到昨天才降到0ppm. 准备再养两天再换水， 让菌落稳定下来。现在水质的参数是：

pH: 7.8-8.0
ammonia: ~0 ppm
nitrite: ~0 ppm
nitrate: 40ppm
calcium: 400ppm
phosphate: 0ppm
KH:6-7
temperature: 79F
salinity: 1.024
add ammonia every day to rise its concentration to 1-2 ppm.

准备看看到时候换水能不能恢复。
由于用的是dry rock和蒸馏水, 水里硅和磷含量几乎没有， 估计爆褐藻的几率应该很
小。
现在主要的问题是如何减少nitrate含量。dry rock里厌氧菌的积累是很缓慢的， 估计

1.Uniformly 15N,13C labeled Ubiquitin T66C in 50 mM phosphate buffer, pH6.5,

RDC值.我目前只看其中三个residue的值: K33,K27,K29.

发帖数: 1704 | 28 不见得，PEI也算是homemade，从公司（一般推荐sigma吧）买粉末自己回来配，调一下
pH就可以用了，这点和Calcium Phosphate倒是很像，但是一来配制的pH的要求没那么
严格简单很多，二来毒性小一些，细胞谱也似乎更广一些，在不少细胞上效果据说可能
好一些，三来DNA/siRNA通吃，效果都不错。目前做高通量转染可以算首选，除非细胞
特殊。
Tranit的试剂其实是抄的Fugene的专利，成分其公司的技术支持直接告诉我和Fugene6
一样，所以用法用量都不用再作优化如果之前用的就是fugene6。价格相对Fugene6便宜
一些，但如果用量很大，还是不够经济。
Calcium Phosphate转染效率低基本就一个原因，complex形成的不够好。假如混匀之后
看不见雾状浑浊，那就可以直接扔掉不必浪费时间和细胞了。原因自然有很多，核心前
面大家都提了是pH的问题，有时候公司的KIT也信不过，我有同事就被promega坑过，换
了Invitorgen才正常点，再后来就再也不用了，因为价格也很贵，自己配又不是放心。

基因枪（金颗粒裹上核酸和蛋白）来暴力转么？因为其实考虑到将来的医疗前景，对于
转干细胞，才是最需要用瞬转的场合。从原理角度上来说，NaAgo适合瞬转，而且适合
针对特定部位进行改造，不是指简单的切除，而是指Knock-In，因为筛选的标记放在
Knock-In片段上即可。NaAgo因为不需要PAM序列，而且脱靶率低，用来做 Knock-In最
合适。
对于你说的小片段DNA被捕捉，我猜你的意思是说那个小片段不小心贴错在什么地方然
后被某个不长眼的聚合酶当成primer 来用了？这个取决于哺乳动物的系统如何处理5'-
phosphate, 因为哺乳动物内部是用的３'-phosphate。那么上游的聚合酶合成到那个被
当成primer的gDNA那里的时候，会出现5', 3'段都有磷酸基的情况。系统如何处理这个

cysDML and cPILOT, 相关工作在ASMS会议报道过。他们所用的方法基本还是基于传统
方法改良如Thiopropyl Sepharose 6B resin和Cys TMT，这些基于抗体和resin方法的
富集效率比较低，从鉴定数目就可以看到。我们的富集效率可以达到97%，很轻松鉴定
到10000+ unique peptides。
目前没有同时含有iodoacetamide+phosphate的tag。phosphate可能会被phosphatase酶
切掉，phosphonate不能，因此更加稳定。
至于为什么要富集Cys peptide,因为其low abundance,essential for the biological
activity and structure, highly sensitive and reactive.

大豆很安全”？由于向卫生部查询发现：管食品与药品安全的部门并不管“转基因大豆
安全”，由此我们必须自己开始寻找安全判断的依据。这就是我们申请向农业部查询发
给孟山都公司的转基因大豆安全证书的依据。

二、农业部查询转基因大豆安全证书
农业部在2011年第一次答复我们最初查询要求时说：批准美国孟山都公司转基因大
豆的四个根据之一，就是我方独立进行的第三方检测，言之凿凿。按理，这份报告应当
就是我国有关方面的第三方检测报告。
2012年2月20日下午，我们一行几个人依据信息公开法规，再次获准前往农业部查
询有关转基因大豆获批安全证书的证明文件。下午2点前，接待人员手持一册10厘米厚
活页插装卷宗等候我们。我们被允许查看并手抄记录，不允许拍照。最初规定只许一个
人看，不允许4人同时查看不同部分。经据理力争才允许我们4人分别同时查看。由于时

应当不是“肌酸激酶”。而是骨骼肌（属横纹肌）损伤后，大量的“肌球蛋白”(
