发帖数: 5365 | 9 从来没有做过B cell,现在要在B cell line里看一个细胞内蛋白,用一样的抗体,flow
cytometry没问题但做cofocal isotype control 的背景很强.目前为止blocking 效果
最好的就是老鼠血清(用的抗体是monoclonal mouse antibody directly conjugated
with Alexa Fluor),但问题是背景没了,信号也弱了好多.这个实验组和isocontrol 比
起来亮不了多少.现在打算用1抗加2抗做,一般是用和二抗同种的血清来做blocking,但
听之前染过这个蛋白的人说,二抗血清对B cell Fc没什么作用.我的问题如下:
1. 不知道这种情况下能不能先做Fc blocking再用二抗血清block?
2.Caltac Laboratories (now belong to Invitrogen)原来有个抗CD32,但不连接荧光
素的已经不卖了,而且那个也是从老鼠养出来的.请问有什么非老鼠的抗体或血清适合做
human B cell FcR blocking?

这是clinical trial里面很常规的作法，如TKI, 如EGFR monoclonal antibody
TKI是检测EGFR mutation
EGFR antibody是检测EGFR overexpression或有correlation的biomarker
到底能否work取决于你选择的biomarker
这个也是需要做clinical trial才能知道的。 |

Amgen 公司试图开发的单抗药物宣布失败。该药品是抗 type 1 insulin-like growth
factor receptor，在细胞和动物模型中有理想的效果。而且，在动物
和细胞模型里面过表达IGF-1会导致癌细胞行为并能被该抗体抑制。
但是在phase III clinical trial中得到的数据表明该药物对病人身上的
多种癌症都没有疗效。
其他类似的单抗药物目前看来效果也不理想，研究者们认为大部分癌症（尤其是固体瘤
）的机理比较复杂，用单一靶点可能是很难抑制癌症的。而且在初期研究中使用的癌症
模型也不能被视为是真正的癌症例子。
另外几个公司目前在试图把 IGF-1R 和 insulin receptor一起阻断，看看能不能得到
更好的效果。

不需要用S35 （用了也未必有说服力，因为高分子量的smear未必就是ubiquinylated
band，审稿者要挑毛病还是很容易的）
你可以在MG132 处理（以及对照）的情况下直接pulldown IkBalpha, 然后用anti-
ubiquitin直接来看pulldown product 上的ubiquitin signal。至于抗体，推荐你用
Millipore monoclonal anti-ubiquitin。 这个方法就比较无可挑剔了，也不涉及放射
性。 |

I compared and it never works.
2 reasons
1. ChIP buffer contains SDS and SDS kills M2 beads (refer to M2 beads
handbook)
2. you never know how much antibody you used. 10 ul of M2 beads perhaps
contain hundreds ug of ab. and saturate your chromatin. you need titration,
but impossible.
you may want to try M2 antibody which is a monoclonal antibody and the same
as the one conjugated with M2 beads.

想法。
俺曾经做过几年癌症研究，也发过几篇说的过去的cancer课题论文，应该说对癌症还是
有一点点皮毛的了解的。现就自己一点点局限的认识，跟大家交流几句。因为俺已经离
开基础研究领域了，要是谁从这点交流中得到一点灵感，万一得了炸药奖或将来卖个牛
x的抗癌药，千万要pm俺一下，好替你高兴。
单刀直入说吧，一句话概括：俺觉得现在癌症基础研究方面有个很大的缺陷，即对分化
（differentiation)投入的力量过于薄弱，而主要精力仍然盯在proliferation和
apoptosis上。
怀疑是不是很多纯生物专业毕业的同仁不太了解癌症的病理分级分期。在病理上，定性
一个癌凶猛与否的最重要指标就是differention程度，而proliferation和apoptosis的
markers如Ki67，p53等只在很少数的癌症诊断中使用。一般而言，分化越低，
prognosis越差。
人类现在治疗癌症的基本或主要手段是什么？绝大多数都是能切则切，首选手术；然后

Gleevec (Gleevec phase 1 is 100%, haha, 88% at phase II and probably because
