不要凝結式, 要會崩成粉末....據說應該就是要這樣, 濕掉的沙會崩碎掉到下層去.
老貓用了十幾年的clay litter從春季開始忽然不肯用了. 紅袋Dr. Elsey也拒用, 只用
Breeze的沸石貓砂(pellets), 但是用五天也會拒用, 按理那是可用一個月的. 太$$$.
所以考慮用松木砂. |

面前的壁炉里的火烧得很旺。整个屋子都充满暖意。
这个壁炉是老式的，燃烧一种处理过的叫做pellet的小木屑。壁炉里有一种装置，匀速
的按照设置的量放出pellet燃烧，将循环空气的导管烧热，风机将加热的与燃烧炉分离
的干净的空气不断送出，屋子里很快就暖和起来。这个壁炉的壁炉台，据说是二战时旧
金山海军总医院的，二战后医院拆除，壁炉整体后来被这个屋子的主人转手买下，安装
在他自建的这栋维多利亚式的房子里。
看着着设计精巧的壁炉，宋加想起小时候在中国，冬天里使用的火盆。那火盆，就是一
个矮木架上，支着一个浅铁盆，上面烧起木材，或者木炭。有时候会在火堆旁暖一壶茶
，或者烤几个红薯。一家人围坐着，欢声笑语。虽然简陋，也算其乐融融。后来宋加从
南方到北京读书，有机会见识了北方人的土炕。冬天里烧热了，捂着被子坐在土炕上，
也是暖融融的。这种土炕，在北方的农村里，仍然是主要的取暖方式。
宋加有些疑惑。几千年的中国，冬天的取暖方式，为何至今没有改进到至少是壁炉的方
式。那种烧炭的方式，废气都是会被直接呼吸的，常常有人因为烧炭火而中毒。而那种

把PELLET打碎成小碎片，然后在37C超声水槽里超声，最后还是有微小的蛋白碎片怎么
都不溶解，测了OD280=2左右，也不算很浓啊，这个浓度的时候这个蛋白一般都是溶解

不过ethanol precipitation的recovery其实在室温下比在低温下更好。温度越低，
recovery越差。很多protocol建议低温，其实是出于degradation的考虑。有可能这个
trizol kit里加了很多酶抑制剂，所以不怕degradation，就可以室温操作了。
isopropanol的沉淀效率比ethanol高，所以需要的用量比ethanol少，适合用于浓度比
较低的核酸沉淀。但是isopropanol比ethanol难挥发，真空干燥用的时间更长，得到的
pellet也不如ethanol precipitation的pellet贴壁贴得牢。
至于你的chloroform wash不够effective的问题，也有可能是你吸取aqueous phase的
时候太贪心，想尽量把上层吸干净，结果反而夹带了一点phenol进来。我经常犯这种错
误，最后不得不再做一次chloroform wash，得不偿失。。。 |

发帖数: 1233 | 10 我不直接回答你的问题，只是提供一个貌似更好的、而且可行的方案。
与其用trizol，加什么氯仿、异丙醇，最后是乙醇这么多步骤，不如用RNAzol一步完成
。pipette的时候，不用担心多吸会有下层液体的污染。因为rnazol根本没有液体分层。
不管用那种试剂，你这种从活体组织提取rna的情况，根本用不着100mg。尤其象肝脏。
有两三粒芝麻大小的组织就完全可以了。当然，还是用1ml试剂比较稳妥。
另外，在加异丙醇（或者别的醇）沉淀rna的时候，不一定按照说明那样离心。可以采
取小一些转速。这样出来的pellet比较松。用酒精洗就会洗得干净。最后溶解的时候也
更容易。不用担心低转速会导致低收获量。你有足够的样品，不存在收获量低的问题。
只要动作快，不用担心常温、低温。比如用rnazol，从匀浆化到形成rna沉淀pellet，
不超过20分钟。从头至尾都有保护：试剂可以保护溶解状态rna不受降解，沉淀状态的
rna也不容易降解。
为了提高匀浆化效率，可采用sonication。10秒钟可以完全打散任何组织。 |

associated with the nucleic acid as well. You can do this with 0.5 M or
higher NaCl, or wash the pellet afterwards, to decrease the protein-DNA
interaction, but some proteins will remain in the pellet.

