就可

我把T=5900K代入L再乘以omega后，得到的曲线总比测量曲线高出一截，而“正确”的
应该是基本重合：
如果我用E=L*omega*cos(66.5deg)，就基本重合了。66.5度是地球自转轴倾斜角。
问题是：在top of atmosphere用卫星测量太阳黑体辐照率，应该和地球自传轴倾角无

我还是没明白二者在根本上有什么区别。一种方法，不用放射性，直接注射BMMC,做动
物损伤模型。 另一种方法，就是我用的方法，用放射性，注射的也都是donor的骨髓（
全骨髓）没有在体外分化的，然后做动物血管损伤模型。既然第一种方法也挺好，为什
么我老板让我用第二种方法，多花6个礼拜的时间等donor bone marrow repopulate

但是sunnyday说的办法问题在于，你不可能把所有分化后的血液细胞都弄死啊，我也不
知道有什么技术可以实现这个（要是照射剂量能把分化细胞都弄死，这个人就已经被弄
死了）。你irradiation老鼠，弄死的还是只是bone marrow干细胞吧，然后你种新的进
去，因为分化细胞的寿命有限，几天之后血液里的分化细胞都是新的干细胞产生的了，
bingo。但是对于AIDS病人来说，这几天时间足够HIV从原来的分化细胞跑到新的干细胞

我用的骨髓是syngeneic，所用的irradiation的dosage是常规的80%，而且是分两次放
射的。另外老鼠骨髓移植后要喝四周的四环素水。另外我用一般的老鼠作recipient一
点问题都没有。所以真不知道是哪里出了问题。

我上周四按照板上的建议，把CFSE浓度降低到0.5uM,10^7cell/ml室温label 8分钟。
昨天取popliteal LN FACS了一下，所有老鼠的都有很多的OTI细胞，也有很好的
proliferation，在wt/ht/mt里边proliferation的区别也看到了。
估计是我之前用的CFSE浓度太高了，T细胞CFSE label以后都死光了。
真心感谢版上大家的帮忙！
需要比较WT和MT的dendritic cell antigen presentation的差异，因为MT的老鼠本书T
/B细胞和wt有差异,所以把老鼠breed 到RAG deficient background里。
实验是这样设计的，把OTI/OTII （2-5×10^6 cell/mouse） 细胞通过静脉注射打进老
鼠体内，一天以后footpad注射OVA+LPS. 再过72小时harvest popliteal lymph node，
看OTI/OTII增殖情况。因为是RAG deficient 老鼠，所以做tail vein injection前没
