周五上午收到perm hardcopy，下午用overnight寄给我签字。
我（东海岸）周六早上九点收到，签字，周六下午 overnight寄出，
周一上午律师收到，同日快递发给同城市公司总部签字
周二上午公司总部用同日发回律师，周二下午四点前律师overnight送出140。
前后五天，包括周六周日。（中间求爷爷告奶奶，说好听话陪笑脸省去XXXX字。） |

几个月等到满180天换工作啊。
律师答应下周尽快给递啦，我们也在争取尽快体检准备材料。律师说如果通过USPS
overnight 的话，因为移民局lock box也是usps的service，他们周六也工作的，所以
周四递交就fedex overnight，周五递交就usps overnight. |

上个星期四收到USCIS的email说有RFE要寄到了。
兴奋的我们赶快请假星期五去办medical然后星期一拿到所有结果。
拿到以后第一件事就是发email给我们公司律师说都办好了medical准备要overnight给
律师。
结果律师说不要着急等收到RFE信再寄给我们。
告诉他排期倒退后他勉强同意我提前寄给他，然后星期一当天我们就收到RFE信件了，
果然是medical和G325A。
律师星期二说收到了把G325A给我准备让我签名，结果到晚上了他们发现他们用的是旧
的(2015年的)。结果耽误一天说星期三给我们新的签然后他们星期四overnight给USCIS。
结果星期三到了一整天律师也不知道在干什么，每次我联系他们他们都说基本办完了不
过每次都没有给我。
到晚上了他们下班了连个email也不给我就走了。
我着急的一直发email给他们问他们到底怎么样了我的G325A呢？结果他们给我来一句今
天来不及了明天给你。。。。。
我气的把HR加公司老板都CC的来了一封狠email结果他们告诉我他们准备加班帮我弄完。

结果工作人员撕了第一页自己留底，复印的第一页给我，把最不清楚的单子粘到信封上
，当时郁闷坏了，字迹模糊，工作人员辩称自己可以看见。说是overnight,结果昨天查
没有update,今天中午才deliver.不知道有没有送对地址，；现在只能等uscis 短信看
有没有收到。所以建议大家还是自己打印label, 别用那个overnight, 压根没
overnight ,不知道什么状况。；吓的以为信丢了，现在还担心呢 |

这简直是晴天霹雳阿。那家里有两辆车的要是找不到有两个parking spots的房子可怎
么办啊？终于明白zipcar是怎么在boston起家的了，zipcar在我以前和现在住的大城市
都没啥市场。我们这虽说parking难找，可也没有overnight parking ban阿。还有
boston地区除了brookline还有那个城市有overnight parking ban？都不想在
brookline找房子了。
还有问问大家green line C和D方便程度是不是差不多去红线的Charles station？ |

现在比较倾向于找一家有注册而且可以overnight 的family care做备份。
在研究过程中看到一些东西跟大家分享：
1. 我们是Kaiser的保险，现在指定医院从St Vincent 换到了新开的Westside Medical
Center，他们自称是加强母婴联系，只设nursery给有健康问题的宝宝，普通宝宝都放
在妈妈房间。另外产房和postpartum room是同一间，护士跟房间ratio 1:3。根据俺上
一次被C的经验，如果这次不能顺产的话，光是妈妈和老二呆在医院可能会有点够呛。
（非常希望VBAC成功。。。）
2. 州政府一般会有管理daycare的办公室可以提供家附近有注册的各类Daycare。
Oregon有这个网站oregonchildcare.org可以查到list，然后挨个联系是否能overnight
。Oregon的注册有三种，除了daycare center以外，有Certified family care 和
registered family care两种，区别看上去主要是允许招生人数和inspection的频率。

上得到很多有用信息，无以回报，随便写上一篇Whitney登山记，希望对后人也能有所

一直和LD商量着爬一把Whitney爽一下，但是苦于没有permit，以为今年是不可能爬了
。经多方查探，终于打听到没有抽到permit的还可以提前两天打电话去预约，如果运气
好的话，有可能能拿到。（这里还要多谢SurfingCat帮忙，提供了很多有用信息！）周
四LD打电话过去一问，正好周六/周日的overnight permit还有3个，急忙预约下了2个
，于是周四、周五两天匆忙购物打包，周六早上10点多就开车驶向Whitney。
1点左右就到了Ranger Station。拿permit的时候被告知，overnight permit必须在周
六当天行进到trail里面去camping，不可以在trailhead camping（其实后来发现，即
便在trailhead camping也不会有太大问题）。于是我们用overnight permit换了周日
的day permit。在这里还领到处理sol |

