should be minimal. assume zero if you just use it for reducing agent for
protein purification. it is a different story if you are using it for redox
potential determination since it requires accuracy.

But I guess you have to use [reducing agent]/[oxidative agent] to fine- tune
redox potential if finer steps are needed.

Just because we don't understand the process, does not mean it is cliche.
I haven't read the paper (I will), but I hope he mention redox signaling
such as glutathione modification, NO modification, etc. For example, NO
modified Hsp90 is selectively acute toxic to motor neurons.

I hate the name of "anti-oxidant." Yes, NAC is an antioxidant. However, α
-Tocopherol chemically speaking is an oxidant. It's metabolite, duraquinone
, is likely the active form (pubmed Guy Miller). Btw, co-enzyme Q10 is an
oxidant, too. I will call vitamin E compunds "redox modulator," and I
believe they hit a very specific target. Btw, the ROS is likely the end
result of the death cascade, not the cause.

你说的是agi-6628,早期的inhibitor?新的agi-221对r172k也适用,他们的cohort里有
一例R172k,也是有效果。idh1和idh2 WT基本就是不同功能...idh1在de novo lipid
synthesis和redox状态维持上更重要。只有neomphoric mutation功能是一致的。
Incyte这点是不错的,不过咱们算这些预期能占到市值的一半,MF,PV类的市场在
ruxolitinib的专利期内还是有2-4 billion的市值的。不过agios这个药celegene要全
部吃下来,最后估计agios会被celegene合并吧

请教一下大家,我们最近develop了一个能在live cell里面定量glutathione的
fluorescent probe,大家有什么建议,可以回答什么important的biological
questions?
Invitrogen卖一个叫thioltracker violet的probe,但是这个不能定量,只能定性的看
一下GSH的变化。尽管GSH probe的文章都发烂了,也没有一个能在live cell里面定量
的。
我们develop了一个probe (ThiolQuant Green)可以很好的定量细胞里面的GSH (
reduced form only)。我们把这个probe用于confocal microscope和flow cytometry,
能很好的和cell lysate GSH measurement correlate.
相比起来redox GFP or YFP,ThiolQuant Green是直接测量reduced form GSH,而不是

It is very useful at monitoring cellular redox states and using microscopy,
you might be able to see if you are depleting specific GSH population (i.e.
mito vs cyto). Get a good price from invitrogen, you could be rich.

有个电化学的问题一直没有想明白。
我有一个石墨电极,放在葡萄糖溶液里。溶液里还有一个参比电极。我加了电压(比如
,0.5V),没有得到氧化电流,因为葡萄糖在石墨上氧化的速度太慢。现在我在石墨上修
饰了酶(可以转化葡萄糖),加同样的电压,得到了电流。我现在做了一个C-V,假设
得到了氧化-还原峰。
请问,这个氧化还原峰是酶的呢,还是葡萄糖的?如果我没有得到明显的氧化还原峰,
又是为什么了呢?
现在情况再复杂一点儿。假设,此酶转化葡萄糖的速度比较慢。于是我在溶液里又加了
一种redox物质A. A在电极上的反应速度几乎为零,但是可以很快的氧化葡萄糖,自己
变成A-。A-又可以很快的被酶还原成A.这样反应速度就快了。我做了相同的实验。也得
到了氧还峰。这个峰又是谁的呢?和上面的峰有何不同?

算,差4.5eV吧; gas phase的计算值,和liquid phase的实验值比较,因为溶液相的
