effects. Many companies tried to screen lead from house-hold or existing FDA

Generally, the kinases for T308 and S473 of Akt are PDK1 and mTOR complex 2
respectively. PI3K causes the production of PIP3 on the membrane, which then
binds to both Akt and PDK1 (which is assumed to be constitutively active).
So far there seems no direct evidence as to how mTORC2 is regulated. However
, inhibition of PI3K in most cases will lead to the suppression of
phosphorylation at S473.

我同意上位的观点,就是有可能在PI3K缺陷细胞或小分子抑制剂的情况下并没有完全
block其活性,我用了一个试验是在PI3K的p85缺陷细胞中检测到仍然有AKT473的调节,
但是否一定说明我这个调节是PI3K-independent的还为时过早。

没想到睡袋这么光明这么孝顺~~帮忙顶
我的个人看法是:首先要搞清楚高血压的原因。是家族遗传问题,还是伴随血糖产生的
,和胰岛素抗性是否有联系,心电图结果如何等。然后才能确定跑步是否有效。假如没
有什么特殊的原因,就是大多数人那样,活动少,饮食没有注意(比如米面特别多),
血脂也高一些,心脏机能弱,那么跑步应该会有一些效果。
有人专门做过这方面的研究,就是跑步是否可以改善高血压:

小弟最近看了看糖尿病的research。 发觉糖尿病人都是insulin resistance,然后
PI2k pathway是inactivation的。 那这不是跟癌症的pi3k pathway activation正相反?
而且如果治疗糖尿病的话,restore PI3k activation那不是很容易得癌症吗?

而cancer metabolism的热跟这两点都关系没那么大,就第一点来说,整个cancer
metabolism里能抱的唯一大腿就是IDH(在癌症里有大量mutation的core component).
其实在发现IDH mutation前,这也是这个领域被人criticize的地方,就是:既然如此
重要,怎么没什么突变。IDH的出现让领域里的人如获至宝,但既变如此,这样的例子
还是太少了。
要说明的是,cancer metabolism是一个比较新(但其实也有上十年了)的领域,它里
面的leaders不是以前做传统metabolism的人,而是做 cancer signaling的人,所以这
些人把自己做的targets像PI3K, mTOR, Myc, P53什么的都说成是cancer metabolism里
的。这些targets在癌症里毫无疑问是非常重要的,但玩的太久了就不容易继续做了,
所以大家在这里换个角度可以继续研究。但这些不仅调控metabolism,也调控很多其他

理砖科的论文,是和别处不同的。多是第一作者为主要的砖工,后边几个帮忙的
,再后边几个打酱油的,最后挂上老板的名字是为通讯作者。做工的人,PhD毕业之后
,往往先做一两轮postdoc,攒几篇paper,然后去找教职——这是二十年前的事,现在
要做到四五轮——有了大paper,后面找postdoc便有底气;倘若有了CNS,便可从AP做
起谋个PI当了,如果有几篇CNS,或者已经当上AP,A/P,那兴许就能评千人锦衣还乡了
。但多数的砖工,智商情商拙计,大抵没有这么顺利。只有牛人,才能当上老板push别
人干活。

当地时间9月20日,科睿唯安(原汤森路透知识产权与科技事业部)公布了最新的“引
文桂冠奖”,预测了22位可能在今年或未来,获得生理学或医学、物理、化学、经济学
诺贝尔奖的学者。
其中,华裔科学家、美国匹兹堡大学医学院病理学特聘教授张远(Yuan Chang)和其丈
夫、同为匹兹堡大学教授的帕特里克·摩尔(Patrick S. Moore)一同上榜生理学或医
学领域的“引文桂冠奖”。上榜原因是这对“夫妻档”科学家一起发现了卡波济氏肉瘤
相关疱疹病毒(KSHV),又名人类疱疹病毒(HHV8)。

理砖科的论文,是和别处不同的。多是第一作者为主要的砖工,后边几个帮忙的
,再后边几个打酱油的,最后挂上老板的名字是为通讯作者。做工的人,PhD毕业之后
,往往先做一两轮postdoc,攒几篇paper,然后去找教职——这是二十年前的事,现在
要做到四五轮——有了大paper,后面找postdoc便有底气;倘若有了CNS,便可从AP做
起谋个PI当了,如果有几篇CNS,或者已经当上AP,A/P,那兴许就能评千人锦衣还乡了
。但多数的砖工,智商情商拙计,大抵没有这么顺利。只有牛人,才能当上老板push别
人干活。

if the migration inhibitor is not specific, what will happen、
I saw a lot of inhibitors, such as mTOR or PI3K inhibtors, growth receptor
inhibitors, they are not cell type specific at all. I am confused how these
inhibitors can kill tumor cells at specific window.

其实是3个问题:
1. GSK的荧光底物筛选模型 是否是 假阳性
这个几乎是铁板钉钉的事实,4个实验室,5个不同模型全部证明
荧光底物结合SIRT1是假阳性
2. SIRT1能否作为糖尿病(衰老)的靶点
直接从酵母,果蝇跳跃到哺乳动物或人,现在证据不足
一篇SIRT1 gene KO, 几篇transgenic mice的结果
我个人觉得not convincing
3. resveratral及类似物是否直接target SIRT1
resveratral研究很多是hype,流行病学的研究结果
无法在临床试验中重复。
reveratrol的靶点太多(IKK,NFkB,AP1,COX,PI3K)
SIRT1只是最近才提上来的热点
考虑到荧光底物模型的不可靠性,我认为SIRT1是否
直接被resvatroal激活还要重新考虑
其他的类似物也要重新认识
Bottom line
GSK花了冤枉钱,现在还在死撑

就一个Lewis C. Cantley挤牙膏挤牙膏一样,做一点发一点,其他人就旁边看,或者跟
着挂点文章。除了PI3K和新发现的PKM2就没啥了哇,将来怎么也就是很有限的几条
pathway有待发现,这是怎么回事?全都是临床的?
