sure whether the inserted Cre work functionally or not before blastocyst
injection.
I recalled that i ever electrotransfer pEGFPN3 to ES, indeed i observed
sporadic GFP. So, i decide to use CMV-Loxp-gfp-Loxp to confirm the
construction.
I feel shame on the third point, how can i make this type of mistakes.

是有未知蛋白bind的,只是不知道是什么。据说,大多数GWAS

从geo上下载chip-seq的data files。上传到genome browser里my data下的customer
tracks里。
customer tracks里有对上传文件格式的要求。可以上传多个文件,同时查看。
上传完以后,你就可以搜索并查看你感兴趣的locus的TF binding peaks了。 |

样,理论上说random integration就被筛掉了啊
至于是不是有可能药筛不complete, KO老鼠里面会不会同时也有inregration,我觉得
是有可能的... 并且founder mice PCR一般发现不了random integration
但是这个事情无所谓啊
你就算有random integration,只要不是在target locus的同一条chromosome上面
你反复breed的过程中,random integration就被breed out了
最后还是只有KO target gene是specific |

有H3K56ac的enrichment,同时也是BrdU enrichment的地方
BrdU意味着active DNA replication?而H3k56ac也是replication和damage response
相关
所以读了一堆文献,感觉经常看到的就是MMS可以induce DSB所以就先用MMS来下手(说
错了请指正)。敢问内行们,如果你们看到BrdU和H3K56ac这样的marker,会接着怎么
做呢

我记得我老板跟我说,gene A and B 在同一个chromosome上面的话,比较难cross
一般情况下,大家都会放弃,选择cross A 和 C, 比如 C gene是B的receptor,这样可
以解释同样的问题
如果A,B 在chromosome上面距离比较远,有可能 重组 成功,但是 需要一定的工作量
如果A,B的gene位置比较近,举个例子,P38MAPK的alpha, beta (MAPK 11,12,13,14)
。。。。 他们隔得非常近 位于同一个locus,所以 一直没有很好的triple KO, quart
KO model
所以说,crispr很强大呀! |

p16INK4a locus)。
但是专门做表观遗传的实验室必定不会研究单个基因的调控,如果他们想讲个故事,说
某某修饰或者某某表观遗传的蛋白是重要的,必然需要一个全基因组水平的分析。至于
得出结论之后,再挑几个区域验证下,我觉得不是什么大问题
而且这技术细胞要固定的,只能看某个时间点。而做single cell的最好是能观察活细

起斑竹/板斧的这个马克,还是唐一把吧。
首先,对于数学基础不是很好的同学(有些人一辈子是学工程的,可数学就是不照,
比如说兄弟我),要求所谓的在知识上的完全地“get educated”是不可能的。对于
这类同学,如果将学习的目标定位在“从理论上全面把握”,十有八九是丢了芝麻
也丢了西瓜,除非你有充足的时间,不干别的,专学这一门课。
其次,我所说的并不是完全为了“how to tackle exams”。事实证明,解题方法的
熟练掌握对于理论层面上的理解是有巨大的帮助作用的,虽然这个帮助的方向似乎
和传统上的看法背道而驰。举个简单的例子,LAPLACE TRANSFORM 的理论推导对于
一个初学CONTROL的人来说几乎是不可能的,更不要提CHARACTERISTC EQUATION或ROOT
LOCUS的理论依据了。站在这个处境下,如果以解题处理步骤为重点,熟练掌握对实
际问题的解析,时间长了,理论层面上的东西自然就理解了(例如ZEROS的定义问题
),同时还成功地应付了考试。
