Hi,
I tried a couple of transfections with the NgAgo plasmid from Addgene and
the 5'P oligo against GFP from the paper, but I did not see a dramatic
reduction in GFP+ as seen using Cas9 and a GFP sgRNA.
Anybody tried as well and came to the same (dead) end?

多谢版上几位好心人的专业教导,昨晚半夜之前我们就基本上决定了听从第二个大夫的意见做手术,但准备要求大夫先try closed reduction.后来国内一骨科大夫的意见反馈回来了,他的意见和版上几位专家的意见也差不多,也说桡骨有不明显的移位.只是说在中国这种情况基本上都是closed reduction.至于是否需要internal fixation他说他不确定,因为他没看到打了石膏的x光片.

有解释为啥Shell Rotella T、Mobil Delvac这些heavy duty diesel oil的Used oil
analysis结果都很突出的原因，普通engine oil是S certification，例如SJ、SM。商
用truck engine oil是C certification， 目前是CI、CJ。C的additive package比S的
牛x很多，但有可能会不兼容乘用车上的catalytic converter。
我的G35目前就在用Shell Rotella T，摩托车下次换油也准备用这个。mobil的
synthetic motorcycle oil 1 quart 12刀，受不了。

For those of you who say that the big size of the J-20 makes it less
manoeverable than the F-22, i say that 5th gen fighters are not supposed to
be dogfighters, so manoevrability is not an issue because BVRAAMs will
decide the fight in future wars as well as AESA radars. I have provided a