最后上样还是粘糊糊的，好不容易上成功了，然后做western，发现pellet里目的蛋白

对于那个第一作者，我觉得学术界的人恐怕对她会有看法，
但不可否认她的表演也许对公众和那些议员真的挺有用的。
至于你说的同位素标记，我可没你那么乐观，如果你去看看她的method，可能会有同感。
她纯化DNA的方法是细胞pellet裂解之后用酚氯仿多次抽提，然后水项用乙醇沉淀。
得到的DNA pellet都没说用70%酒精洗一遍就用buffer溶解了，然后溶液直接测放射性。
就算用70%酒精洗了一遍，最后得到的DNA里面也会有微量的盐污染。
我没有定量算过他们加到培养基里的放射强度和最后测到的放射强度间的关系，
有兴趣的朋友们可以自己算算。
最大的问题是她们没有阴性对照，不知道背景会有多大。

If you have protein in +IPTG sample, then if it is mostly in pellet, that
tells you the solubility in this buffer is probably not great. |

I am using Miltenyi anti-NKp46-biotin-bead- MS column kit. The first step is
to get splenocyte from spleen by gentlMACS without enzyme treatment. A very
simple protocol, just put spleen into the PEB buffer and let gentleMACS run
m_spleen_01 program once, and filter, spin, aspirate, resuspend the pellet.
The problem comes from the resuspending process. The small tight pellet will

新近表达一个蛋白，连上GST差不多116kD，昨天小试了一下，按照supernatant,
pellet的顺序，跑胶然后用gelcode
染，貌似大部分蛋白都是在pellet。
我不确定是不是真出现inclusion body了。
我没有在sonication前后留样品，所以我没法知道是不是因为sonication的缘故。

我昨天把1st binding后的supernatant又加beads了，看好没好些今天。
我觉得N-lau还是挺好使的。能solubilize大部分pellet。
我500ml的culture提了差不多160ug的蛋白。。。
我下次买点你说的B-Per试试好了～ （不过看成分也差不多）
我现在的protocol基本上是：(For 500ml culture)
1. thaw pellet on ice
2. add 18ml GST buffer (配方见上)，add 1% PI, add 2ml lysozyme (10mg/ml)，
mix on ice for 30 seconds
3. pass thru 20G syringe to reduce viscosity, also add DNAse 40ul (1mg/ml).
4. sonication (it will become clear)
5. 10000rpm, 40min，4deg
6. collect supernatant for GST beads binding

发帖数: 363 | 20 每个步骤都是别人都知道的,我只是组装了一下. 最大不同是别人是先Urea变性,提出来
再复性.我是利用Triton X-100可以洗脱SDS的特点利用SDS变性,然后Triton X100洗脱
复性,然后过柱. 肯定不会对所有蛋白适用,但是起码我遇到的都work (包括一个>100
Kd的酶).
我是用的sonication water bath, 也可以用probe sonication再加SDS,都OK,但是注意
dilution的比例和体积.

0.2mM MgCl2), or in RNase-free water for short-term storage.

vitrogen is from Advanced BioMatrix, Inc (5005-B)

纯化过程中用detergent啊，lyzosome啊
DNase还有RNase啊
结果破碎细胞以后离心得到的pellet，还有得到的复合物pellet都超级难resuspend的
一整块像Jelly的东西
能弄成大小碎块但是没有办法混匀
用针头反复吸吹也不行
加Uear到5M也不行
参考了一些提纯histone protein的protocol，
加HCl到0.25M也没有办法完全溶解

deoxycholate （DOC）就有很多沉淀出来，不知道会不会对蛋白得率有影响？

1. to one volume protein, add 1/100 volume of 2% DOC, keep on ice for 15 min
2. add 1/10 volume 100% TCA and keep on ice for 1 hr
3. centrifuge at 10,000 g for 10 min at 4C
4. wash the pellet in acetone to get rid of residual TCA
5. resolubilise the pellet in appropriate buffer, and add Laemmli sample
treatment buffer to protein solution

reagent也提取到了mRNA,nanodrop测出的浓度有800ng/ul，但是我试了cyclophilin，
gapdh，18s几个内参，都测不到CT值。已经重复几次了，郁闷的不行。请教各位，有什

用2M urea加1% triton溶1h就出来了。但是也遇到过一次RFP的需要用8M urea才能溶的
，那一次纯化出来效果非常差。所以刚才centuribob贴出这个方法后，我觉得非常有用
，很可能可以用在我们的inclusion body上面。 |
