plethora of different cells. But the first one we looked at, of a leaf of
all things, absolutely blew my mind. I can't describe how I felt in words,
it was a feeling of looking at the building blocks of life. And when I saw
the organelles shifting around and actually saw that it was alive...not to
sound kinda girlie or anything but I felt like crying. It was so beautiful.

很多organelle localized的protein的turnover时间就是比较长。
即使mRNA knockdown了,蛋白降解还需要很长的时间。

概念。但是印象中PFA penetration的速率好像是1小时1mm左右,你自己可以估计一下
。我想多半还是太大,应该需要slice,并且需要4度overnight。小sample 2hr足够fix
的不必overnight,overfix也不好。但是好像很多人都是丢进fixative过夜。所以应该
区别也不大。但是最好是4度,否则cellular extraction和其他artifacts会比较严重
。sample stress在化学固定过程中或多或少,不可避免。除非高压冷冻迅速fix
sample。在电镜下会看的比较明显。plasma membrane会很wavy,一些organelle形态异
常,甚至aggregate。

modification. Some scientist in the field believe a lot of acetylation
modification is through direct chemical transfer of Acetyl-CoA without the
help of HAT espeically in organelle enviroment favoring nucleophilic
addition.
As far as crotonyl-modification. My friesd published in a good journal
stating HDAC3 is the de-crotonylase...my lab results are exactly opposite.

discussed in the paper. so we cannot distinguish between the cyto and mito
pools of GSH. this will certainly be our next step.
In terms of toxicity, we have not measured IC50 yet. during the 1-2 h
experiment, we did not observe any apparent toxicity. However, this does not
mean it does not have long term toxicity.
The highest probe concentration we used was 1 uM. For HeLa cells, we can use
