接测量才能有效定量化. Mitotic chromosome究竟是有个rigid protein scaffold,
DNA绕在上面; 还是一个DNA network, protein只是提供网格节点的稳定性? 以前猜东
猜西, 也不如抓住一个chromosome, 测量它的力学响应, 以及MNase对这个力学响应的
理解深刻. 结论是mitotic chromosome是一个DNA network. 而且是一个非常松散的网
络.

neurons and glia usually cannot survive for long term in NBM+B27. With that
said, there is no pure neuronal culture. Anyway, cultured neurons do exhibit
some glial properties, such as glucose transporter expression, metabolic
activity, etc.
For embryonic glial culture, we use DMEM/F12 (1:1) +10% FBS. Because glial
cells are mitotic whereas neurons are post-mitotic, neurons will be easily

淋巴结，呈阴性，从而认为没有癌细胞扩散至盆腔内的淋巴结。现在已看到病理检查报
告：
This uterine smooth muscle tumor shows diffuse moderate to severe cytologic
atypia. There is a hemorrhage and focal necrosis. In addition, the mitotic
rate is as high as 10/10hpf. Based these parameters, it is classified as an
intermediate grade leiomyosarcoma.
下面是我的大致翻译：
这一子宫平滑肌瘤显示从中度到快速扩散非典型细胞。此外，有出血和病灶坏死。细胞

For Q5, if they use CSF extract( a kind of Xenopus extract, which should be
natually mitotic extract, once you add Calcium, a good CSF extract will start
to cycle.), they can avoid the possible artifact. The problem is in their
paper they use all human proteins(separase, securin and cohesin.) If they use
CSF extract, the proteins they can analyze for MS are Xenopus proteins. What I
don't understand is why they use human proteins.

After the addition of calcium or fertilization which causes a transient
increase in cytoplasmic calcium conc., the CSF will be inactivated, following
by APC activation, cyclin B destruction and mitotic exit. And this process is
roughly 30 min. It is easy to collect phosphorylated separase before the
addition of calcium. To collect the unphosphorylated one, like you say, you

的影响？
尤其是RNA浓度低的样品。因为要调成和浓度高的样品一样的总量，所以要加的比较高
的体积。这样在反转录体系中这个低样品的洗液就多，抑制反转录酶的活性很强。所以
你虽然放了一样多的总RNA，但是大多都没有被反转录。当你所qPCR时，你这个样品的
Ct值会很高，意思就是表达很低。所以就是给你表达低的假现象。你做qPCR时，可以留
意一些低浓度样品的Ct值。很多人发表的qPCR的文章后来表明结果相反，有一些原因是
这些研究者没有考虑这个原因。
这是我给另外一个贴子的回复
Qiagen柱子提取的RNA总是含很多的洗液。后面会影响到反转录酶的活性。如果每个样
品的浓度不一样，你做qPCR时，要调成一样的浓度，这样你做反转录的每一个样品的反
应液里含有的洗液浓度就不一样。不一样的洗液浓度，抑制反转录酶的程度也不一样。
这样你最后出来的qPCR结果肯定不准确。有的样品里表达低，可能是因为洗液多而抑制
了反转录酶造成的，而不是真正的表达低。如果你同一组的样品，有的表达高，有的表

发帖数: 144 | 43 你如果用的是Qiagen 或其他公司的柱子提的RNA，那你有没有考虑过RNA样品中的洗液
的影响？
尤其是RNA浓度低的样品。因为要调成和浓度高的样品一样的总量，所以要加的比较高
的体积。这样在反转录体系中这个低样品的洗液就多，抑制反转录酶的活性很强。所以
你虽然放了一样多的总RNA，但是大多都没有被反转录。当你所qPCR时，你这个样品的
Ct值会很高，意思就是表达很低。所以就是给你表达低的假现象。你做qPCR时，可以留
意一些低浓度样品的Ct值。很多人发表的qPCR的文章后来表明结果相反，有一些原因是
这些研究者没有考虑这个原因。
这是我给另外一个贴子的回复
Qiagen柱子提取的RNA总是含很多的洗液。后面会影响到反转录酶的活性。如果每个样
品的浓度不一样，你做qPCR时，要调成一样的浓度，这样你做反转录的每一个样品的反
应液里含有的洗液浓度就不一样。不一样的洗液浓度，抑制反转录酶的程度也不一样。
这样你最后出来的qPCR结果肯定不准确。有的样品里表达低，可能是因为洗液多而抑制
了反转录酶造成的，而不是真正的表达低。如果你同一组的样品，有的表达高，有的表

有时候看到有的人可以一堆一堆地数出来，感觉好厉害啊！
觉得这期science比较有趣，虽然没有那种看得超感动的文章，但是还是有不少很值得
一看的paper的。

但是作者什么的老是不太记得
有时候看到有的人可以一堆一堆地数出来，感觉好厉害啊！
觉得这期science比较有趣，虽然没有那种看得超感动的文章，但是还是有不少很值得
一看的paper的。

人已经说了，哺乳动物细胞是由importin T介导的（STH like that）
另外，我觉得可以做几个thought experiment
1,如果只有M phase plasmid才能趁着核膜解体进核，那酵母里转染基本是mission
impossible了（因为酵母核膜不解体）。
2，如果只有M phase plasmid才能进核，我们转染293T是不可能在24小时后达到80%的
transfection efficiency without doubled cell number.
3,如果M phase plasmid才能进核，是否表达外源质粒早就会被发展成鉴定是否post -
mitotic cell的标准了。
…………我觉得还有很多例子。
最后弱弱地说一下，我之所以想到这些粒子，因为我似乎看到过把质粒转进去再pull
down下来看sequence specific histon deposition的，啊哈哈 |
