Sean他们让效率提高百万倍，所以可以一个细胞一个细胞研究，他的观点是白血病里面
ms是由CSC的，但是melanoma那些通过普通assay发现的CSC marker都不对，ms每个细胞
都可以变成新瘤 |

improve. Erlotinib's resistance is pretty normal. PLX's resistant is pretty
normal too. And melanoma becomes much more aggressive in resistant patients
after PLX treatment. Basically hospice is the only way to go after that.

其实我也不知道哪种更好,更容易接种. 我们的目的是造肿癌模型后,用化疗药物诱导AP

trial只接受晚期垂死病人。

首先效果并没有楼上说的好。Ipilimumab（anti－CTLA－4） objective response
rate在10%左右（各个地方PhaseIIItrial的效果不一），最好的在MSKCC，也在15%左右
。真正有survival benefit的人群在10%不到，提高的average surivival只有3个月。
考虑到toxicity，这个药目前只在PhaseIII和VI的melanoma上用，就是因为那些病人存
活率不超过5%，等于死马当活马医来治疗。希望能有那么10%不到的人能够中奖且收益
。你说的50%的response，包括了CR，PR，和SD，意义不大。在其他的恶性度比较高的
肿瘤包括NSCLC（肺癌）和prostate cancer Ipilimumab效果不好。其实CTLA－4在老鼠
上效果就只对high immunogenic的肿瘤有效。
anti－PD－1看起来效果比Ipilimumab好，但也只能说稍好，副作用稍小，适用的肿瘤

另一侧flank再打1million BMDCs，7天后flank种1million B16细胞。第一次实验tumor
完全没长。第二次，现在是种了B16后第8天了，打了KO DCs的老鼠tumor很明显，可是
打了WT DCs的还是没有tumor。本来还挺兴奋的，觉得差距挺大，可是WT干脆不长的话
是不是technial problem啊，B16一般几天开始有palpable的tumor呢？不长可能是什么
原因呢? |

的时候也有过不长肿瘤的情况,还很兴奋.最后发现可能是实验细节上的问题.
我是做疫苗的,后来我就直接对那些不长的老鼠再接种一遍B16. 要是保护作用是真的,
那就还是不会长,或者至少要慢的很多. 不了解你的实验目的,或者也可以这样确认肿瘤
确实是不长. |

另一半老鼠身上的肿瘤全都长了。唯一的问题可能出在bone marrow derived DCs上。
可是我就是想不出DC究竟出了什么问题才会让B16完全不长的。不过我打算还是take你
的suggestion，等到14天要是WT还是没动静，就再challenge一次。等到问题解决了来

发帖数: 242 | 40 如果你能好好读一些关于CTLA-4治疗效果的文章，就会知道这个药物和许多化疗药也没
有什么大的不同：第一，毒性大的很，第二，只对melanoma有效，第三，有效10－15％
，第四，。。。。你自己study吧。PD-1和PD-L1那就好太多了（也要你自己study才会
印象深）。这些药物一旦批准，CTLA-4就是垃圾了。James Allison现在就是拼命打压
PD-1/PD-L1，有双重的目的。我的观察，这个Allison是个搞politics的高手，不是他
的credit也敢claim。Lieping Chen和YongJun Liu都太低调了，也是中国人的通病。这
个Lieping Chen，不仅是发现PD-L1，还根据它的功能第一个搞出治疗抗体，anti－PD-
1也是根据这个原理做的。这是个比CTLA-4好太多的药物，得到的credit比Allison可差
多了去了。 |

treatment and placebo groups were given antiPD1 at the end of the trial. But
these trials are really difficult to get enrolled.
As far as I know, BMS antiPD1 NSCLC inidcation has been approved by FDA. Not