其实包太大了有很多缺陷，比如包的自重太大，承重系统不好设计等等，
所以对一般人来说，第一个overnight的包，最好大小就是60-65L（男生），
买比较popular的作overnight backpack的牌子，不要贪便宜，

爬switchback时所有的人都在往下走，就我一个人往上，心里有点慌，爬快了就有点胸
闷气短的感觉。后来放慢了点速度才好的。过了trail camp的高度还是有点挑战的。
5点多开始下山，天黑前就我最后一个人在那儿下山。下山时到了trail crest前东西吃
完了，水喝完了，在那里爬那段你说的300ft时走两步歇一回。过了trail crest下来一
团漆黑在那儿数97/99/100-switchback。
day hike有day hike的好处。overnight也有overnight的好处。我上次是在trail camp
overnight，背了很重的东西上去。

也应该满足，如果按字面意思理解楼主这样飞不应该算是back-to-back吧。当然航空公

持。我坚持一段时间煮浓浓的白银耳莲子百合汤的时候，皮肤确实会从里而外的更加丰
润。
白银耳莲子百合：
干百合，加水泡overnight
莲子，去心的，加水泡overnight
白银耳，买贵的，大朵的，泡2-3大整朵，加开水泡overnight
第二天，放材料进高压锅，注意不要把底下的沙子放进去。白银耳本来是大朵，你这时
候用手仔细把白银耳撕成小片，每片/块大概大拇指大小就行。我妈的经验是材料是整
个容量的2/3，上面的水是没过材料多个1/3来。高压锅上汽之后半小时或更多，有时候
真不一定多久，反正就是炖到银耳还是小片状，但是胶质都出来，粥是粘粘的胶水状。
我喜欢放糖，否则没味道。吃的时候再放糖。 |

用walmart货，然后慢慢用到中等品牌。摸索几年后，我现在皮肤应该没怎么老化，还
木出现皱纹，而且紧致度和光泽度也都还可以，我应该是找到了适合自己的方式：
1. 护肤最重要之一防晒： 紫外线一是晒黑，二是最可怕的致老！皱纹都是因为ROS和
紫外线引起的老化～所以，对于所有mm，无论什么年龄层，都应该一年四季，无论下雨
下雪还是阴天，都要在白天擦防晒霜。我脸上一年四季用spf40的，在夏天出门我会擦

本来这个yield就不高,我每次为了测个序,都至少要从10ml overnight culture里小抽
有关不表达的问题,建议你所有clone(一个construct做8~12个),先煮菌,然后挑阳性的
用1ml small scale 测试表达情况,简单做法是
overnight culture,1:100稀释到1ml LB中,37度,3hr,然后用终浓度0.2~1mMIPTG
induce
37度3hr,然后取100ul,离心,扔掉90ul上清,加10ul 2xSDS loading buffer,不用重悬
直接煮10min,跑胶
有的时候这种条件可能表达时间不够看不出来表达,就要用更麻烦的small scale
就是2ml体系,其他都一样,但是要37度养到od 0.3,然后换18度养到od 0.6,再induce
induce后用18度,200rpm,overnight(8~16hr),然后再看whole cell蛋白看有没有表达
如果上面两个都没表达,基本上就没啥希望了 |

然后用FPG这种能识别8-oxoG的酶来切，被切的基因组位点会产生nick，阻碍PCR
polymerase，基于这样的原理做qPCR定量来研究某段序列对于oxidative damage的反
应。
好像这种做法比较少见，大部分DNA damage的研究都是global的。但也有很好的先例比

说肯定能抽干了。你那种指针型的真空度表是不管用的，我们实验室手套箱的真空度表
还能指到低于零呢。你可以去借一个digial vacuum gauge，买一个也可以才200多块钱
。DMF的沸点是153度，你去查一下真空度和boiling point的关系表(很多catalogue的
背面都有），心里有个数，要
大大的低于算出来的这个要求，然后overnight 或者抽几天，别忘了添加液氮哦。别说
沸点是153度的DMF，就是沸点是200多度的东西给与足够的真空度和时间都是能抽干的
。不过要有耐心，我如果用我们手套箱上的油泵，有的时候overnight都抽不干普通溶
剂。一般来说如果5g以下的东西overnight都抽不干（除非你的东西量很多），就是这个方法不work
嗯，我的建议就是上digital vacuum gauge 和液氮冷阱，一定能稳妥地搞定。 |

and then pending data for the US GDP, and after GDP first reaction happend...
all these happend overnight.
if you play single stock, play overnight is better, otherwise, I guess the

but the difference between china overnight rate and that in other country is
that overnight rate in other country is a market behavior but in china is
more like a government behavior.
So it is actually a very strong signal that central bank of China is holding
the money tight and the housing market is the place that we really need to
worry about. |
