2014年1月《自然—方法学》（Nature Methods）上发表年度特别报道，将“单细胞测
序”（Singled out for sequencing）的应用列为2013年度最重要的方法学进展。
近几年来，基于单细胞测序技术的科学研究取得了突飞猛进的发展，其成果有望为一些
重要的医学问题提供新的解决方案。文章总结了2013年单细胞测序技术对于人类早期发
育、癌症以及神经科学研究等几个重点领域的最新应用成果。文章特别指出，来自中国
北京大学的一些研究团队在这方面完成了许多优秀的工作，做出了突出的贡献。
单细胞基因组扩增新技术（MALBAC）最早由哈佛大学谢晓亮教授发明，相关论文2012年
发表于《科学》 （Science）杂志。该方法通过形成闭合环来抑制DNA片段被重复地复
制，以保持DNA扩增的均匀性，解决了传统方法对单细胞基因组扩增的强烈偏好性的问
题。这项突破在单个细胞水平实现了全基因组93%的高覆盖率，同时也能准确检测单个
肿瘤细胞中的染色体拷贝数异常。

Nature methods 上其实还提到了UCSD的 Kun Zhang lab做的用 microfluidics 实现的
单细胞分离和测序。好像是去年nature biotech的文章。感觉办法很直接很高效的，不
知道成本是否会高。 不过sunney Xie的办法肯定不是唯一的。

线发表了北京大学程和平教授研究组的一篇研究论文，研究人员研制出新一代微型化双
光子荧光显微镜，重量仅2.2克，可佩戴在动物头部，实时记录数十个神经元和上千个
神经突触的动态信号;横向分辨率可达0.65微米，成像质量可与商品化大型台式双光子
荧光显微镜相媲美。
人们观察大脑多通过解剖，大脑处在静止、无生命的状态。程和平教授说，新一代微型
化双光子荧光显微成像改变了观察细胞、亚细胞结构的方式。可在动物觅食、哺乳、打
斗、嬉戏、睡眠等自然行为下，或在学习前、中、后长时间观察神经突触、神经元、神
经网络、远程连接的脑区等多尺度、多层次动态变化。
据介绍，该显微镜成像分辨率高、速度快，并具备多区域随机扫描和每秒1万线的线扫
描能力;采用柔性光纤束进行荧光信号接收，解决了动物活动、行为由于荧光传输光缆
拖拽而受到干扰等难题。其效果远优于目前领域内主导的由美国脑科学计划核心团队所

Also Newton's method could fail to converge if starting too far away from

working in China because the potential application of OR in chinese industry
is tremendous. I certainly will choose OR with no doubt. Many similar programs
also teach and specialize in OR: industrial engineer, operational management,
etc.
For studying quantative method business, you might be facing competition from
every quantative and statistical programs. Useful but not very specific.
