这些invitrogen卖的probe，我们都研究过，而且我也和invitrogen R&D联系过，
invitrogen他们承认他们的probe都不能在live cell里面定量，所以对我们的probe很
感兴趣。
我先讲一下为什么commercially available的probe不能很好的在live cell里面定量：
1.现在卖的所有的GSH或者thiol probe都是基于不可逆反应的，例如monochlorobimane
。一般按照invitrogen的protocol，加10-20 uM的probe和GSH反应，反应后，产物发出
荧光signal。这里的主要challenge是GSH的浓度非常高，一般是1-10 mM。如果是uM的
probe和mM的GSH反应，如果给足够的反应时间的话，不论你是1 mM GSH，还是10 mM
GSH，所有的probe都会被consume掉，所以得到同样的荧光output。
2. one can argue，如果GSH和probe反应足够慢的话，低浓度的GSH反应就会更慢，高

fluorescent probe，大家有什么建议，可以回答什么important的biological
questions？
Invitrogen卖一个叫thioltracker violet的probe，但是这个不能定量，只能定性的看
一下GSH的变化。尽管GSH probe的文章都发烂了，也没有一个能在live cell里面定量
的。
我们develop了一个probe （ThiolQuant Green）可以很好的定量细胞里面的GSH (
reduced form only)。我们把这个probe用于confocal microscope和flow cytometry，
能很好的和cell lysate GSH measurement correlate.
相比起来redox GFP or YFP，ThiolQuant Green是直接测量reduced form GSH，而不是

对于其中一个基因,他设计了4个probe
重复10次以后 这4个probe对于该基因表达的结果相矛盾
比如 probe a 表明 model 1 > model 2 > model 3
probe b 表明 model 2 > model 1 > model 3
probe c 表明 model 3 > model 2 > model 1
我记得这种情况下好像比较谨慎的做法是只评论probe,而不对基因表达作出判断
不知道有没有其它解释这个问题的统计方法 |

有没有看过这其中是否有snp？
2种假设
1，probe之间的差异认为是noise，无视，取median/mean代表整个gene
2,认为probe之间的区别来自splicing，在probe层次讨论差异表达
其实还是要看他数据的质量，比如同一个model下10次单个probe的可重复性究竟如何？
还有如果是设计的custom probe，是按照什么选的，能不能保证probe的质量，避免
cross hybrdization之类的。。。 |

个问题：
1）请问这个是不是用疏水性的probe就会好点，这个具体是什么公司的，什么probe呢？
2）我现在基本上是probe contaminate了就扔掉了，现在用的是Veeco的probe，大约22
刀一个。有没有其他的比较可靠的又便宜的supplier呢。

discussed in the paper. so we cannot distinguish between the cyto and mito
pools of GSH. this will certainly be our next step.
In terms of toxicity, we have not measured IC50 yet. during the 1-2 h
experiment, we did not observe any apparent toxicity. However, this does not
mean it does not have long term toxicity.
The highest probe concentration we used was 1 uM. For HeLa cells, we can use

。我曾经在5'UTR region做过两个probe，效果都很不好，全杂到rrna上了，结果后来

打开av_extracted view会发现自动cross-probe了，
cross-probe指的是把原来schematic和extract 以后的R, C, L, fet的schematic中的
对应节点关联起来。
但是如果是用的calibre的PEX extract生成calibre view的话，
打开calibre v_view会发现是一个新的R, C, L, fet schematic,
如何也实现cross-probe呢？

GTP and incorporated) in sourthern Blot. However, all failed. The probe is
single-stranded RNA, it should blot the DNA template. I don't know what's

很多（total RNA和polyA RNA都是），真正的条带相对很微弱。可是奇怪的用这个
probe做的FISH的结果很干净，信号看着一点不像backgroud. target出现在部分neuron
里，non-neuron里完全没有。
想请教试验大牛们，这FISH结果能相信吗？
如果FISH能crosshyb到rRNA上，看上去啥样？
怎样能改善northern的结果呢？polyA RNA仍然不能完全清除rRNA,所以还是有crosshyb
.
