【舍恩事件】
舍恩事件是一起于2002年揭发的大规模学术论文造假丑闻。事件主角扬·舍恩 (Jan
Hendrik Schön) 1970年生于德国费尔登，1997年获康斯坦茨大学物理学博士学位
并留校任教，1998年加入贝尔实验室，于贝尔特拉姆·巴特洛格教授指导下做材料学研
究，2001年成为正式员工。之后的4年里，他与20多人合作在许多大型国际期刊上发表
了超过80篇论文，在《自然》和《科学》上发表的第一作者署名的论文就有17篇。这些
论文在当时被认为于分子电路、分子半导体、有机激光、高温超导和纳米科技领域取得
突破进展，震动了整个科学界。2001年，他获得了德国最重要的科学奖项之一——奥托
·克隆-韦伯银行奖。

假设现在我把球球tip放在一悬空薄膜表面,然后做力-距离曲线,
根据所得的数据,可以得到力的大小和膜的形变距离,
问题是,如何由这些数据计算薄膜的力学性质,如:杨氏模量?
由于本人主要从事炼金(chem)的工作,对材料物理了解甚少,望各位多多

Lieber这种话都说出来了.
Schon 以后不管怎么说都会比较难混了. 其实他做的过分了, 偶第一次看science 12月
的那篇paper的时候就觉得有点不对了, 和nature的比较fig完全一样, 后来加了个
correction说是加一个0.1的factor, 那有这么巧的. 按他的说法, nature的monolayer,
science的是diluted看做single molecule的, 很简单的直觉上也应该知道monolayer更
接近bulk一些. 就算不考虑物理内涵, 实验上普遍观察到的current leakage,
两个结果就不应该是简单的线性关系.
偶现在做理论, 上个礼拜跟一个postdoc讨论molecular transistor的问题, 他science
的那篇paper很难用理论解释, 因为外加小电场对分子轨道的改变微乎其微.

的一些原因：cells condition, monolayer overlapping之类的问题.当然commercial
trypsin function本身就比较复杂，有时有些人就用较纯化或重组的trypsin和胶原酶之
类成份确定的混合物去消化。
我的感觉是你是否让你的宝贝细胞刚长得90%confluence后，hanks washing triple,再用
0.05%trypsin在不同温度（Rm, 36）消化一下看有没有提高。
实在不行就定些Cell Dissociation Buffer or Enzyme Free Dissociation
Reagents(60% confluence)，但我总认为是花拳秀腿，但也许就对你的实验管用。
另外培养界面，polypropylene, glass, teflon-coated 都是option,但我对这些不熟悉
。可能还有几个较流氓的方法，冰水超声,内置tiny cover slides, 瞬时低渗 |

transfection, 不知道翻译准不准）。前一次做的感染细胞放了一星期，monolayer都
要100%了，还没死，怀疑感染效率低。现在试第二次，已经转染后72-96小时了，发现
转染了bacmid的细胞除了长得比不加bacmid的对照长的慢点，好像很难看出来
invitrogen册子上说的细胞变大，出芽（这个我尤其难分辨）什么的（我平时也就用
200X inverted microscope观察），大家有比较明显的感染细胞和不感染细胞的对照图
推荐我瞧瞧么？我不想一直等到细胞dislodge了，或者裂解了才能知道感染成功没有。 |

If you grow adherent cells in monolayers, it's better to perform the
crosslink in petri dishes.
The basic principle is that: you want to take a snapshot of all kinds of
interactions with the minimum disturbance to the cells. |
